Keyhole method for high-speed human cardiac cine MR imaging.

Although electrocardiographic (ECG)-gated magnetic resonance (MR) imaging is widely used for cardiac imaging, it has several disadvantages, such as long imaging time, respiratory artifacts, and motion artifacts induced by arrhythmia. An MR image can be acquired within about 0.3 seconds by using a fast gradient-echo imaging method. When this method is continuously applied, only two to three images can be obtained during a single cardiac cycle. The goal of this study is to obtain cine MR images in a single cardiac cycle using fast gradient-echo imaging combined with the "keyhole" method. The optimal conditions for the keyhole method for cardiac cine imaging were obtained by computer simulation based on a simplified cardiac model. When the read-out direction was set parallel to the cardiac short axis, left ventricular motion was almost correctly reproduced by the keyhole method with acquisition time reduced to one-fourth. J. Magn. Reson. Imaging 1999;10:778-783.

Interspecies transmission of feline immunodeficiency virus from the domestic cat to the Tsushima cat (Felis bengalensis euptilura) in the wild.

Feline immunodeficiency virus (FIV) was isolated from a wild-caught Tsushima cat (Felis bengalensis euptilura), an endangered Japanese nondomestic subspecies of leopard cat (F. bengalensis). Phylogenetic analysis of the env gene sequences indicated that the FIV from the Tsushima cat belonged to a cluster of subtype D FIVs from domestic cats. FIVs from both the Tsushima cat and the domestic cat showed similar levels of replication and cytopathicity in lymphoid cell lines derived from these two species. The results indicated the occurrence of interspecies transmission of FIV from the domestic cat to the Tsushima cat in the wild.

A null lesion in the rhodopin 3,4-desaturase of Rhodospirillum rubrum unmasks a cryptic branch of the carotenoid biosynthetic pathway.

The carotenoids accumulated by a mutant Rhodospirillum rubrum ST4, containing a single Tn5 lesion in the pathway for carotenoid biosynthesis, were analyzed by HPLC, 1H NMR spectroscopy, and field desorption mass spectrometry. The main carotenoid was identified as 3,4,3',4'-tetrahydrospirilloxanthin, and the four minor carotenoids were identified as rhodopin, 3,4-dihydroanhydrorhodovibrin, 3', 4'-dihydrorhodovibrin, and 1,1'-dihydroxylycopene. The C-3,4 and C-3',4' bonds of all 5 carotenoids are saturated, and they have 11 conjugated double bonds. With the exception of rhodopin, which is a normal intermediate of the wild-type pathway, all of the carotenoids are not naturally occurring. The Tn5 lesion was assigned to rhodopin 3,4-desaturase which is proposed to catalyze dehydrogenation at both ends of the symmetrical spirilloxanthin derivative. An unexpected finding was that the enzymes following rhodopin 3,4-desaturase are still able to end-modify the 3,4-, and 3',4'-saturated precursors and that the order of methylation and hydroxylation is not obligatory. It is proposed that the observed nonnatural carotenoids can be explained by the inclusion of a cryptic branch, unmasked by the absence of rhodopin 3,4-desaturase, in the established linear pathway for spirilloxanthin biosynthesis. This is the first example of latent branching of the carotenoid biosynthesis pathway exhibited by a carotenoid mutant of a phototrophic bacterium.

Characterization and localization of thromboxane A2 receptor in human and guinea-pig nasal mucosa using radiolabelled (+)-S-145.

1. TxA2 receptor (TP-receptor) antagonists such as S-1452 and Bay u 3405 have been shown to be effective in alleviating nasal blockage in patients with allergic rhinitis as well as guinea-pig allergic rhinitis models. The present study was conducted to examine the existence and localization of the TP-receptor in human and guinea-pig nasal mucosa by in vitro receptor binding autoradiography using radiolabelled (+)-S-145, which is a potent and specific TP-receptor antagonist with an extremely slow dissociation rate. 2. We ascertained the binding specificity of [3H]-(+ )-S-145 in human and guinea-pig platelet membranes by comparing the ability of four TP-receptor ligands of U-46619, (+)-S-145, I-(+)-S-145 and Bay u 3405 to displace the specific binding of [3H]-(+)-S-145 and [3H]-U-46619. The rank order of potency (Ki) for the displacement was correlated highly with that determined from [3H]-U-46619 binding to the same preparations. 3. Quantitative autoradiography using a radioluminographic imaging plate system, in which the radioactivity of [3H]-(+)-S-145 is expressed as photostimulated luminescence (PSL) per area (mm2), revealed that specific binding of [3H]-(+)-S-145 to human and guinea-pig nasal mucosa was saturable. Scatchard analysis showed about three fold higher affinity and two fold greater maximal binding to the nasal mucosa of humans than that of guinea-pigs: the KD and Bmax values in human mucosa were 2.82+/-0.35 nM and 6.47+/-0.33 PSL mm(-2) and those in guinea-pig mucosa were 8.23+/-1.93 nM and 3.37+/-0.66 PSL mm(-2), respectively. 4. Specific [3H]-(+)-S-145 binding to cryostat sections of human and guinea-pig nasal mucosa was displaced by another TP-receptor antagonist, Bay u 3405, and a TP-receptor agonist, U-46619. The order of potency (Ki value: nM) was (+)-S-145 (2.5) > Bay u 3405 (15.4) > > U-46619 (359.6) in human nasal mucosa and (+)-S-145 (22.8) > U-46619 (49.8) approximately Bay u 3405 (62.1) in guinea-pig nasal mucosa. These rank orders showed rather good correlation with those obtained for the respective platelet membranes. 5. Autoradiographs of human nasal mucosa demonstrated that specific [125I]-(+)-S-145 binding sites mainly exist on the smooth muscle layers of venous sinusoids and arterioles in the lamina propria, with few or no binding sites in the epithelium and nasal gland. 6. We concluded that radiolabelled (+)-S-145 can be used as a TP-receptor ligand for autoradiographic study, and that the TP-receptor is exclusively located on smooth muscle layers of venous sinusoids and arterioles in the nasal mucosa. The potent vasoconstrictive activity of TxA2 may cause reduction of local blood flow followed by mucosal oedema probably through mechanisms of vascular injury such as ischaemia-reperfusion.

Functional evaluation of telemedicine with super high definition images and B-ISDN.

In order to determine whether a super high definition (SHD) image running at a series of 2048 resolution x 2048 line x 60 frame/sec was capable of telemedicine, we established a filing system for medical images and two experiments for transmission of high quality images were performed. All images of various types, produced from one case of ischemic heart disease were digitized and registered into the filing system. Images consisted of plain chest x-ray, electrocardiogram, ultrasound cardiogram, cardiac scintigram, coronary angiogram, left ventriculogram and so on. All images were animated and totaled a number of 243. We prepared a graphic user interface (GUI) for image retrieval based on the medical events and modalities. Twenty one cardiac specialists evaluated quality of the SHD images to be somewhat poor compared to the original pictures but sufficient for making diagnoses, and effective as a tool for teaching and case study purposes. The system capability of simultaneously displaying several animated images was especially deemed effective in grasping comprehension of diagnosis. Efficient input methods and creating capacity of filing all produced images are future issue. Using B-ISDN network, the SHD file was prefetched to the servers at Kyoto University Hospital and BBCC (Bradband ISDN Business chance & Culture Creation) laboratory as an telemedicine experiment. Simultaneous video conference system, the control of image retrieval and pointing function made the teleconference successful in terms of high quality of medical images, quick response time and interactive data exchange.

Acute sodium deficiency reduces gustatory responsiveness to NaCl in the parabrachial nucleus of rats.

Sodium-deprived rats ingest excessive amounts of salt solutions at high concentrations which are normally avoided. This phenomenon, salt appetite, is known to be controlled by the sense of taste. In the present study, a possible involvement of the parabrachial nucleus (PBN) in sodium appetite was examined by recording PBN neuron responses to 12 different taste stimuli in sodium-deprived and control groups of urethane anesthetized rats. Sodium deficiency was induced by two injections of furosemide 24 and 22 h before the recording. Taste responses to 0.3 and 0.5 M solutions of NaCl were significantly lower in the sodium-deprived rats than in controls. In gustatory responses to other taste solutions except for quinine hydrochloride solution, no differences were detected between the two groups of the rats. Correlation coefficients of responses among sodium salts and those between 0.5 M NaCl and sweeteners were larger in the sodium-deprived rats than in controls. The results indicate that PBN neurons in sodium-deprived rats are critically implicated in induction of quantitative and qualitative changes of gustatory sense to sodium salts.

The Hansenula polymorpha PEX14 gene encodes a novel peroxisomal membrane protein essential for peroxisome biogenesis.

We have cloned the Hansenula polymorpha PEX14 gene by functional complementation of the chemically induced pex14-1 mutant, which lacked normal peroxisomes. The sequence of the PEX14 gene predicts a novel protein product (Pex14p) of 39 kDa which showed no similarity to any known protein and lacked either of the two known peroxisomal targeting signals. Biochemical and electron microscopical analysis indicated that Pex14p is a component of the peroxisomal membrane. The synthesis of Pex14p is induced by peroxisome-inducing growth conditions. In cells of both pex14-1 and a PEX14 disruption mutant, peroxisomal membrane remnants were evident; these contained the H.polymorpha peroxisomal membrane protein Pex3p together with a small amount of the major peroxisomal matrix proteins alcohol oxidase, catalase and dihydroxyacetone synthase, the bulk of which resided in the cytosol. Unexpectedly, overproduction of Pex14p in wild-type H. polymorpha cells resulted in a peroxisome-deficient phenotype typified by the presence of numerous small vesicles which lacked matrix proteins; these were localized in the cytosol. Apparently, the stoichiometry of Pex14p relative to one or more other components of the peroxisome biogenesis machinery appears to be critical for protein import.

The effect of L-menthol stimulation of the major palatine nerve on subjective and objective nasal patency.

To determine whether L-menthol stimulation of the major palatine nerve can affect nasal patency, we noted subjective and measured objective nasal patency before and after L-menthol stimulation of the palatal mucosa. L-Menthol stimulation of the palatal mucosa enhanced nasal sensation of airflow but nasal resistance was unaffected. By contrast, L-menthol stimulation after local anesthesia of the palatine nerve or of the nasal mucosa substantially diminished the subjective response of increased nasal patency but without affecting objective nasal patency. It is suggested that the effect of both direct stimulation by L-menthol of the major palatine nerve and its vapor action on the sensory nerve endings of the nasal mucosa produce an increase in the sensation of nasal patency without an increase in objective nasal patency.

Expression of prolactin gene in mouse placenta during late pregnancy: detection of mRNA and its translation product.

To examine the existence of PRL messenger ribonucleic acid (mRNA) in the mouse placenta during late pregnancy, reverse transcriptase-polymerase chain reaction (RT-PCR) and Southern blot analysis were carried out followed by nucleotide sequence analysis of cDNA. Total RNA extracted from each tissue was reverse-transcribed, followed by PCR with two oligonucleotide primers specific or a part of mouse PRL (mPRL) cDNA. An amplified RT-PCR product of predicted size was detected in all samples from the placenta of days 16 and 18 pregnant mice. This product was specifically hybridized with a probe overlapping an entire sequence of mPRL cDNA in Southern blot analysis. Nucleotide sequence analysis also provided evidence that the amplified cDNA had a nucleotide sequence completely identical to the mPRL cDNA sequence reported previously. Furthermore, mPRL with a slightly bigger molecular weight than that of pituitary PRL was detected in the placenta of days 12, 14, 16 and 18 pregnancy by immunoblot analysis. These results suggest that PRL mRNA and its translation product are synthesized in mouse placenta during late pregnancy.

Peroxisomal remnants in peroxisome-deficient mutants of the yeast Hansenula polymorpha [corrected].

We have analyzed the presence of peroxisomal remnants ('ghosts') in three peroxisome-deficient (per) mutants of the yeast Hansenula polymorpha, namely delta per4, delta per5 and delta per10. Under peroxisome-inducing growth conditions peroxisomal membrane proteins (PMPs) were normally synthesized in cells of these mutants. In addition, these cells contained clusters of small membraneous vesicles, which were absent in cells grown under peroxisome-repressing growth conditions. These structures displayed typical peroxisomal properties in that they proliferated upon overproduction of Per8p, the H. polymorpha peroxisome proliferation factor. Moreover, in delta per4 and delta per5 these vesicles were susceptible to glucose-induced proteolytic degradation.

Another interpretation of burst imaging as a variation of line projection imaging.

Burst imaging and DUFIS realize fast imaging without rapid switching of magnetic field gradients. In this paper, some new aspects of burst imaging are explored. Interestingly, when one-dimensional Fourier transform is applied, Burst imaging becomes similar to line projection scanning, a concept proposed earlier by Mansfield and Maudsley (P. Mansfield, A. A. Maudsley, Planar spin imaging by NMR. J. Magn. Reson. 27, 101-119 (1977)). The line scanning interpretation is applied to two-dimensional Burst imaging, and the relationship between its two-dimensional k-space trajectory and line scanning interpretation is discussed.

An international comparison of characteristics of the sensation of nasal obstruction between Canadian and Japanese patients.

To determine differences in perception of nasal obstruction in Canadians and Japanese, we assessed subjective and objective nasal patency of 48 patients in Canada and 43 patients in Japan. Mean severity of the sensation of nasal obstruction in Canadian patients was significantly higher than in Japanese patients, while no significant differences in nasal resistances were found. Complaints of nasal obstruction in Canadian patients were directly concerned with nasal breathing, while Japanese patients complained of indirect matters, such as "unable to concentrate on job or study" or "nasal obstruction or nasal speech pointed out by other persons." The differences might be due to national characteristics.

Heterologous complementation of peroxisome function in yeast: the Saccharomyces cerevisiae PAS3 gene restores peroxisome biogenesis in a Hansenula polymorpha per9 disruption mutant.

PER genes are essential for the biogenesis of peroxisomes in the yeast Hansenula polymorpha. Here we describe the functional complementation of a H. polymorpha per9 disruption strain (delta per9) by a heterologous gene. The Saccharomyces cerevisiae Pas3p, a homologue of per9p, restored peroxisome biogenesis and peroxisomal protein import in the delta per9 mutant, allowing it to grow again on methanol as sole carbon and energy source. This result shows that heterologous complementation of peroxisome function in yeast is indeed feasible and furthermore suggests that H. polymorpha delta per9 may be the candidate of choice to attempt the isolation of Per9p homologues from higher eukaryotes by functional complementation.

Mutagenic activation of 3-methoxy-4-aminoazobenzene by mouse renal cytochrome P450 CYP4B1: cloning and characterization of mouse CYP4B1.

A new P450 responsible for mutagenic activation of 3-methoxy-4-aminoazobenzene (3-MeO-AAB) which is a potent procarcinogen was purified from renal microsomes of male mice using an index of umu gene expression. The purified P450 had high bioactivation toward 3-MeO-AAB and also 2-aminofluorene and 2-aminoanthracene. The antibody against this P450 completely inhibited mutagenic activation of 3-MeO-AAB of mouse renal microsomes. With immunoblotting, this form was present abundantly in renal microsomes of male mice but not in those of female mice. This P450 was also present in pulmonary microsomes of male and female mice but not in hepatic microsomes. The NH2-terminal amino acid sequence analysis indicated that this form belonged to the CYP4B subfamily. Thus, mouse kidney cDNA library was screened with rat CYP4B1 probe. The cDNA-deduced amino acid sequence of isolated cDNA consisted of 511 amino acids and bore 90, 86, and 84% similarities to rat, rabbit, and human CYP4B1, respectively. The NH2-terminal amino acid sequence of the purified renal P450 and amino acid sequence of BrCN-digested peptides from the purified P450 agreed with the cDNA-deduced amino acid sequence. These results suggest that CYP4B1 is a major form in renal microsomes of male mice and plays a major role in mutagenic activation of 3-MeO-AAB. In extrahepatic tissue, CYP4B1 may contribute to chemical carcinogenesis.

A case of acquired petrous cholesteatoma associated with insidious middle ear infection treated by staging the surgical procedures.

Surgical treatment of petrous cholesteatoma is difficult, especially in infected cases, since it often involves vital structures. We report the case of a patient successfully treated for an infected petrous cholesteatoma by staging the surgical procedures to reduce the risk of intracranial complications. The patient, a 53-year-old man, was referred to our hospital because of vertigo during coughing or strenuous effort. The left side mastoid cavity was open to the external ear canal and wholly covered with cholesteatoma epithelium with purulent discharge. The superior basal turn of the cochlea, superior and posterior semicircular canals, and roof of the internal auditory canal were eroded. Conservative treatment was not effective in eradicating the otorrhea. Four weeks after the first operation (radical mastoidectomy), the second operation was conducted following a combined middle cranial fossa and transmastoid approach. The postoperative course was uneventful. Normal facial nerve function was preserved and unsteadiness disappeared, but hearing could not be preserved. The MRI examination, performed one year after surgery, did not reveal any evidence of residual cholesteatoma.

[Antihypertensive effects of repeated oral administration of cilnidipine, a novel calcium antagonist, in 2K1C renal hypertensive dogs].

Antihypertensive effects of repeated oral administration of cilnidipine in 2K1C renal hypertensive dogs were compared with those of nicardipine. On the first day, oral administration of cilnidipine (3 mg/kg) or nicardipine (3 mg/kg) markedly lowered both systolic and diastolic blood pressure 1 hr after administration. The hypotensive effects of cilnidipine were longer compared with those of nicardipine. Both drugs elevated the heart rate and plasma renin activity. On the 8th and 15th days, similar responses were obtained by repeated administrations of cilnidipine and nicardipine. After withdrawal of these drugs, no rebound phenomena in blood pressure were observed. The changes in mean blood pressure were correlated with plasma cilnidipine or nicardipine concentrations that were obtained at each time of blood pressure measurement (r = -0.598; P < 0.001 and r = -0.594; P < 0.001, respectively). These results suggest that stable and long-acting antihypertensive effects of cilnidipine for 15 consecutive days in renal hypertensive dogs are related to the change in plasma drug concentrations.
