Male Donryu rats and B6C3F1 mice were treated with dietary 3-methoxy-4-aminoazobenzene (3-MeO-AAB) (0.06% or 0.09%), and subcellular fractions of the liver were examined for ability to mutagenically activate 3-MeO-AAB and 2 amino acid pyrolysate components using Salmonella typhimurium TA 98 as a tester strain. The treatment resulted in striking induction of enzyme(s) for the mutagenic activation of these aromatic amines in rats, but the effect was smaller in mice. The enzyme(s) involved in the reaction was characterized as an isotype of microsomal cytochrome P-448.

The production of late placental alkaline phosphatase (ALP) isoenzyme and the co-presence of a small amount of tissue-unspecific ALP isoenzyme was confirmed in the newly established uterine cervical epidermoid cancer cell line SKG-III. Sodium butyrate (3mM), which has been shown to modulate oncodevelopmental gene expression, was examined after introduction of the agents into confluent cultures. 72 hours after the first sodium butyrate treatment, the amount (per mg cell protein) of total ALP activity increased to 5 times that of control cells and tissue-unspecific insoenzyme occupied about two thirds of the concentration of the modulated ALP. Treatment of SKG-III cells with prednisolone (5 micrograms/ml) for 48 hours caused a 3.5 times increase in total ALP activity, and most of the modulated ALP was occupied by late placental isoenzyme. From these data, it was concluded that sodium butyrate mainly induced tissue-unspecific isoenzyme, while prednisolone induced late placental isoenzyme. The results suggested that SKG-III cells had at least two ALP isoenzyme genes and chemical agents such as sodium butyrate or prednisolone could modulate the ALP isoenzyme profile of these cultured cells in vitro.
Luteinizing hormone-releasing hormone (LH-RH; 100 nM-50 microM) reduced the sensitivity of the nicotinic ACh-receptor in amphibian sympathetic ganglion cells and skeletal muscle end-plates. Analyses of LH-RH action, based on a Michaelis-Menten type kinetics, revealed that LH-RH depressed the maximum response (Vmax) of the dose-response curve of ACh currents without changing the affinity (Km) of ACh to the receptor. It was suggested that LH-RH reduced the sensitivity of nicotinic receptor by acting on a certain allosteric site of the receptor-ionic channel complex. Probably, LH-RH reduces the ACh current by decreasing the number of channels available.
Young (3-day-old) embryonic chick hearts have slowly rising action potentials (APs) dependent on slow Na+ channels, and these slow APs are blocked by verapamil, but not by Mn2+ (1 mM). In contrast, old embryonic hearts possess slow Ca2+-Na+ channels which can be blocked by both verapamil and Mn2+. In the present experiments, we compared the effects of a number of Ca antagonistic drugs on the spontaneous APs recorded from intact 3-day-old embryonic chick hearts (ventricles). At 10(-5) M, verapamil, D-600, and nifedipine abolished the spontaneous APs (3 X 10(-6) M produced some depression of Vmax). Electrical stimulation did not elicit APs, and long-duration hyperpolarizing pulses (applied because the maximum diastolic potential was moderately decreased) did not allow APs to be elicited by stimulation. This blockade was not reversed by elevation of [Ca]0 from 0.6 mM (control concentration used to reduce contractions) to 3.6 mM or 5.4 mM. However, lowering of [Na]0 from 137 mM to 68.5 mM facilitated the blockade. Washout of the drugs for 30 min produced nearly complete recovery. In contrast to the effects of verapamil, D-600, and nifedipine, 10(-5) M diltiazem, bepridil, and mesudipine had little or no depressant effect on Vmax; the same was true for 1 mM Mn2+. These results indicate that verapamil, D-600, and nifedipine are blockers of slow Na+ channels (as well as of slow Ca2+-Na+ channels), whereas diltiazem, bepridil and mesudipine do not block slow Na+ channels. Therefore, the Ca antagonistic drugs vary in their ability to block slow Na+ channels.
After about 12 days of gestation, fetal macrophages begin to appear in the subepidermal mesenchyme of rat fetuses. The macrophages are ultrastructurally characterized by cytoplasmic vacuoles, abundant polyribosomes and long filopodia. Immunocytologically, they possess Fc and complement (C3) receptors on the cell surface and are capable of immune phagocytosis, Latex or carbon phagocytosis, and glass adherence. From 15 days of gestation, lysosomal granules and micropinocytic vesicles gradually develop, together with an enlargement of Golgi complexes, whereas the number of polysomes and the number and size of cytoplasmic vacuoles are gradually reduced when gestation ends. Finally, the macrophages become amoeboid. Non-specific esterase and endogenous peroxidase activities are always absent in these macrophages. In culture experiments with cell suspensions prepared from the mesenchyme, fetal macrophages show a similar maturation process. Autoradiography with 3H-thymidine demonstrates a high proliferative capacity of the macrophages, particularly during the fetal stage.
A modified procedure of PAP-immunohistochemistry with the use of a rabbit antiserum against serotonin was applied to investigate the pattern of serotonin-containing nerve fibers in the spinal cord of the monkey, Macaca fuscata. The majority of descending serotonin fibers in the white matter is located immediately below the pia mater in the ventrolateral funiculi. Lamina I and the outer zone of lamina II are supplied with numerous serotonin fibers. In the intermediate gray, two prominent bundles composed of longitudinal fibers, i.e., lateral and medial longitudinal serotonin bundles, were recognized at the lateral column and in the vicinity of the central canal, respectively. The motoneurons of the anterior horn are encompassed by fine networks of serotonin fibers and terminals. The results obtained from studies with the monkey spinal cord closely resemble those characteristic of the dog spinal cord as presented in a previous paper, except for portions of the lumbar level. In segments L3-L4, intercalated cell groups between the medial and lateral motor nuclei receive particularly rich inputs of serotonin fibers in the same manner as the neurons of the nucleus intermediolateralis. This peculiar finding may suggest the presence of a specialized nucleus in the anterior column of the simian and also human spinal cord.
Detailed comparative analysis of the organization of serotonin fibers in the anterior column of the mammalian spinal cord (rat, guinea pig, cat, dog and monkey) was carried out by use of the indirect antibody peroxidase-antiperoxidase (PAP) method. The plexus formation of serotonin-containing varicose fibers around the alpha-motoneurons in the monkey anterior horn was in much closer apposition to the cell bodies in comparison with the spinal cords of the rodents and carnivores. The results may suggest that anterior horn motoneurons in the simian spinal cord are intimately innervated by serotonin fibers in a manner different from that of rodents and carnivores. Furthermore, the small cell groups endowed with particularly dense networks of serotonin fibers were demonstrable in the anterior horn of L1-L2 segments of rats, and L3-L4 of guinea pigs and monkeys; however, in the lumbar levels of the carnivores this was not the case. Hence it seems doubtless that there exists in the lumbar anterior horn of the rodent and primate spinal cords a cell group with an unknown specialized function.
The lumbar spinal cord of the rat was studied by combined retrograde fluorescent labelling with 4',6-diamidino-2-phenylindole-2HCl (DAPI) and immunoperoxidase procedure using serotonin antiserum. A peculiar small neuronal group endowed more densely than other anterior horn neurons with serotonin-like immunoreactive fibers was recognized in the anterior column of lumbar segments L1-L2. At the same time, this small nucleus was shown to contain the motoneurons innervating the cremaster muscle by means of retrograde labelling with DAPI. It is tentatively suggested that the bulbospinal descending serotonin system is particularly intimately connected with the function of the cremaster muscle.
A comparative study of serotonergic innervation on motoneurons in the brainstem of various mammals (mouse, rat, guinea pig, dog, cat and monkey) was carried out using a sensitive immunohistochemical method. Except for the extraocular muscle nuclei, the motor nuclei of the cranial nerves received rich inputs from serotonin neurons, in all species examined--rodent, carnivore and primate. The motoneurons of the monkey were innervated by varicose serotonin fibers, in a manner different from that of other species, i.e. their cell bodies and proximal dendrites were tightly encircled by a large number of serotonin-containing varicose fibers. At the ultrastructural level, a predominant population of axosomatic contacts was confirmed in the cranial motor nuclei of the monkey, particularly in the nucleus ambiguus.
19-Iodinated and 6 beta-iodomethyl-19-nor derivatives of cholesterol and 17-ketosteroid labeled with 131I were tested in rats to determine the critical structural features required for maximal adrenal uptake. The introduction of the 17-keto group in place of the 17 beta-side chain of cholesterol caused most of the radioactivity to be taken up by the thyroids. Fluorination at the C-3 position had deleterious effects on the adrenal concentration and led to the loss of adrenal specificity. A beta-hydroxy group at the C-3 position is substantially required for adrenal uptake.
The preparation and antigenic property of 3 beta-hydroxy-5-cholen-24-oic acid-bovine serum albumin (BSA) conjugate in which the hapten is linked to the carrier protein through an (O-carboxymethyl) oxime bridge at the C-19 position on the steroid nucleus is described. Antibody raised against antigen in the rabbit possessed high titer and specificity to 3 beta-hydroxy-5-cholen-24-oic acid, exhibiting no significant cross-reactions with various bile acids.
The preparation and antigenic property of 3-dehydrolithocholyglycine-bovine serum albumin (BSA) conjugate in which the hapten is linked to the carrier protein through an (O-carboxymethyl) oxime bridge at the C-3 position on the steroid nucleus is described. Antibody raised against antigen in the rabbit possessed high titer and specificity to lithocholylglycine, exhibiting no significant cross-reaction with free lithocholic acid or lithocholyltaurine.
When cassava (Manihot esculenta Crantz) root was cut into blocks and incubated under laboratory conditions, the blocks showed more widespread and more even symptoms of physiological deterioration than those under natural conditions. Thus, the tissue block system has potential for biochemical studies of natural deterioration of cassava root. The changes in cyanide content and linamarase (linamarin beta-d-glucoside glucohydrolase; EC 3.2.1.21) activity in various tissues during physiological deterioration were investigated. Total cyanide content increased in all parts of block tissue after 3-day incubation. The degree of increase in cyanide was most pronounced in white parenchymal tissue, 2 to 3 millimeters thick, next to the cortex (A-part tissue), where no physiological symptoms appeared. On the other hand, linamarase activity was decreased in all parts of block tissue after a 3-day incubation. A time course analysis of A-part tissue indicated a clear reciprocal relationship between changes in total cyanide and linamarase activity; total cyanide increased, while linamarase activity decreased. Free cyanide constituted a very small portion of the total cyanide and did not change markedly.
