Relationship between monensin and sulphur amino acid requirements in broiler chickens.

1. Diets containing 80, 100, 125, 150, 175 or 200 mg monensin/kg were fed to broiler chickens from 0 to 28 d in cages that prevented access to excreta. 2. Growth was depressed with 125 mg monensin or more/kg and food intake tended to decrease. Feathering was adversely affected with 175 mg or more/kg. 3. In a similar experiment, diets containing 8.8, 9.1, 9.4, 9.7 and 10.1 g total sulphur amino acids (SAA)/kg were supplemented with 125 or 80 mg monensin/kg and compared with a diet containing 8.8 g SAA and 33 mg robenidie/kg. 4. With 125 mg monensin/kg, body-weight gain was significantly less than that of birds receiving robenidine if dietary SAA content was 9.4 g or less/kg. With 9.7 g SAA or more/kg, gain in birds receiving monensin was similar to that of birds receiving robenidine. 5. Monensin at 125 mg/kg therefore appears to increase SAA requirement.

Establishment of a human cancer cell line from carcinoma of the gingiva.

A new human epithelial cell line (MK-1) has been established from a well differentiated squamous cell carcinoma of the gingiva. It has been subcultured 90 times during the past 23 months. The population doubling time of the MK-1 cell line was 28 h. A colony forming ability with 18% plating efficiency was shown. Chromosomal analysis revealed an aneuploid human male karyotype with 22 marker chromosomes. The model chromosome number was 75. Scanning electron micrographs of MK-1 cells showed numerous microvilli and intercellular bridges. Transmission electron micrographs of MK-1 cells showed epithelial characteristics with desmosomes, tonofilaments and membrane-coating granules. Hetero-transplantation of cultured MK-1 cells produced tumors, the histology of which was similar to that of original ones. The ultrastructure of the tumors showed the presence of abundant tonofibrils and keratohyalin granules. This MK-1 cell line is a model useful for the study of keratinization of squamous cell carcinomas.

The density and distribution of intramembrane particles in erythrocytes from persons with muscular dystrophies.

Intramembrane particles in the freeze-fractured faces of erythrocyte membranes from persons with several different types of muscular dystrophy are examined. The density of particles in the protoplasmic face was lower than that of controls, although not at a statistically significant level. No altered pattern of aggregation of these particles was observed.

Sulfoconjugation and glucuronidation of salicylamide in isolated rat hepatocytes.

Sulfoconjugation and glucuronidation of salicylamide by isolated hepatocytes were examined with various concentrations of salicylamide and sodium sulfate. The ratio of sulfate to glucuronide formed changed markedly, depending on the concentrations of salicylamide and inorganic sulfate in the medium. The apparent Km value of sulfoconjugation for salicylamide was 0.006 mM, while its Vmax value varied depending on the concentration of inorganic sulfate (e.g., 2.1 and 0.5 nmoles/min/10(6) cells at 1.2 and 0.5 mM inorganic sulfate, respectively). The Km and Vmax values of glucuronidation for salicylamide were 0.19 mM and 1.28 nmoles/min/10(6) cells, respectively, in the absence of sodium sulfate. Glucuronidation was suppressed in the presence of inorganic sulfate. The suppression could be attributable to the competitive consumption of salicylamide by sulfotransferase. Additional in vivo experiments revealed that an extra amount of salicylamide markedly lowered the blood inorganic sulfate levels of rats. The significance of the finding is discussed in conjunction with the variation of the Vmax value of sulfoconjugation with the inorganic sulfate concentration.

Some quantitative evaluation of first-pass metabolism of salicylamide in rabbit and rat.

The first-pass metabolism of salicylamide (SAM) was studied in rabbits and rats paying main attention to the amounts of the glucuronide (SAMG) and sulfate (SAMS) formed. The roles of the intestine and liver for SAMG and SAMS formation during the first-pass were also examined quantitatively. Further, an equation was developed to estimate the hepatic first-pass metabolism based on the experiments of intravenous and oral administration and intestinal perfusion experiment. Significant species differences were found in these two animals. The extent of the first-pass metabolism was larger in rabbits (ca95%) than in rats (ca60%). Of the 95% first-pass metabolism in rabbits, about 30% is due to the intestine whereas the remaining 65% is due to the liver. Of the 60% in rats, about 20 and 40% are estimated to be due to the intestinal and hepatic first-pass metabolism, respectively. In rabbits SAMG and SAMS are formed together in nearly equal amounts in the liver and at the ratio 4 : 1 in the intestine. In rats, on the other hand, SAMG is exclusively formed in the intestine and SAMS in the liver. In this context, the equation described here was applied to the data of Gugler et al. (J. Pharmacol. Exp. Ther., 195, 416-423 (1975)) who carried out a study on the first-pass metabolism of SAM in dogs, and consequently, reported that glucuronidation occurs primarily in the liver whereas sulfation occurs primarily in the intestine in dogs.

[Non-invasive quantitative evaluation of aortic regurgitation by ultrasonic pulsed Doppler flowmeter (author's transl)].

A usefulness of non-invasive and quantitative evaluation of aortic regurgitation (AR) was investigated by measuring the aortic arch flow from the suprasternal notch with ultrasonic pulsed Doppler flowmeter (UPDF). Measurement of the blood flow was made on 19 patients with AR, calculating the regurgitant ratio from the waves of relative flow volume, and the estimated grade of AR was compared with the severity judged from the cineaortogram. The results were as follows: 1) The aortic arch flow was recorded in 17 out of 19 patients with AR. 2) Except mild cases, the diastolic regurgitant flow was recorded in patients with AR. 3) The regurgitant ratio obtained by the UPDF correlated well with the grade of the severity of AR judged from the cineaortogram.
