Inhibition of microtubule polymerization by synthetic estrogens: formation of a ribbon structure.

Dienestrol, meso-hexestrol, and dl-hexestrol, synthetic nonsteroidal estrogens, were shown to be inhibitors of microtubule assembly in vitro using microtubule proteins isolated from porcine brains. The order of activity of the synthetic estrogens as inhibitors of microtubule assembly is: dienestrol greater than diethylstilbestrol greater than meso-hexestrol greater than dl-hexestrol greater than isodienestrol. The activity of dienestrol as an inhibitor was of the same order as that of (+)-griseofulvin, as determined by turbidity measurement. Electron microscopic observation revealed that twisted ribbon structures are formed from microtubule proteins in the presence of some synthetic estrogens (dienestrol, meso-hexestrol, and dl-hexestrol).

Immunohistochemical study of the cells infiltrating human renal allografts by the ABC and the IGSS method using monoclonal antibodies.

Sixty renal allograft tissues obtained from 29 patients were stained with hematoxylin and eosin. These tissues were histologically classified into 4 patterns according to the distribution pattern of the infiltrating cells: normal, focal, focal-diffuse, and diffuse types. Clinical signs of acute rejection were observed in 88% of the patients with the diffuse type infiltration, and 83% of those with the focal-diffuse type infiltration but in only 13% of those with the focal type infiltration. Twenty-four renal allografts were analyzed by the ABC and the IGSS methods using monoclonal antibodies. The number of T cells (Leu 1) accounted for about 80% of the total number of infiltrating cells; 2-8% of the cells were B cells (Leu 12); about 10% were NK/K cells (Leu 7); and 4-6% were monocytes/macrophages (Leu M3). As to helper/inducer T cell (Leu3a) and killer/suppressor T cell (Leu2a), which are T lymphocyte subsets, there were more Leu3a- than Leu2a-positive cells in focal type tissue, but there were more Leu2a- than Leu3a-positive cells in focal-diffuse and diffuse type tissue. In most cases that developed clinical signs of acute rejection, there were more Leu2a- than Leu3a-positive cells. The Leu3a/Leu2a ratio in most of the AZA-administered cases dropped immediately after the transplantation and maintained a low value, but in the CSA-administered cases it decreased gradually post-transplant.

Antimicrobial activity of tertiary amine covalently bonded to a polystyrene fiber.

Tertiary amine was covalently bonded to a polystyrene fiber and examined for antibacterial activity. The tertiary amine covalently bonded to a polystyrene fiber (TAF) showed a high antimicrobial activity against Escherichia coli. TAF exhibited a stronger antibacterial activity against gram-negative bacteria (E. coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, Salmonella typhimurium, and Serratia marcescens) than against gram-positive bacteria (Staphylococcus aureus and Streptococcus faecalis) or Candida albicans. This activity against E. coli was accentuated by 0.1% deoxycholate or 10 mg of actinomycin D per ml, to which E. coli is normally not susceptible. This implies that TAF causes an increase of the bacterial outer membrane permeability. On the other hand, the antimicrobial activity was inhibited by adding Mg2+ or by lowering the pH. This suggest an electrostatic interaction between the bacterial cell wall and TAF. Scanning electron microscopy showed that E. coli cells were initially attached to TAF, with many projections on the cell surface, but then were apparently lysed after contact for 4 h. Taken together, these results imply that bacteria initially interact with TAF by an electrostatic force between the anionic bacterial outer membrane and the cationic tertiary amine residues of TAF and that longer contact with TAF damages the bacterial outer membrane structure and increases its permeability.

[Physiological and self-rated affective/cognitive responses of the neurotic personality to an interpersonal stress situation].

The effects of approach by an unacquainted male to high- or low-neurotic (on the MPI scale) female subjects on physiological responses (heart rates and eye blinks) and self-rated affective/cognitive responses (tension, anxiety and apparent size of the male) were examined. (a) In the first trial, non-neurotic subjects showed an abrupt increase of HR near the personal space boundary, however showed a rapid habituation at the second and third trials. In contrast, self-rated affection (tension and anxiety) increased gradually as the male approached, and habituation was slow at the later trials. (b) Neurotic subjects displayed higher tension than non-neurotic subjects, while tension, anxiety and heart rate were less habituating. (c) INDSCAL analysis revealed that these three self-rated indices clustered together, while the physiological ones did not. (d) It was suggested that the eye blink response had a tension reducing function.

[Clinical use of autologous frozen blood in the surgery of three esophageal cancer patients].

Autologous frozen blood transfusion (AFBT) has advantages both of autologous and frozen blood transfusion. In AFBT there are no remarkable adverse effects which often emerge after usual heterologous blood transfusion. As a rule, four hundred milliliters of blood were drawn twice from patients and reserved as autologous frozen blood (AFB) preoperatively. It is supposed that radical operation of esophageal cancer using only AFB is difficult to perform because of various kinds of preoperative risks. In this paper three cases of radical esophageal cancer operations, in which only AFBT were used are reported. Pre- and postoperative liver functions were uneventful. RBC counts, Hb and Hct dropped after drawing blood and did not recover until the day of operation. Postoperatively, they deteriorated further but recovered to initial values without any specific treatment within 5 months after operation. Pre- and postoperative PaO2 values of AFBT were not different from those of the usual blood transfusion. Thus using only AFBT, esophageal cancer operations were performed without any disadvantageous effects.

Renal effects of dichloroacetate in vivo.

We have investigated the effect of dichloroacetate (DCA) on the urinary excretion of lactate and pyruvate in a patient with congenital lactic acidosis and in healthy human controls. DCA administered orally in doses of 50 mg/kg and 30 mg/kg decreased plasma lactate and pyruvate of both the patient and the controls, while the urinary excretion of lactate and pyruvate was increased. However, these urinary increments were too small to contribute to the decrease of plasma lactate and pyruvate.

Molecular cloning of unintegrated closed circular DNA of porcine retrovirus.

Viral DNA of unintegrated closed circular form was isolated from a swine kidney cell line (SKL) which was infected with a porcine retrovirus Tsukuba-1 (PRetV) produced from a swine malignant lymphoma-derived cell line. Shimozuma-1 and cloned using a lambda phage vector, Charon 21A. One of ten independent clones contained the 8.3 kb DNA fragment as an insert, which was thought to be a full length of viral DNA molecule carrying a long terminal repeat (LTR) sequence. We have analyzed this insert by mapping the recognition sites of some restriction endonucleases by Southern blot hybridization with appropriate probes.

Supraspinal depressant action of diazepam on the crossed extensor reflex.

The effects of diazepam on the crossed extensor reflex (CER) were compared between intact and spinally transected rats and also between IVth and lateral ventricular administration. The CER of the chronic spinal rat was not influenced by 0.05-2 mg/kg doses of diazepam, which was 5-200 times larger than the ED50 of the intact preparation. The IVth ventricular injection required only half as much diazepam compared to the lateral ventricular injection and the elapsed time between drug administration to the peak effect was significantly shorter. According to these findings, benzodiazepines seem to act primarily on the supraspinal structure near the IVth ventricle and thus depress the CER.

Simultaneous detection of B-cells and T-cells by a double immunohistochemical technique using immunogold-silver staining and the avidin-biotin-peroxidase complex method.

A new double immunohistochemical technique for the simultaneous detection of B-cells and T-cells was investigated, using tissue preparations obtained from human axillary lymph nodes and rejected renal allografts. The specimens were immunostained first for the demonstration of B-cells, by the immunogold-silver staining (IGSS) method using Leu-12 monoclonal antibody, and then for T-cells by the avidin-biotin-peroxidase complex (ABC) method using Leu-1 monoclonal antibody. With the present methods, both B-cells and T-cells were clearly detected and distinctively identified without cross-linking of antibodies or double reaction of enzymes.

Tetrodotoxin secreting glands in the skin of puffer fishes.

Unique exocrine glands or gland-like structures were found in the skin of several species of puffer fishes of the genus Takifugu. The glands of T. pardalis and T. vermiculare porphyreum consisted only of secretory cells with a large vacuole. These cells were completely enclosed by epithelial cells with developed microfilaments, except at their opening to the lumen. The contents of the large vacuole in the peculiar secretory cell were forced out when the puffer was stimulated. Exocrine glands or gland-like structures with peculiar secretory cells were also found in the skin of T. poecilonotus, T. niphobles and T. vermiculare radiatum. A high concentration of TTX was detected in the gland contents collected directly from live specimens of T. pardalis. We therefore conclude that these glands are TTX secreting glands.
