Do all daughter cells enter the "indeterminate" ("A") state of the cell cycle? Analysis of stathmokinetic experiments on L1210 cells.

The results of ten different stathmokinetic experiments on L1210 cells are analyzed to determine whether all the postmitotic cells enter the exponential AG1 compartment of the cell cycle characterized by exponentially distributed transit times. The analysis is based on a mathematical model coherent with the generalized A-B-transition hypothesis. An original fitting procedure is introduced to estimate the fraction of cells entering AG1 as well as other parameters of the cell cycle. Results of the analysis suggest that either all or nearly all postmitotic cells enter G1A. The results are discussed with respect to the validity of the A-B-transition hypothesis of the "probabilistic" model of the cell cycle. Some systematically occurring discrepancies that do not conform with generally accepted cell cycle models are apparent from the analysis of this data.

Linkage analyses in type I diabetes mellitus using CASPAR, a software and statistical program for conditional analysis of polygenic diseases.

We have developed software and statistical tools for linkage analysis of polygenic diseases. We use type I diabetes mellitus (insulin-dependent diabetes mellitus, IDDM) as our model system. Two susceptibility loci (IDDM1 on 6p21 and IDDM2 on 11p15) are well established, and recent genome searches suggest the existence of other susceptibility loci. We have implemented CASPAR, a software tool that makes it possible to test for linkage quickly and efficiently using multiple polymorphic DNA markers simultaneously in nuclear families consisting of two unaffected parents and a pair of affected siblings (ASP). We use a simulation-based method to determine whether lod scores from a collection of ASP tests are significant. We test our new software and statistical tools to assess linkage of IDDM5 and IDDM7 conditioned on analyses with 1 or 2 other unlinked type I diabetes susceptibility loci. The results from the CASPAR analysis suggest that conditioning of IDDM5 on IDDM1 and IDDM4, and of IDDM7 on IDDM1 and IDDM2 provides significant benefits for the genetic analysis of polygenic loci.

Mathematical model of the proliferation cycle of lymphoblastic leukemia cells.

In the paper a system of equations is described, which enables computation of the mean number of leukemic cells in the blood as a function of time. After formulation of the problem in the form of cybernetical model, the equations are derived. Then the results of modeling obtained with the aid of a digital computer are described. The paper concludes with short summary.

Analysis of different variants of erythrocyte survival using digital computers.

In the paper a method is presented, enabling parameter estimation of erythrocyte survival patterns, what is an important element in the diagnosis of haemolytic anaemias. The method is based on a computer program, fitting the experimental data to eight variants of theoretical patterns with use of a weighted least squares criterion. A simple statistical test of approximation quality is described and several interpretational examples are given. The program may be easily handled by a physician with small programming experience.
