Effect of 17 beta-estradiol on the proliferation of osteoblastic MC3T3-E1 cells via human monocytes.

Effects of human monocyte-conditioned medium on the proliferation of osteoblastic MC3T3-E1 cells were investigated in serum-free cultured condition. Monocyte-conditioned medium significantly stimulated osteoblast proliferation at the concentration between 10 and 30%, compared to that in the absence of monocytes. 17 beta-estradiol directly stimulated osteoblast proliferation at the concentrations of 10(-8) and 10(-10)M. On the contrary, the conditioned medium prepared by monocytes cultured in the presence of 17 beta-estradiol at the concentrations of 10(-8) and 10(-10)M significantly inhibited osteoblast proliferation. Present data indicate that in addition to direct effect on osteoblasts, 17 beta-estradiol affected osteoblast proliferation presumably through modulating the release of several local regulators of bone turnover from monocytes. The effect on osteoblastic activity via monocytes might be linked to the coupling of osteoclast and osteoblast actions.

Circulating monocyte (macrophage)-specific antibodies in patients with autoimmune thyroid diseases.

We investigated the presence of circulating monocyte-specific antibodies (monocytotoxic activities) by a complement-dependent cytotoxicity test and the relations between these monocytotoxic activities and other immunological indices in patients with autoimmune thyroid diseases. Antibodies reactive for monocytes (macrophages) were found in the sera from patients with autoimmune thyroid diseases. These antibodies were present in both IgG and IgM fractions and specific for monocytes since they were absorbed by monocytes but not by lymphocytes or granulocytes; furthermore, lymphocytotoxic and granulocytotoxic activities were not changed after the absorption of the sample sera by monocytes. Also, these antibodies did not have cross-reactivity to thyroid-specific antigens demonstrated by absorption tests and their specificity was different from anti HLA-DR antibody demonstrated by a flow cytofluorometric analysis. Monocyte-specific antibodies are reactive for autologous monocytes as well as allogenic monocytes. Patients who had positive monocytotoxic activities had high levels of TSH receptor antibodies (TRAb) and antimicrosomal antibodies in Graves' disease, and monocytotoxic activities were significantly correlated with the levels of TRAb in Graves' disease. These results suggest that the monocyte-specific antibodies (monocytotoxic activities) were significantly correlated with the immunological activities in Graves' disease.

[Application of nitrides to dental casting. 2. Feasibility as coating materials for Co-Cr alloy].

Nitrides used in this experiment are hBN, A1N and Si3N4. We studied feasibility of nitrides as wax pattern coating materials for Co-Cr alloy in an atmosphere of argon. Wax patterns were painted with coating material of nitrides (first investing), then a mixture of phosphate bonded investment and water was used in outer investing (second investing). The temperature of mold was 800 degrees C, and an argon arc vacuum-pressure casting machine was used. We cast the thin specimens in Co-Cr alloy (composition Co: 63 wt%, Cr: 27 wt%, Mo: 5 wt%, Bal.: 5 wt%; melting point: 1,380 degrees C). Surfaces of cast specimens were investigated using the unaided eye, a scanning electron microscope and a surface roughness analyzer. Furthermore, the thick specimens (globular form: phi 6.5 mm) using coating material of hBN were investigated using the unaided eye. Results were summarized as follows; 1. The nitrides controlled remarkably the reaction between cast specimen and investment. 2. The nitrides had the good effect of separation. 3. The cast surface coated the nitrides had less clinging than uncoated one, and this indicated that its cleansability was improved. 4. The nitrides decreased the surface roughness (Rz) of the cast to about 1/3. 5. hBN had the best effect in nitrides. 6. hBN also had the effect in a thick globular specimen.

Glycosaminoglycans in 3'-methyl-4-dimethylaminoazobenzene-induced rat hepatic cancer.

The changes in glycosaminoglycans in livers of rats with 3'-methyl-4-dimethylaminoazobenzene-induced hepatic cancer were examined and compared with those in fetal liver. The incorporation of 35S into sulfated glycosaminoglycans in hepatic cancer tissue was also studied after i.p. injection of Na(2)35SO4. The major component of glycosaminoglycans in healthy adult rat liver was heparan sulfate (61.7%), with hyaluronic acid (21.1%), dermatan sulfate (13.1%), and chondroitin sulfate (4.0%) as minor components. The quantities of all of the examined glycosaminoglycans were higher in tumors than in normal liver, but chondroitin sulfate and hyaluronic acid were more prevalent in the tumors (about 51 and 7 times higher, respectively). As a result, the share of heparan sulfate was decreased (37.1%) in the tumors. The high content of chondroitin sulfate (about 30 times) and the decreased share of heparan sulfate (29.0%) were also observed in fetal liver. The incorporation of 35S into individual glycosaminoglycans varied markedly. Approximately 90% of the label in glycosaminoglycans of healthy liver was found in a heparan sulfate fraction 4 hr after injection. In hepatic cancer tissue, however, 35S incorporation into both chondroitin sulfate and dermatan sulfate fractions was increased about 6.5- and 5.6-fold, respectively, of those in healthy liver.

Serum and liver glycylproline dipeptidyl aminopeptidase activity in rats with experimental hepatic cancer.

Changes of glycylproline dipeptidyl aminopeptidase (GPDA) and gamma-glutamyl transpeptidase (gamma-GTP) activities were compared in the serum and liver tissue of rats with hepatic cancer induced by 3'-methyl DAB. Serum glycylproline dipeptidyl aminopeptidase activity in rats with the azo dye-induced hepatic cancer was significantly higher than that in healthy rats, but the increase was not so extensive compared with that of gamma-glutamyl transpeptidase. The specific activity of glycylproline dipeptidyl aminopeptidase was decrease in the microsomal fraction and increased in the supernatant fraction of hepatic cancer tissue, whereas that of gamma-glutamyl transpeptidase was increased in both microsomal and supernatant fractions. These results suggest that the mechanisms, whereby serum activities of these two enzymes were increased in rats with hepatic cancer, were different from each other.

Electrophoretic fractionation of serum gamma-glutamyl transpeptidase in human hepatic cancer.

Serum gamma-glutamyl transpeptidase from patients with various hepatobiliary diseases was fractionated by polyacrylamide gradient gel slab electrophoresis to study the specific patterns of gamma-glutamyl transpeptidase fractions in hepatic cancer. On zymograms of normal serum gamma-glutamyl transpeptidase, a total of 10 fractions was observed. Additionally, fractions I', I" and II' were recognized in sera from hepatocellular carcinoma patients. Among these, fraction I', which migrated slightly, but significantly, slower than fraction I was the most specific; it was found in 55% of the hepatocellular carcinoma patients. Fractions I" and II' were also relatively specific, each was observed in about 29% of these patients. Fractions V to IX were observed in few hepatocellular carcinoma cases. Fraction I' is thought to be a hepatoma-related fraction, highly specific for the serum of hepatocellular carcinoma patients. Fractions I" and II' were also thought to be hepatoma-related fractions of gamma-glutamyl transpeptidase. We suggest that fractions I', I" and II' may be useful in the diagnosis of hepatocellular carcinoma.

Serum glycylproline dipeptidyl aminopeptidase activity in human hepatic cancer.

A method for the assay of glycylproline dipeptidyl aminopeptidase activity in the serum is described. The enzyme activity determination in sera from patients with hepatic cancer opens a possibility in the differential diagnosis of hepatobiliary diseases, because of the significantly higher values found in some cases of hepatic cancer. No correlation with alpha-fetoprotein was found.

Carcinoembryonic proteins in gastric carcinoma metastatic to the liver.

Two cases of liver metastases from gastric carcinoma are described in which the simultaneous occurrences of alpha-fetoprotein (AFP), carcinoembryonic antigen (CEA) and carcinoplacental alkaline phosphatase (CPALP) were demonstrated in the sera and tumor tissues. AFP was detected not only in the tumor tissues but also in the liver tissue adjacent to the tumor, while the other 2 carcinoembryonic proteins were not detected in the non cancerous liver tissues. The characteristics of CPALP in Case 1 were almost similar to the Nagao isoenzyme, based on enzyme tests involving L-leucine, L-phenylalanine and EDTA inhibitions, heat-stability and Michaelis constant, except for electrophoretical slower moving, while that in Case 2 were identical to variant type CPALP (Warnock).

The glycosaminoglycans in human hepatic cancer.

A method is proposed for the analysis of glycosaminoglycans that were isolated from human liver, combining cellulose acetate electrophoresis and enzymatic digestion with mucopolysaccharidases. The major constituent of glycossaminoglycans in the healthy liver is heparin sulfate and/or heparin (about 65%), with approximately equal quantities of dermatan sulfate and hyalauronic acid (about 13.5 and 13%, respectively) and a small amount of chondroitin sulfate. These components, especially chondroitin sulfate and hyaluronic acid, are markedly increased in hepatic carcinomas.

Hypopituitarism associated with Cogan's syndrome; high-dose glucocorticoid therapy reverses pituitary swelling.

A 70-year-old woman with Cogan's syndrome first presented with central diabetes insipidus and then developed secondary hypothyroidism. Magnetic resonance imaging revealed a diffuse pituitary swelling without evidence of tumor. High-dose glucocorticoid therapy administered to treat Cogan's syndrome was very effective in suppressing the inflammatory process, and resulted in the reversal of the pituitary swelling and partial recovery of thyroid stimulating hormone secretion. This is the first case of hypopituitarism associated with Cogan's syndrome, a form of autoimmune vasculitis. The glucocorticoid-responsive pituitary lesion is best explained by autoimmune hypophysitis which shows pituitary swelling and is known to often associate with other autoimmune phenomena.
