Eustachian tube function in the older adult.

This investigation examined the effects of the aging process on the adequacy of Eustachian tube function. Older adults presenting normal hearing sensitivity and sensorineural hearing loss and normally hearing young adults served as subjects. Eustachian tube function was assessed using electroacoustic immittance measurement. Tympanograms were analyzed relative to pressure and function (amplitude) changes after Eustachian tube testing. One-way analyses of variance and chi 2 analyses revealed no significant differences between groups and no significant relationships between group membership and tympanometric measures.

The antigenic composition and protein profiles of eumycetoma agents.

The protein profiles of different eumycetoma agents were compared by SDS gel electrophoresis. Dendrograms confirmed the homogeneity of isolates of Pseudallescheria boydii but amongst Madurella species, particularly isolates identified as M. grisea, there were substantial differences in protein composition. However using Western blotting reference isolates of the different species showed distinct antigen patterns in response to immune rabbit sera. In particular there was little evidence of cross reactivity between M. mycetomatis and M. grisea. However this specificity was not apparent when human sera from patients with different eumycetoma infections were compared in an ELISA system using the same antigens. It is possible that the formation of a mycetoma grain may limit a patient's exposure to antigens which confer specificity, an explanation which may also account for the variability in antibody responses seen.

The effect of sulconazole on the antigenic composition of two dermatophyte species.

Trichophyton rubrum and Trichophyton interdigitale have been grown in liquid culture in the presence of sulconazole. The antigenic activity of detergent extracts of intact organisms was analysed following SDS-PAGE and the probing of Western blots with homologous antisera raised in rabbits and with sera from patients with dermatophyte infections. Differences in protein-band patterns were noted; some bands present in control samples were absent in azole-treated samples and vice versa. These differences were reflected in antigenic band patterns, especially among components of approximate molecular weight of 30-40, 50-60 and 92-100 kDa.

A new selective enrichment procedure for isolating Pasteurella multocida from avian and environmental samples.

A selective enrichment procedure, using two new selective media, was developed to isolate Pasteurella multocida from wild birds and environmental samples. These media were developed by testing 15 selective agents with six isolates of P. multocida from wild avian origin and seven other bacteria representing genera frequently found in environmental and avian samples. The resulting media--Pasteurella multocida selective enrichment broth and Pasteurella multocida selective agar--consisted of a blood agar medium at pH 10 containing gentamicin, potassium tellurite, and amphotericin B. Media were tested to determine: 1) selectivity when attempting isolation from pond water and avian carcasses, 2) sensitivity for detection of low numbers of P. multocida from pure and mixed cultures, 3) host range specificity of the media, and 4) performance compared with standard blood agar. With the new selective enrichment procedure, P. multocida was isolated from inoculated (60 organisms/ml) pond water 84% of the time, whereas when standard blood agar was used, the recovery rate was 0%.
