A kinase able to phosphorylate exogenous protein synthesis initiation factor eIF-2 alpha is present in lysates of mengovirus-infected L cells.

Infection of mouse L929 cells by mengovirus resulted in the expression of a kinase activity that selectively phosphorylated the small, 38,000-molecular-weight subunit of eucaryotic initiation factor 2 and histone H2. This kinase activity was independent of host cell RNA synthesis and was located in the postribosomal supernatant (S-100 fraction) early after infection (up to 3 h). At later times after infection (5 h), kinase activity was also associated with the polysome fraction. The kinase present in the S-100 fraction bound strongly to DEAE-cellulose, its peak activity eluting at 0.5 M KCl. Kinase activity was independent of the presence of exogenous double-stranded RNA, and KCl at concentrations greater than 0.1 M inhibited eucaryotic initiation factor 2 phosphorylation. The 67,000-molecular-weight phosphoprotein activated in interferon-treated cells by double-stranded RNA was not detected by standard phosphorylation assays in lysates from mengovirus-infected cells. Labeling of this protein in vivo during 5 h of infection was also not detected. The DEAE-cellulose-purified mengovirus kinase inhibited protein synthesis in reticulocyte lysates, and the inhibition was not reversible by high concentrations of poly(I).poly(C).

Staining of elastic tissue by Verhoeff's iron hematoxylin.

The elastic system of normal human skin was studied by light and immunofluorescence. Oxytalan, elaunin et elastic fibers were stained by weigert, orcein, gomori and Verhoeff. By Verhoeff's method all elastic system of dermis is shown, with other three stains, only elaunic and elastic fibers are well visualized. These results are confirmed by immunofluorescence studies.

Fibroblast changes in cutaneous ageing.

With ageing there are progressive modifications in the connective tissue of the dermis. In ten young subjects the collagenous bundles are thick and the fibroblasts are active cells in close contact with collagen fascicles. In ten elderly subjects collagen fibres are fragmented and the fibroblasts are quiescent, without any contact with collagen. In ageing the most important lesion is the destruction of the relationship between fibroblasts and interstitial matrix. A role for fibronectin in this adhesion is suggested: in old subjects the papillary network of fibronectin is poorly developed. Furthermore, in the fibroblast we can see architectural changes in the cytoskeleton; this modification breaks up the cytoskeleton - plasma membrane - fibronectin unit and explains the secretory and metabolic changes observed in ageing, the dysfunction of the cell-interstitial matrix unit, and also the structural changes.

Dermis ageing.

On ageing major changes within connective components of the dermis can be seen: collagen looses its regular and fascicular appearance, while ground substance increases, elastic material decreases and the fibroblast cell population becomes "at rest". The resulting dermal ageing is different according to individuals and is related to genetic back ground and exposition to multiple aggressions. This review presents these changes of the superficial dermis and the role of fibroblasts in the conservation of the connective tissue integrity.

[Localization of apoprotein B in human dermis. A study using optical and electronic immunoprocedures].

The structural localization of apo B, the major protidic constitute of low density plasma lipoproteins, was investigated in human dermis using an immunofluorescence procedure and an immunoperoxidase technique. Apo B was found attached to plasmic fibroblast membranes and along collagen fibers. Dermal areas containing apo B were more numerous in patients with coronary disease than in subjects of the same age with healthy coronary vessels. These results are consistent with those of biochemical determinations: concentrations of apo B in the aorta and skin are parallel and vary according to the severity of atheromatous lesions.

Striae: morphological aspects of connective tissue.

The study of 15 abdominal striae in women aged 25 to 57 shows important histological modifications in the skin. The collagen is fragmented and the ground substance is abundant. Fibroblasts are globular, quiescent, and lose all signs of fibrillar secretion. In the light of the recent biochemical data, our results suggest that the striae are the consequence of fibroblastic dysfunction, due to abdominal distension. Comparison with scarred and normal skin indicate that striae are a special entity belonging to the group of connective dystrophies.

The effect of beta-aminoproprionitrile on the lingual artery and aorta of the rat: a light microscopic study.

The effects of chronic toxicity of beta-aminoproprionitrile (BAPN) on the lingual artery and aorta of the rat were compared. Thirty-two three week old weanling male white Wistar rats were given 1 g/kg/day (for five days weekly) of a 40% saturated solution of BAPN fumarate by intragastric tube. The animals were divided into four groups of eight animals each and sacrificed after one, three, six and nine weeks respectively. For each group, four controls were used. Cross-sections of the lingual artery and aorta stained with hematoxylin-erythrosin-safranin, alcian blue, Masson's trichrome and Verhoeff's iron hematoxylin were studied. BAPN had no effect on the lingual artery. Early changes were seen in the aorta at six weeks. Characteristic lesions were found at nine weeks involving the elastic tissue, myocytes and ground substance. The elastic lamellae were thinner, lost their normal orientation, and became swollen and finally fragmented. The myocytes of the media changed orientation and migrated into the intima. The proteoglycans increased and fibrosis supervened at the end stage. Failure to induce lesions in the lingual artery may be explained by early structural maturation and a low metabolic turnover rate in muscular arteries, decreased exposure to or greater protection from mechanical stress.

Long term lathyrism and atherogenic diet in the rat. Protective action of pyridinol carbamate.

In the rat, prolonged administration (7 months) of Beta-Aminopropionitrile in association with a hyperlipidic diet caused the formation of widespread pronounced atheroma. The addition of Pyridinol Carbamate during the treatment minimized and retarded the appearance of lipid overload lesions. The histological modifications were found together with an increase in the free cholesterol fraction. These two observations explain the protective role of Pyridinol Carbamate on the wall of the aorta.

Chronic lathyrism and atheromatosis in the rat. Protective effect of metformin.

Chronic administration of beta-aminopropionitrile fumarate (BAPN) (1 g/kg/day for 9 weeks) produced morphological changes of the aorta wall and in dermis, and biochemical changes of the aortic wall in the rat. Chronic administration of BAPN did not produce plasma lipid abnormalities such as those due to hyperlipidic diet. 9 weeks of BAPN followed by 10 months of a hyperlipidic diet increased the aortic cholesterol level and induced atheroma. The diet alone only produced an endothelial lipid overload and increased the aortic cholesterol level but less than BAPN and hyperlipidic diet together. Addition of metformin to BAPN prevented the formation of atheromatous lesions in the aorta and minimized the level of lipids in the aortic wall and of the dermis.

Age dependent biochemical changes in dermal connective tissue. Relationship to histological and ultrastructural observations.

A biochemical study of skin biopsies of young normal subjects and two groups of aged subjects, one active and the other confined to bed, was made and the results compared with histological observations. Skin biopsies were incubated with 3H proline and 14C glucosamine and connective tissue components were sequentially extracted. In each extract the specific activity of total and collagenous protein was determined. With age a decrease was observed in both the amount of protein and the 3H proline incorporation of collagenous extracts. A parallel increase in 14C glucosamine incorporation was also noticed in extracts containing glycosaminoglycans and structural proteins. These results agree in part with the changes observed by light and electron microscopy, which revealed an increase in ground substance and fragmentation of collagen bundles in the dermis of older subjects.

Chronic lathyrism. Plasma, aorta and rat skin treated for five months with beta-aminopropionitrile with or without high fat diet.

Previous experiments proved that white Wistar rats, resistant to spontaneous or experimental atheroma, present arteriosclerosis (parietal aorta lesions) and atheroma lesions (fat deposits) after a 9 week-treatment by Beta-aminopropionitrile (BAPN) followed by a high fat diet. The present work shows that BAPN alone, administered for 5 months, does not induced atheroma; its formation needs both lipid humoral disorders and parietal aortic lesions. It confirms certain facts noted in the aging of man and in the formation of atheroma.
