Cell surface antigens of chemically induced sarcomas of the mouse. I. Murine leukemia virus-related antigens and alloantigens on cultured fibroblasts and sarcoma cells: description of a unique antigen on BALB/c Meth A sarcoma.

As background for a serological definition of the unique antigens of chemically induced sarcomas, we have typed a series of fibroblast and sarcoma cell lines of BALB/c and C57BL/6 origin by cytoxicity and absorption tests for murine leukemia virus (MuLV)-related cell surface antigens and known alloantigens. 7 of the 17 cultured lines expressed the range of cell surface antigens associated with MuLV (GIX, GCSA, gp70, p30), and this was invariably associated with MuLV production. In nonproducer lines of C57BL/6 (but not BALB/c) origin, a MuLV-gp70-like molecule was found on the surface of fibroblasts and sarcoma cells. The alloantigenic phenotype of these MuLV+ and MuLV- cell lines was H-2D+, H-2K+, Thy-1.2+ or -, PC.1+ or -, Lyt-1.2-, Lyt-2.2-, Ia.7-, and TL.2-. A unique antigen was defined on the BALB/c ascites sarcoma Meth A with antisera prepared in BALB/c or (BALB/c X C57BL/6)F1 mice. Tissue culture lines derived from this tumor were MuLV-, which facilitated serological study of the antigen. Absorption analysis indicated that the antigen was restricted to Meth A; it could not be detected in normal or fetal BALB/c tissue MuLV+ or MuLV- fibroblast lines, 12 syngeneic or allogeneic sarcomas, or normal lymphoid cells from 13 different inbred mouse strains.

A new hemoglobin beta chain variant in sheep.

A new hemoglobin beta chain variant referred to as Hb-E is observed in four Indian sheep breeds. Hemoglobin E is electrophoretically and chromatographically undistinguishable from hemoglobin B. However, polypeptide chain separation by urea-polyacrylamide disc gel electrophoresis and urea CM-cellulose chromatography reveals the new beta chain. The betaE chain has a slower anodic mobility than betaB chain at alkaline pH. The adult sheep hemoglobins could be classified into eight categories, namely AA, AB, AE, AC, ABC, BB, BE and EE, according to their sub-unit composition. The occurrence of betaE chain in homozygous condition or in combination with either one of the two allelic beta chains betaA or betaB, indicate that it may be an allelic variant. Gene frequencies of three hemoglobin beta chain alleles observed during the study are as follows: A = 0.192; B= 0.682; E = 0.126.

The nitroblue tetrazolium test in juvenile rheumatoid arthritis and the stimulation of granulocytes by patients sera.

The spontaneous NBT test was used in 57 children with juvenile rheumatoid arthritis (JRA) and another connective tissue diseases. The patients were grouped according to the various manifestations and divided into different stages of activity. The patients with tendosynovitis and unknown arthralgia showed a negative test. Patients with morbus Wissler without exudative joint manifestations were also NBT-negative, to a large degree. By contrast, all manifestations with joint involvement showed a high per centage of positive results in the NBT test. In a second investigation, the granulocytes of normal persons were stimulated by sera of patients with various manifestations of JRA. The sera of patients with joint manifestations all caused a stimulation of the granulocytes of normal persons, compared with the controls, in which pooled human serum was employed. Stimulation of granulocytes with serum of Wissler patients proved impossible, however.

Endodermal sinus tumors of the ovary in children. The role of radiation therapy in relation to the clinical course.

Endodermal sinus tumor of the ovary in children has been characterized by its general resistance to therapy and its almost uniform fatality. The multidisciplinary treatment of 5 such cases is described. The author's experience suggests that these tumors are responsive to irradiation. The rationale for a combined approach is discussed, and radiation treatment volumes and doses are suggested.

[Studies with the NBT test in various clinical forms of juvenile rheumatoid arthritis].

Fifty-six healthy neonates and children as well as 33 patients with juvenile rheumatoid arthritis and other diseases of the locomotor system were examined using a modification of the NBT test. In healthy newborn infants there was observed an increase in the number of NBT-positive cells between the first and tenth days. The test results obtained for the 33 patients with juvenile rheumatoid arthritis were positive in 73%, with positive results being obtained especially in those patients where the disease was in its second stage of activity. Highest values were obtained in patients with a predominantly articular manifestation of the disease. Stimulative effects produced by immune complexes are considered to be chiefly responsible for this. However, this method of examination is of small practical value from a differential diagnosis point of view.
