Analysis of the glycoproteins of murine tumour cell lines with 125I-concanavalin A in two-dimensional electrophoresis gels.

The combination of two-dimensional gel electrophoresis and post-electrophoretic staining with 125I-labelled concanavalin A was used to compare the glycoproteins of murine tumour cell lines. Comparison between different cell lines showed that there were about eight common glycoproteins. The rest of the glycoproteins were generally unique to particular cells. Thus the P815 cell could be distinguished from 13 other murine cell lines by its glycoprotein pattern. The specific glycoproteins of each cell line were unaffected by culture in vivo, virus infection or hybridisation. Different clones from the same cell line gave identical patterns. Crude membrane preparations and glycoproteins purified from cell lysates by affinity chromatography on concanavalin/agarose gave the same patterns as whole cells. Thus the glycoproteins of murine tumour cells appear to be a stable characteristic which can provide specific markers for the identification of tumour cell lines.

Leukotriene B4: biological activities and the cytoskeleton.

The aggregation and chemokinesis of either rat or human polymorphonuclear leucocytes (PMNs) induced by leukotriene B4 isomer III (LTB4) are inhibited significantly by colchicine (10(-6) - 10(-3)M) and vinblastine (10(-7) - 10(-4)M). Random migration of the leucocytes is inhibited by colchicine 10(-3)M and vinblastine 10(-4)M. Cytochalasin B (4 X 10(-7) - 4 X 10(-6)M) caused the aggregation of rat PMNs but inhibited their random migration. The aggregation of the PMNs induced by LTB4 was enhanced by cytochalasin B but the chemokinesis was inhibited. It is suggested that both microtubules and microfilaments may be involved in the aggregatory and chemokinetic responses of PMNs to LTB4.

Silicate nephrotoxicity in the experimental animal: the missing factor in analgesic nephropathy.

Three silicon-containing compounds (magnesium trisilicate BP, crushed quartz and crushed Arran granite) were added to the drinking water (250 mg/l) of three groups of male guinea pigs for four months. At autopsy all animals receiving magnesium trisilicate showed a focal tubulo-interstitial nephritis mainly affecting the distal nephron. Similar but less intense lesions were encountered in two animals receiving crushed quartz. No renal lesions were found in the control group or in animals receiving crushed Arran granite. The concentration of soluble silicates in drinking water, measured as elemental silicon (Si) by atomic absorption spectroscopy (AAS) was determined for twenty-eight locations in different regions of Britain. Following ingestion of magnesium trisilicate, significant increases in urinary excretion of Si were demonstrated in two healthy adults using atomic absorption spectroscopy.

Complete nucleotide sequence and organization of the Moloney murine sarcoma virus genome.

The complete nucleotide sequence of a mammalian transforming retrovirus. Moloney murine sarcoma virus, has been determined. MSV, recombinant virus derived of helper viral and cellular sequences, possesses termini resembling prokaryotic transposable elements. The viral genome has the coding capacity for the Moloney murine leukemia virus gag gene product and contains large deletions in pol and env genes. A large open reading frame encompassing its cell-derived sequences codes for its putative transforming protein. The nature of some of the important domains in the viral genome has been established, and their structure is discussed in relation to their function.

Biological activities of leukotriene B4.

Leukotriene B4 isomer III is released from polymorphonuclear leucocytes, monocytes, eosinophils and macrophages in vitro and has been detected and measured in human synovial fluid in vivo. Its most prominent biological activities are to induce the aggregation of and to stimulate the movement (chemokinesis and chemotaxis) of leucocytes in vitro and it acts as a cytotaxin in vivo. In addition, it increases vascular permeability in vivo when administered together with the vasodilator, PGE2.

Aggregation of polymorphonuclear leucocytes (PMNs) by leukotriene B4: effects of cyclooxygenase products and metabolic inhibitors.

Leukotriene B4 isomer III (LTB4) stimulates the aggregation of rat PMNs in vitro. The effects of deoxyglucose, iodoacetate, dinitrophenol, cyclohexamide, colchicine, prostaglandins and thromboxane B2 (TXB2) on aggregation were examined. The results demonstrate that, first, aggregation is an active process, the energy being supplied by glycolysis and not mitochondrial respiration. The response can be elicited in the absence of extracellular glucose. Synthesis of protein does not occur during aggregation. An intact microtubular system is required for the expression of a full aggregation response. Prostaglandins E1, E2, F2 alpha and TXB2, products of the cyclooxygenase pathway of arachidonic acid metabolism, partially inhibit LTB-4 induced aggregation by a mechanism which has not yet been elucidated. The prostaglandins and TXB2 themselves do not promote aggregation.

Leukotriene B4: an inflammatory mediator in vivo.

Leukotriene B4 (LTB4 isomer III), which promotes the movement and aggregation of leucocytes in vitro also stimulates the chemo-attraction of leucocytes and their adherence to vascular endothelium in vivo. These effects were observed directly in the hamster cheek pouch preparation and on histological examination of sections from the rabbit mesentery. Intravenous injection of LTB4 (isomer III) into the rabbit resulted in a profound but transient neutropenia. Intradermal injection of LTB4 (isomer III) in the rabbit produced a rapid accumulation of neutrophils and this effect was also observed in skin chambers applied over abrasions on the rabbit back or the human forearm.

Isomers of leukotriene B4 possess different biological potencies.

Rat elicited polymorphonuclear leucocytes (PMNs), when exposed to the ionophore A23187, release three isomers of leukotriene B4. The three isomers have been purified and tested for their ability to induce the chemokinesis of human PMNs in vitro, the aggregation of rat PMNs in vitro and changes in vascular permeability in rabbit skin in vivo in the presence of PGE2. The results demonstrate that all three isomers are biologically active and that the enzymatically produced isomer, in which the conjugated triene contains one cis and two trans double bonds, is more potent than the two diastereoisomers of LTB4 which contain all trans double bonds in the conjugated triene and which are produced by non-enzymatic hydrolysis.

Leukotriene B4: a mediator of vascular permeability.

1 Intradermal injection of leukotriene B4 (LTB4) or prostaglandin E2 (PGE2), 1 to 100 ng per skin site produced little or no change in plasma exudation in the rabbit, guinea-pig and rat. 2 Intradermal injection of LTB4 or PGE2 together with bradykinin (500 ng) resulted in a significant potentiation of the plasma exudation produced by bradykinin alone in the rabbit and guinea-pig. 4 LTB4 (1 to 10 ng) had no effect on blood flow in the rabbit skin, in contrast to PGE2 which was a potent vasodilator in this species. 5 It is concluded that LTB4 is a mediator of vascular permeability and that this effect can only be observed in the presence of a vasodilator such as PGE2.

Association of HLA B8 with spontaneous resolution in sarcoidosis.

HLA may influence the natural history of some diseases. HLA frequencies have been compared in 164 healthy control subjects, 50 patients with sarcoid lung fibrosis, and 37 patients with sarcoidosis that resolved spontaneously. B8 was increased significantly in the resolved group compared to both healthy control subjects (p less than 0.001) and the fibrotic group (p less than 0.01). The results support the findings of other investigators, and are consistent with the hypothesis that inherited host factors, perhaps related to immune response, influence the clinical expression of sarcoidosis.
