Studies on nuclear estradiol receptors in human mammary carcinomas.

Nuclear estradiol receptors (NER) were measured in human breast carcinomas by extracting nuclei with 0.6 mol/l KCl and adsorption to hydroxylapatite. NER were found in 34/58 tumors containing cytoplasmic estradiol receptors (CER) and in only 8/34 tumours without CER. NER correlated with CER but not with cytosol total estrogens or progesterone. NER levels also showed no significant correlation with either tumor stage, grade, cellularity, size, or presence or absence of axillary node metastases.

Assay of estradiol receptors in human breast carcinomas using the gamma emitting ligand 16 alpha [125I]iodoestradiol.

Estradiol receptors (ER) were measured in human breast carcinomas using the gamma emitting ligand 16 alpha [125]iodoestradiol. Competition experiments with excess unlabeled steroids showed that the iodoestradiol was bound specifically by the ER. The K4 of the iodo and tritium-labeled estradiol for the receptor were similar with values of 3.4 x 10(-18) M and 2.1 x 10(-10) M respectively. A comparison of ER determination in 25 human breast carcinomas using both [3H]estradiol and [125I]estradiol gave a correlation coefficient value of 0.97. It is concluded that the use of [125I]estradiol should greatly simplify the ER assay and facilitate its introduction into the routine hospital laboratory.

Peroxidase activity in human breast carcinomas: correlation with histological and biochemical parameters.

Peroxidase activity was found in 68% of 56 human breast carcinomas. Median activity was higher in ductal carcinomas than in tumors with lobular components. Activity did not correlate with tumor grade or stage, or the presence or absence of axillary node involvement. Activity, however, correlated with tumor cellularity, lymphocyte infiltration levels, and total protein and alkaline phosphatase levels. It is concluded that variation in peroxidase activity in human breast tumors is at least in part due to lymphocyte infiltration. Furthermore, this contamination of breast tumors with lymphocytes may explain the lack of correlation between estradiol receptor levels and peroxidase activity.

Serum sialic acid and CEA concentrations in human breast cancer.

The concentration of bound sialic acid in the sera of 56 normal subjects and 65 subjects with breast cancer was measured, in order to determine (1) whether serum sialic acid concentrations are raised in breast cancer and (2) whether the concentration of sialic acid in serum reflects tumour stage. The amount of sialic acid in serum was compared to serum carcinoembryonic antigen (CEA) values. Urinary hydroxyproline and serum alkaline phosphatase concentrations were used as indicators of bone and liver involvement. Erythrocyte sedimentation rate (ESR) was also measured. Significantly elevated serum sialic acid concentrations were found in breast cancer, and showed correlation with tumour stage. Serum sialic acid values did not correlate with CEA values. The results suggest that measurement of serum sialic acid concentrations may be of adjunctive value in assessing tumour stage.
