Thalamic degeneration in X-chromosome--linked copper malabsorption.

Thalamic degeneration was present in 5 autopsied cases of X-chromosome-linked copper malabsorption (X-cLCM), Menkes' kinky hair disease. Among the thalamic nuclei, those in the formatio paraventricularis, intralamellaris, and extralamellaris were spared. The nuclei projecting to the granular cortices had severe neuronal depopulation. The thalamic nuclei that send axons to the agranular cortices were less often and less severely involved. The thalamic afferent system was intact except for degeneration of the red nucleus. Cerebral cortical lesions varied from case to case and usually were less marked than thalamic neuronal changes.

Degeneration of the cerebellar system in X-chromosome-linked copper malabsorption.

Examination of the cerebellar system of 5 autopsied patients with X-chromosome-linked copper malabsorption led to the discovery that among cerebellar afferent systems, only the dorsal spinocerebellar tract showed consistent degenerative changes. Cerebellar cortical lesions comprised granule cell loss and Purkinje cell abnormalities that included nuclear and cytoplasmic degenerative changes with cell loss and deficient dendritic arborization as well as the presence of somatic sprouts. Perisomatic baskets were often absent in the area of sprouting Purkinje cells. Cerebellar cortical topographical analysis revealed relative sparing of the caudal portion of the cerebellum. The nodulus was intact in all patients. The dentate nucleus and the superior cerebellar peduncle were preserved. The red nucleus was consistently degenerated.

Sparing of the Onufrowicz nucleus in sacral anterior horn lesions.

Sacral anterior horns were examined with special reference to the Onufrowicz nucleus in 3 patients with Werdnig-Hoffman disease, 2 with healed anterior poliomyelitis, and 1 who had undergone amputation of both legs above the knee. None of these patients showed symptoms of either urethral or anal sphincter disturbances. The Onufrowicz nucleus of the second sacral anterior horn appeared normal in all the patients examined, though the lateral portion of that horn and the medial nuclear group of the third sacral segment were frequently involved. These findings support the view that the Onufrowicz nucleus may be innervating the external sphincter muscle of the urethra and the anus.

"Glial bundles" in the spinal cord late after paralytic anterior poliomyelitis.

"Glial bundles" in the proximal portion of the anterior spinal roots, identical to those in Werdnig-Hoffmann disease, were observed in 3 patients many years after attacks of acute paralytic anterior poliomyelitis. These structures should therefore be considered a nonspecific astrocytic reaction secondary to the destruction of motor neurons and their axons rather than a malformation specific for Werdnig-Hoffman disease.

Localization of olivo-cerebellar fibers in inferior cerebellar peduncle in man.

The localization of the olivo-cerebellar fibers in the inferior cerebellar peduncle was examined in a patient who showed severe degeneration of the left olivary nucleus due to an old hemorrhagic infarct in the contralateral cerebellum. Other precerebellar nuclei which send their cerebellipetal axons to the inferior cerebellar peduncle, such as the external cuneate nucleus, the lateral reticular nucleus and the arcuate nucleus, were normally preserved. The dorsal spino-cerebellar tract and the vestibular nuclei, together with the juxtarestiform body were also intact. Degeneration of the inferior cerebellar peduncle was noted only in its ventromedial portion contralateral to the olivary lesion. As a result, it is concluded that the olivo-cerebellar fibers seem to be located in this portion of the inferior cerebellar peduncle.

Immunological studies on CS-1170, a new semisynthetic cephamycin antibiotic.

The antigencity of CS-1170, a newly developed semi-synthetic cephamycin, and it's cross-reactivity with beta-lactam antibiotics were studied. The antigencity was confirmed by the following tests: 1)the passive hemagglutination test with anti-CS-1170 antisera of guinea pigs immunized with CS-1170 plus Freund complete adjuvant, 2)lymphocyte proliferation response in vitro stimulated with this drug in the guinea pigs, 3) the PCA test using mice immunized with CS-1170-protein conjugates. The antibody to CS-1170 appears to be directed to the acyl side chain because cyanomethylthioacetylglycine, an univalent acyl side chain of the CS-1170, can significantly cross-react with the antibody. A methoxy group on the C-7 alpha-position of the antibiotic plays no important part for the antigenic specificity of the molecule. The cross-reactivity of CS-1170 with cefazolin, cephalothin, benzylpenicillin and ampicillin was only to a minimal extent in the passive hemagglutination test. The cross-reactivity of CS-1170 and the related antibiotics was not observed among them in the PCA system with IgE anti-CS-1170. These findings support a conclusion that CS-1170 is one of beta-lactam antibiotic with an immunologically minimal cross-reactivity to the related antibiotics.
