[Acetaldehyde adducts in the cerebral cortex of ethanol-fed mice].

To investigate the neurotoxicity of acetaldehyde covalent adducts, immunohistochemical staining for acetaldehyde adducts using the antibody against acetaldehyde adducts, was performed in the cerebral cortex of ethanol-fed (withdrawal) mice. In the ethanol-fed mice, the degeneration in the cerebral cortex was found, while the protein epitope related to acetaldehyde was found in the cerebral cortex, liver and adrenal cortex. No histochemical and immunohistochemical changes in the tissues from the control mice were found. It is possible that acetaldehyde adducts may effect on the cerebral cortex as the neurotoxicity which cause psychosis such as delirium and hallucination after alcohol drinking.

Effects of football training on ventilatory and gas exchange kinetics to sinusoidal work load.

The purpose of this study was to evaluate the effects of football training on maximal oxygen uptake (VO2max), ventilatory threshold (VT) and kinetics of ventilation and gas exchange variables to sinusoidal work load. The sinusoidal work load during cycling exercise was varied from 30 watts to 60% of VO2max (60% VO2max) with a period of 2 min. O2 uptake (VO2), CO2 output (VCO2), minute ventilation (VE) and heart rate (HR) were measured on a breath-by-breath basis using a computer system. Training periods were continued for 9 months. Six males who had no football experience were performed in which VO2max, VT, and the kinetics of each variable were measured for 3-month intervals (PRE, TR.3, TR.6, and TR.9). They usually underwent football training, such as sprint and strength training for 2-3 hours day-1, 6 days week-1. Mean VO2max was significantly increased at TR.3 and TR.6. VT was also significantly increased during the training period. The amplitude of VO2, VCO2, VE, and HR responses during sinusoidal exercise unchanged during the training period. Phase shifts to work load in VO2 and HR responses did not significantly change during the 9 months, but the phase shift in VCO2 and VE responses significantly continued increasing as the training intervals progressed. These results suggest that football training does not significantly affect the development of the kinetics of VO2 and HR during submaximal exercise, but that it dramatically increases VO2max and VT values.

[The present and future of clinical physiologic tests].

The symposium on clinical physiologic tests was designed in the 44th congress of all central laboratory divisions of national university hospitals at 28, 29 May in 1997. The title of symposium was "the present and future of clinical physiologic tests" which was named by professor M. Itoh. Four symposists were nominated among the specialists on clinical physiologic tests of members of above congress. Professor M. Itoh and Professor H. Kawaguchi spoke about Erectric Cardio Graphy (ECG) and main heart function tests and Professor M. Kambe spoke about pulmonary function tests and Professor M. Okada spoke about the using methods of clinical physiologic test data in remote and distant medicine. The special feature of clinical laboratory division in Japanese large hospitals is that has clinical physiologic tests part and the clinical laboratory division of hospitals in foreign countries has no clinical physiologic tests part. So, we expect more development of clinical physiologic tests part in nearly future.

Positional reproducibility of the patient's head normalized by a robotized chair-based automatic head-positioning system, Racs-N.

With a view to normalization of the head orientation without the use of a fixation jig for the measurement of jaw movement employing an X-ray television, we have been developing an automatic head-positioning system, Racs-N, consisting of a head-position sensor and a robotized chair. The head position is monitored in 6 degrees of freedom (d.o.f.) by using a position-sensitive detector (PSD) camera-based photostereometry. If they are incorrect, the robotized chair rectifies them automatically by adjusting the position and orientation of the head support in 5 d.o.f. (except the vertical position). The rectified position, however, has a certain margin of error, due to the mobility of the head relative to the head support. To improve this, we employed an iterative correction in that the positioning is repeated until the determined position converges. In experiments employing several subjects, this particular correction was verified to converge by more than six iterations and to be effective for the reduction of head-position errors.

Direct intraperitoneal insemination in ovarian hyperstimulation cycles induced with gonadotropin-releasing hormone agonist.

Direct intraperitoneal insemination (DIPI) is one of the least invasive strategies of assisted reproduction. Unexplained infertility, cervical factors and male sub-fertility are major indications for DIPI. This is a report of pregnancy that occurred as a result of a novel trial of DIPI, in which ovarian stimulation took place using a long gonadotropin-releasing hormone (GnRH) analogue and human menopausal gonadotropin (HMG) protocol in a patient who had been unsuccessfully treated with intrauterine insemination.

Factors affecting the development of oocytes and embryos in IVF-ET.

In our study in hMG/hCG protocol, there is correlation between basal FSH level and number of oocyte retrieved (Rs = -0.4334, p = 0.039). FSH was not useful in predicting poor responders in an in vitro fertilization program where pituitary desensitization is routinely used. There is correlation between day 2, day 3 serum immunoactive inhibin level and number of follicle developed with hMG alone, or with hMG in conjunction with pituitary desensitization using a buserelin intranasal spray. The patients were divided by relationship between E2 peak and inhibin peak. The most of oocytes retrieved, oocytes fertilized and embryos cleaved were obtained when hCG was injected on the E2 peak day following the inhibin peak day. Inhibin concentration in aspirated follicular fluids was low and estradiol concentration was high in pregnant group. There is correlation between acceleration index of inhibin and number of oocyte retrieved in both protocols. Inhibin levels were significantly lower (P < 0.05) in women older than 36 years of age, although estradiol responses were not so influenced by age. These data suggest that: (1) In the early follicular phase, inhibin may be a valid index to predict ensuing follicular growth and follicular development. (2) The serum inhibin responses to maximal ovarian stimulation may be a sensitive and early index of declining ovarian function with advancing age. (3) Inhibin is an important factor for the diagnosis of pregnancy outcome.

Suppression of actinomycin D-induced apoptosis by the NS3 protein of hepatitis C virus.

The NS3 protein of hepatitis C virus is a multifunctional protein that is indispensable for virus replication. Little is known, however, about the possible effects of the NS3 on host cell function(s). In the present study, we demonstrated that NIH3T3 cells constitutively expressing a carboxy-terminally truncated NS3 (NS3DeltaC) were more resistant to actinomycin D-induced apoptosis than the control cells. We also observed that induction of p53 expression by actinomycin D treatment was weaker in the NS3DeltaC-expressing cells than in the control cells. However, induction of WAF1 expression by the same treatment was not different between the two groups. Taken together, our results suggest the possibility that expression of NS3DeltaC suppressed actinomycin D-induced apoptosis of NIH3T3 cells through at least partly, if not solely, a p53-dependent, WAF1-independent pathway.

Genomic structure of human BST-1.

BST-1 is an ectoenzyme expressed on human bone marrow Stromal cells and myeloid lineage cells, having both ADP-ribosyl cyclase and cyclic ADP-ribose (cADPR) hydrolase activities. In mouse, BST-1 is also expressed on lymphoid progenitors. We isolated chromosomal DNA segments of the human BST-1 gene. The human BST-1 gene consisted of nine exons and eight introns. The length of each exon was very similar to that of the Aplysia ADP-ribosyl cyclase gene. The flanking region of the BST-1 gene contained several potential binding sites for nuclear factors, NF-kappa B, p53, NF-IL6, CREB, PEA3, E2A, C/EBP, AP3, AP2 and SP1 and consensus sequences for gamma-IRE and ISRE like element.

Human BST-1 expressed on myeloid cells functions as a receptor molecule.

We have previously identified and cloned BST-1 as a molecule which is overexpressed on the bone marrow stromal cell lines derived from patients with rheumatoid arthritis and which has the ability to support the pre-B cell growth. BST-1 is a glycosylphosphatidylinositol-anchored ectoenzyme having ADP-ribosyl cyclase and cyclic ADP-ribose hydrolase activities. In this report, we demonstrate that human BST-1 was expressed on monocytes, granulocytes in the peripheral blood of healthy donors, and macrophages matured in vitro. Cross-linking of BST-1 with a polyclonal anti-BST-1 antibody induced tyrosine phosphorylation of a 130-kDa protein (p130) in the human myeloid cell lines U937 and THP-1. Cross-linking of BST-1 overexpressed on a transfectant induced tyrosine phosphorylation of p130, dephosphorylation of the 100-kDa protein, and growth inhibition. These results suggest that BST-1 can deliver signals into cells and function as a receptor.

High-efficiency full-length cDNA cloning by biotinylated CAP trapper.

We have devised a method for efficiently constructing high-content full-length cDNA libraries based on chemical introduction of a biotin group into the diol residue of the cap structure of eukaryotic mRNA, followed by RNase I treatment to select full-length cDNA. The selection occurs by trapping the biotin residue at the cap sites using streptavidin-coated magnetic beads, thus eliminating incompletely synthesized cDNAs. When this method was used to construct a mouse brain full-length cDNA library, our evaluation showed that more than 95% of the total clones were of full length, and recombinant clones could be produced with high efficiency (1.2 x 10(7)/10 micrograms starting mRNA). The analysis of 120 randomly picked clones indicates an unbiased representation of the starting mRNA population.

Positron emission tomography study of the alterations in brain distribution of [11C]methamphetamine in methamphetamine-sensitized dog.

We newly prepared a MAP-sensitized dog by repeated MAP treatment and studied the brain distribution of [11C]MAP in the normal and the MAP-sensitized dog using PET. The maximal level of accumulation of [11C]MAP in the sensitized dog brain was 1.4 times higher than that in the control. No difference was found in the metabolism of MAP between the two conditions. The significant increase of [11C]MAP in the MAP-sensitized brain indicates that subchronic MAP administration causes some functional change in the uptake site of MAP. The pharmacokinetic change may, in part, account for behavioral sensitization.

Localization of sympathetic and parasympathetic neurons innervating pancreas and spleen in the cat.

The localization of sympathetic and parasympathetic neurons innervating the pancreas and spleen was studied in the cat utilizing retrograde transport of wheat germ agglutinin conjugated horseradish peroxidase (WGA-HRP). Injection of WGA-HRP into the pancreas resulted in retrograde labeling in the whole of the solar plexus, while injection of WGA-HRP into the spleen also resulted in heavy labeling in the celiac ganglia bilaterally. Only a few labeled neurons were distributed in the superior mesenteric ganglion. With respect to parasympathetic innervation, HRP-labeled pancreatic and splenic neurons were found throughout the rostrocaudal extent of the dorsal motor nucleus of the vagus nerve (DMV) bilaterally. Although pancreatic neurons in the DMV were mainly observed in limits rostral to the obex, splenic neurons were centered at the level of the obex.

Immunocytochemical detection of p53 protein from pancreatic duct brushings in patients with pancreatic carcinoma.

BACKGROUND: It is often difficult to distinguish pancreatic carcinoma preoperatively from chronic pancreatitis. Therefore, we have developed a new method of detecting p53 immunoreactivity in cytologic material obtained by endoscopic retrograde pancreatic duct brushing (ERPDB). METHODS: Twenty-eight patients with prominent strictures of the main pancreatic duct demonstrated by pancreatography including 20 ductal cell carcinoma and 8 chronic pancreatitis were studied. The ability to distinguish between these two groups preoperatively by conventional cytologic examination was compared with p53 immunocytochemistry using ERPDB: RESULTS: The sensitivity, specificity, and overall accuracy of conventional cytologic examination in distinguishing ductal cell carcinoma from chronic pancreatitis were 60%, 100%, and 71% respectively. In comparison, the sensitivity, specificity, and overall accuracy of p53 immunocytochemistry in distinguishing were 90%, 100%, and 93%, respectively. The sensitivity of p53 staining of specimens from patients with carcinoma of the body or tail of the pancreas (90%) was the same for those with tumors of the head of the pancreas (90%). CONCLUSIONS: These results suggest that p53 immunocytochemistry using ERPDB in conjunction with conventional cytologic examination can help differentiate ductal cell carcinoma from chronic pancreatitis preoperatively.
