[Acquired idiopathic sideroblastic anemia (author's transl)].

The authors consider the clinical and biological data of Acquired Idiopathic Sideroblastic Anemia (AISA). The physiopathology of the syndrome is discussed; the relationships between pathologic sideroblastosis, dyserythropoiesis and ferrokinetic modifications are pointed out. The associated abnormalities of granulocytic and megacaryocytic series linked AISA to other myelodysplasia.

[Persistence of orientation-selective cells of the primary visual cortex in kittens enucleated unilaterally at birth and reared in darkness].

In kittens dark reared (6 weeks old) orientation selective cells are no longer recorded in the primary visual cortex, while in kittens of same age, enucleated at birth unilaterally and reared in identical conditions, 30% of visual cortical cells are shown to be orientation selective and in addition respond preferentially to horizontal and vertical orientations.

Early development of visual cortical cells in normal and dark-reared kittens: relationship between orientation selectivity and ocular dominance.

1. 535 units were recorded in the primary visual cortex in twelve normally reared and fifteen dark-reared kittens aged between 8 and 50 days. These results were pooled with a previous study of 582 units recorded from thirty-five kittens reared in similar conditions. 2. These 1117 cells were classified into four functional classes of neurones: (a) visually unresponsive cells, (b) non-specific cells which were sensitive to spots or slits of light moving in any direction, (c) immature cells which were preferentially activated by a rectilinear stimulus but unselective regarding its precise orientation and (d) specific cells that appeared to be as selective for orientation as the simple or complex cells in the adult cat. 3. The results confirm that cells having the same orientation-specific response properties as adult cortical visual neurones are present in the earliest stages of post-natal development, independently of visual experience. However, to maintain and develop these specific cells after the third week of post-natal life, visual experience is necessary. 4. The ocular dominance of visual cells is not constant from the earliest stages of development. A significant increase in binocularly driven neurones occurs with age. 5. Before 3 weeks of age, whatever the rearing conditions, there are more specific cells coding horizontal and vertical orientations than those coding oblique orientations. These 'horizontal and vertical detectors' are preferentially driven by the contralateral eye. 5. After 4 weeks of age, specific neurones are found at all orientations in normally reared kittens. At this stage of development the ocular dominance is independent of orientation preference, of the functional class of neurones considered and of the rearing conditions. The proportion of binocularly driven cells is slightly below adult standard. 7. A hypothesis of differential plasticity is proposed: contralateral, monocular 'horizontal and vertical detectors' are supposed to be stable; they would remain so until they become binocular. Binocular cells, for which competition between two inputs occurs, are the labile units which can be despecified or specified under the control of visual experience.

[Stabilization of operant responses during a program of variable interval reinforcement as a preliminary investigation of sensory thresholds using conditioned suppression in the cat].

Cats were trained to press a key during a variable interval schedule of reinforcement with a limited hold contingency. The main modifications of the response rate were observed when the subject missed one reinforcement or obtained more than one reinforcement during the limited hold. The existence of a zone of lesser variability in the mean response rate is shown to be linked to the occurrence of the last reinforcement. This result suggests an original methodological contingency concerning the use of conditioned suppression in psychophysics. The schedule of sensory stimulation should be established for each subject during that period of lesser variability between two reinforcements.

Pure acute monocytic leukemia. A study of 12 cases.

Twelve cases of pure acute monocytic leukemia in adults were studied. They were selected on the basis of the morphology of the blast cells on Romanowsky-stained smears of blood and bone marrow, as well as positivity of the cells for the naphthol ASD acetate esterase reaction specifically inhibited by sodium fluoride. There was no sex predominance. Neoplastic involvement of the skin and/or gingiva was very frequent. The leukemic proliferation in blood and bone marrow consisted of monoblasts, promonocytes and monocytes. The peroxidase reaction was negative or only faintly positive. Serum and urinary lysozyme levels were increased. The blast cells retained their ability to stimulate, in vitro, colony formation by normal bone marrow cells used as targets. All of these characteristics permit specific identification of this type of acute leukemia. The prognosis is grim: only five of 12 patients achieved complete remission, and four of these five had relapses in less than 14 months; the median survival was five months.

Role of sodium and urea in the renal concentrating mechanism in Psammomys obesus.

Micropunctures were performed at the tip of Henle's loops and vasa recta accessible at the extrarenal surface of the papilla in a desert rodent (Psammomys obesus) studied under mild NaCl ((NaCl 4%, 0.0375 ml/min) and mild urea (urea 4%, 0.0375 ml/min) loading conditions. In Nacl loaded animals, it was confirmed that solute addition (mainly sodium) contributes in a large proportion to the concentrating process along the thin descending limb. Comparison of sodium and urea concentrations in the loops with those in vasa recta at the same level of the papilla demonstrated that 1. the transepithelial sodium gradient was compatible with a diffusion transport of this ion from the interstitium to the thin descending limb; 2. the sodium concentration higher in interstitium than in the loop fluid was not compatible with the existence of a purely passive concentrating process in inner medulla as was recently proposed [8], 3. the transepithelial urea gradient was very limited which indicates that this solute does not play an important part in the concentrating process. In urea loaded psammomys, solute addition (mainly urea) to the thin descending limb fluid was still present but water abstraction was enhanced as compared to salt loaded animals, probably on account to the higher interstitial urea concentration. It is, thus, brought to evidence that the relative contribution of water abstraction and solute addition to the concentrating process along the thin descending limb can vary in a given species as a function of the physiological state.

Functional segmentation of the rabbit distal tubule by microdetermination of hormone-dependent adenylate cyclase activity.

Rabbit distal convoluted tubules (DCT) microdissected from collagenase-treated kidneys were observed to contain up to four portions of a different appearance under stereomicroscopic examination: (1) a DCTa portion (generally very short), located right after the macula densa (MD) and resembling the portion of the limb (CAL) located before the MD; (2) a constant, "bright" portion, DCTb; (3) a constant, "granular" DCTg portion which, in most DCT, is connected to a portion of the collecting tubule of a similar "granular" appearance (CCTg); (4) many DCT having contacts with the kidney capsule in the superficial cortex were observed to contain an additional portion of a "light" appearance, DCTl, resembling the portion of the collecting tubule (CCTl) to which these superficial DCT are always branched. The hormone-dependent adenylate cyclase (AC) contained in these different portions was investigated by sectioning microdissected distal structures into successive samples according to the above-mentioned criteria, and by measuring with the help of a previously described micromethod, the enzyme activity contained in each single sample under one of the following conditions: control, parathyroid hormone. (PTH l U/ml), vasopressin, (AVP 10(-6)M), isoproterenol (10(-6)M), fluoride (5 X 10(-3)M). Highly significant and reproducible AC stimulations by these hormones were obtained for the following portions, respectively: DCTa, DCTg and CCTg with PTH; DCTl and CCTl with AVP; DCTg, CCTg and CCTl with isoproterenol. From these data, it is concluded that (a) the distal convoluted tubule can no longer be regarded as a single well-defined functional structure; (b) DCTa is actually a short CAL portion extending beyond MD, (c) DCTg and CCTg are two portions of a same functional segment; (d) similarly, DCTl belongs to the functional segment mainly constituted by CCTl; and, finally, (e) DCTb is the only functional segment which is entirely located in the distal convoluted tubule, i.e., included between the macula densa and the first branching with another tubule.

Visual cortical cells: their developmental properties in normal and dark reared kittens.

582 units were recorded in area 17 in twenty-one normally reared kittens, and fourteen dark reared ones, aged between 8 and 42 days. 2. Four classes of neurones were defined: (a) non-activable cells that cannot be excited by any peripheral stimulation; (b) non-specific cells that are sensitive to a visual stimulus moving in any direction; (c) immature cells that are preferentially excited by a rectilinear stimulus but are unselective for its precise orientation; (d) specific cells that appear to be as selective for orientation as the simple or complex cells of the adult cat. 3. 23% of the cells presenting adult characteristics of specificity in terms of directionality and orientation are present in both the normally reared and in the dark reared kittens as soon as the first visual response (12 days) appears. 4. In normally reared kittens the proportion of specific cells increases with age, while that of the non-specific cells decreases. 5. Up to 3 weeks of age there is no significant difference in the proportion of the different types of cells in the dark reared from the normal kittens. But thereafter in the dark reared kittens the specific cells tend to disappear while the non-specific cells increase in number. 6. This confirms the view that cells with some highly specific response properties of the adult visual cortical neurones, especially in relation to orientation specificity, are present in the earliest stages in the absence of all visual experience. However, visual experience is necessary to maintain and develop these specific cells after the third week of post-natal life.

[Acute monoblastic leukemia. Clinical and therapeutic aspects in 20 cases].

Twenty cases of acute monoblastic leukemia are studied according to definite criterias: cytology, cytochemical staining, lysozyme production. The study points out the tumoral characters: hematodermy, gingivitis, central nervous system leukemia and the lack of important bone marrow incompetence. 47 per cent of complete remissions are obtained with daunorubicin, aracytin combination therapy. Other useful chemical agents, and prospects for maintenance therapy are discussed.

Vasopressin dependent adenylate cyclase in single segments of rabbit kidney tubule.

AVP dependent adenylate cyclase activity was measured in single pieces of 8 different tubular segments isolated from collagenase treated rabbit kidneys. High responses were observed in all the tested portions of the collecting tubule, that is its cortical branched part (BCT), its cortical straight part (CCT) and its outer medullary part (MCT). Dose response curves indicated in CCT: 2 fold threshold stimulation at 10(-11) M AVP, 27 fold stimulation at 10(-6) M AVP, half maximal stimulation at about 10(-9) M AVP. Both the medullary (MAL) and, to a lesser extent, the cortical (CAL) portions of the thick ascending limb were also observed to contain AVP sensitive adenylate cyclase (for MAL: 2 fold threshold stimulation at 10(-9) M AVP, 9 fold stimulation at 10(-7) M AVP, half maximal stimulation at 5 X 10(-9) M AVP). In contrast, nearly no responsiveness to AVP was observed in the proximal convoluted tubule, in the thin descending limb of the loop and in the distal convoluted tubule (DCT). The limited response obtained in DCT (which is a structure generally considered as a target site for AVP) as well as the clearcut effect elicited by AVP in MAL (the functioning of which is not known to be controlled by ADH) were expected observations; their possible physiological implications will be discussed.

[Vasopressin-stimulated adenyl cyclase activity in the ascending portions of rabbit kidney nephron loops].

The adenylate-cyclase activity contained in the successive segments of the loop of Henle was measured using single tubular pieces isolated by microdissection. Vasopressin (10 (-6) M) had no significant effect on either the pars recta or the thin descending limb; whereas, it stimulated the enzyme contained in the thin ascending limb (8.9 fold as a mean value) and that contained in the medullary portion of the large ascending limb (14.7 fold).

Receptive field characteristics and plastic properties of visual cortical cells in kittens reared with or without visual experience.

The functional organization of visual cortical cells was studied in two groups of five week old kittens: one group normally reared, the other reared in total darkness from the third day after birth. The following results were obtained: 1. The cells of the normally reared kittens were similar to adult cells except for some aspects of immaturity. In contrast, the cells of the dark-reared kittens were totally non-specific. Their receptive field showed neither orientational nor directional properties. 2. The distribution of cells according to the ocular dominance was not different in either group and was similar to that previously described for "adult" cells. 3. A few hours of visual experience was sufficient to provide specific receptive field properties to the cortical cells of a dark-reared kitten. 4. Conditioning exposure with an oriented grating induced changes in orientational sensitivity in normally reared kittens but not in dark-reared kittens.
