A 56 year-old Japanese woman died suddenly after complaining of abdominal pain. Autopsy revealed typical aortic dissection with cardiac tamponade. Histologically, generalized granulomatosis with giant cells resembling sarcoidosis was found. The aorta was most strikingly affected so that granulomatous aortitis was regarded as the cause of the aortic dissection.
The structure of precursors of dog and rabbit atrial natriuretic polypeptides was determined by nucleotide sequence analysis of cloned cDNA of mRNA encoding the peptides. The dog and rabbit precursors are 149 and 153 residues long having 23- and 25-residue putative signal peptides at their N-termini respectively. The 28-residue peptide with identical sequence to that of human, which has potent natriuretic activity, is located at the C-terminus of the dog precursor. The 28-residue peptide of identical sequence to that of mouse/rat is located at C-terminus of rabbit precursor followed by additional Arg-Arg sequence which is also found in rat/mouse precursors and is apparently removed during processing.
The dopaminergic inhibition of the hydrosmotic effect of vasopressin was studied by in vitro perfusion of the cortical collecting tubule isolated from the rabbit kidney. Arginine vasopressin (AVP) 100 microU/ml increased hydrosmotic water permeability (Pf, 10(-3) cm/s) by 11.4 +/- 1.59. Although dopamine 10(-5) M added alone to the bath did not affect Pf, the combined administration of 10(-5) M dopamine and 100 microU/ml AVP reduced the hydrosmotic effect of AVP to to 2.37 +/- 0.34. This inhibitory effect of dopamine was reversed by the simultaneous addition of 10(-5) M metoclopramide, a D2-antagonist. These observations suggest that dopaminergic receptors also exist in the cortical collecting tubule. The dopaminergic inhibition of the hydrosmotic action of AVP may be at least in part responsible for the diuretic action of dopamine.
Two monoclonal antibodies (nos. 6008 and 6016) were raised against human gamma interferon (IFN-gamma) derived from E. coli harboring the recombinant cDNA for IFN-gamma, and one (3710) against a synthetic peptide representing its C-terminus amino acid sequence of 20 residues. The monoclonal antibody against the synthetic peptide (3710) reacted either with IFN-gamma or the synthetic peptide. One monoclonal anti-IFN-gamma (6008) did not react with the synthetic peptide, while the other (6016) showed a weak binding with the peptide. The binding of the monoclonal antibody against the synthetic peptide (3710) with IFN-gamma was not inhibited by 6008, but to a certain extent by 6016. A 2-site '1-step' radioimmunoassay was developed in which 6008 was fixed on a solid-phase support, and the test sample together with radiolabeled 3710 was added for the binding with it. The assay was rapid with a sensitivity capable of detecting a few ng/ml of IFN-gamma.
Antibody to hepatitis B core antigen of immunoglobulin A class was determined in the serum of patients infected with hepatitis B virus by a sandwich-type solid-phase radioimmunoassay with monoclonal antibodies. The antibody, as defined by a sample to normal ratio greater than 2.1, was detected in all of 39 patients with acute hepatitis, with titers varying widely depending on the time of blood sampling. In persons with persistent infection, the antibody was detected in only 2 (4%) of 46 asymptomatic carriers of the virus, contrasting with the positivity in as many as 15 (41%) of 37 patients with chronic persistent hepatitis, in 45 (94%) of 48 patients with chronic active hepatitis, and in 40 (87%) of 46 patients with liver cirrhosis with or without hepatocellular carcinoma. The mean +/- SE titer of antibody in chronic persistent hepatitis (3.8 +/- 0.9) was significantly lower than those in chronic active hepatitis (13.8 +/- 3.2) and cirrhosis with or without carcinoma (25.6 +/- 6.1) (p less than 0.001). Based on the results obtained, the antibody may reflect hepatic injury in the persistent hepatitis B virus infection.
An expression plasmid, pHY202, was constructed which directs the synthesis of a fusion protein encoded by the gag sequence of human T-cell leukemia virus type I (HTLV-I) inserted into the lacZ' gene. Escherichia coli cells harboring pHY202 produced the 43-kDal LacZ'-Gag fusion protein with a yield of approx. 0.3% of total soluble proteins. The fusion protein is specifically recognized by monoclonal antibodies against the Gag proteins p19 and p24, and could be applicable for the diagnosis of HTLV-I infection, because almost all sera from HTLV-I carriers gave a positive response in the enzyme-linked immunosorbent assay (ELISA) employing the LacZ'-Gag hybrid protein purified by immunoaffinity column chromatography.

Mycoplasma pulmonis m53 was inoculated intraarticularly in the bilateral hind footpads and bilateral knee joints of BALB/c mice. Mycoplasmas were recovered from the affected joints over 20 weeks accompanying acute or subacute inflammation. Intensive deposition of immunoglobulins, a complement (C3) and mycoplasma cell antigens occurred in synovial and adjacent connective tissues. The histopathologically intact kidneys, brain, and lungs showed deposition of IgG and the complement on the endothelial cells of blood vessels. An IgG-rheumatoid factor like substance (RFLS) was detected in the serum of the mice by an enzyme-linked immunoabsorbent assay. Persistence of mycoplasma cells and immune complexes in the articular tissues might cause the prolongation of inflammatory responses in murine mycoplasmal arthritis.
The diagnostic potential of cloned cDNA copies of human rotavirus (strain WA) genome segments for the detection of rotavirus in clinical specimens has been determined. A hybridization assay in which a mixture of 32P-labeled cDNAs representing the 11 rotavirus segments was used as a probe compared favorably with three frequently used diagnostic tests for rotavirus in terms of both specificity and sensitivity. Significantly, clinical isolates could be readily distinguished when cloned cDNA copies of individual genome segments were used independently as a probe. In assays in which genome RNA from rotaviruses of known subgroups and serotypes were tested, cloned probes that encode nonstructural viral proteins hybridized efficiently to genome RNAs of all strains, whereas cloned probes corresponding to genome segments 6 and 9 exhibited the potential for differentiating strains of different subgroups and serotypes. Cloned cDNA copies of rotavirus genome segments therefore offer considerable potential for improved general diagnosis of rotavirus in clinical specimens, as well as for epidemiological studies in which virus isolates can be distinguished on the basis of nucleotide sequence homology of individual genome segments.
In order to confirm whether dopamine inhibits the antidiuretic action of vasopressin in mammalian kidney, we examined interactions among arginine vasopressin (AVP), dopamine and haloperidol in water-loaded ethanol anesthetized rats. The submaximal dose of AVP causing antidiuresis was 80 microU in this preparation. Dopamine at the doses of 0.11, 1.1 and 11 micrograms/100 g body weight (i.v.) inhibited the antidiuretic effect of 80 microU AVP by 18 +/- 7, 27 +/- 6 and 36 +/- 14%, respectively. The effect of 1.1 micrograms/100 g body weight dopamine in inhibiting the action of AVP was completely reversed by haloperidol at 2.3 micrograms/100 g body weight. Single administration of dopamine or haloperidol was without effect on urine flow. These observations support the view that dopamine inhibits the antidiuretic action of vasopressin by dopaminergic receptors also in the mammalian kidney.

The erythrocytes from the vitamin E-deficient rats showed no changes in comparison with the erythrocytes from the vitamin E-supplemented rats in lipid peroxides, membrane fluidity, lipid composition, and activities of defensive enzymes against the lipid peroxide formation. Once erythrocytes from vitamin E-deficient rats were exposed to H2O2, the erythrocyte membrane fluidity was significantly reduced, which was presumed to be accompanied by an increase in the hemolysis and lipid peroxide formation. Large decreases in arachidonic acid and phospholipid were also observed. These findings suggest that the reduced membrane fluidity is due to the loss of phospholipid from the erythrocytes, and raises the possibility that the reduced fluidity might contribute to the peroxidative hemolysis of the vitamin E-deficient rat erythrocytes.
The receptor for polymerized human as well as chimpanzee serum albumins has been identified on the 55-amino acid polypeptide coded by the pre-S region of hepatitis B virus DNA. Monoclonal antibodies were raised against a synthetic polypeptide of 19 amino acid residues representing a hydrophilic region of the pre-S amino acid sequence deduced from hepatitis B virus DNA. Sheep erythrocytes fixed with glutaraldehyde were coated with monoclonal antibody against the synthetic polypeptide to develop a hemagglutination assay for pre-S polypeptide. The pre-S polypeptide was detected in the serum containing hepatitis B surface antigen particles along with hepatitis B e antigen, with titers in parallel with those of the receptor for polymerized human serum albumin.
A case of glioblastoma multiforme of spinal cord followed by intracranial dissemination was reported. A 20-year-old man was admitted on Sep. 22, 1981, with motor disturbance of both legs and loss of urinary control. Neurological examination showed spastic paraplegia with bilateral Babinski signs. Abdominal and cremaster reflexes were absent. There was no abnormalities in cranial nerves and upper limbs. Cerebral CT was normal at that time. Myelography showed complete block at the level of 8th thoracic vertebra. Spinal angiography revealed abnormal vascularity at the level of Th7 fed via anterior spinal artery. Intramedullary tumor was partially removed through 5th-8th thoracic laminectomy. Total 5600 rads of irradiation was administered after the operation. The postoperative course was deteriorating. Two months after the operation paresis of both upper extremities had been developed. State of consciousness had been disturbed due to increased intracranial pressure which was subsided for a time while by ventriculoperitoneal shunt. Repeated CT showed high density spots scattered in basal cisterns spreading to all ventricular systems. He expired Feb. 21, 1982. Autopsy revealed that the whole spinal cord was covered by tumor tissue. Basal cisterns were filled with tumor tissue. The tumor was infiltrated subependymally into ventricular systems with intramedullary invasions in some places. Histological diagnosis was glioblastoma multiforme.
