Antagonistic actions of S-145 on vascular and platelet thromboxane A2 receptors.

We studied the actions of a potent thromboxane A2/prostaglandin in H2 (TP) receptor antagonist, (+/-)-(5Z)-7-[3-endo-[(phenylsulfonyl)amino]bicyclo [2.2.1]hept-2-exo-yl]heptenoic acid (S-145) on vascular and platelet receptors in the pig. S-145 showed almost the same affinity for both receptors in ligand binding studies with [3H]U46619 or [3H]SQ29, 548. The binding affinity of S-145 was 5-8 times higher than that of SQ29,548, a well-characterized TP receptor antagonist. However, S-145 inhibited U46619-induced contractions of pig coronary arteries with an IC50 value 52.5 times lower than that of SQ29,548, and was approximately equipotent with SQ29,548 in inhibiting U46619-induced secondary aggregation of pig platelets. Detailed kinetic studies on [3H]S-145 binding revealed that the apparent discrepancy between the pharmacological potency of S-145 in platelet and vascular systems was not due to tissue selectivity, but its small association constants for both receptors.

A fluorescence sandwich ELISA for detecting soluble and cell-associated human interleukin-2.

A fluorescence sandwich ELISA was developed for the quantitation of human interleukin-2 (IL-2) using a combination of two anti-human IL-2 antibodies, L61 and biotinylated anti-IL-2 rabbit IgG. The minimum level of IL-2 detectable by this ELISA was 0.3 pg/ml, indicating that this method is about 100 times more sensitive than the usual colorimetric human IL-2 ELISA. The ELISA specifically detected human IL-2 in cell culture supernatants and serum, and also in extracts from mitogen-stimulated peripheral blood mononuclear cells. All of the IL-2 species detected in the supernatants, sera and cell lysates were shown to have a molecular weight of 15,000, as estimated by size exclusion high performance liquid chromatography.

Comparison of vascular and platelet thromboxane A2/prostaglandin H2 receptors in the pig.

We compared the properties of vascular and platelet thromboxane A2/prostaglandin H2 receptors in the pig. The binding profiles of U46619, several prostaglandins and thromboxane A2/prostaglandin H2 receptor antagonists to the aorta and platelet receptors were almost the same irrespective of whether the agonist ([3H]U46619) or the antagonist ([3H]SQ29,548) radioligand was used, except that the receptor density in the latter was 4 times higher than that in the aorta. The antagonists suppressed U46619-induced contraction of pig coronary artery and secondary aggregation of platelets at potencies comparable to their KI values in the binding experiments. On the other hand, the responses of the artery specimens to U46619 and the prostaglandins differed from those of the platelets. Thus, the binding sites in the vascular and platelet receptors seem to be the same or quite similar, but there seem to be different mechanism(s) leading the agonistic binding signal to final responses.

A case of the pathogenesis of choledochocele in relation to dysfunction of the sphincter of Oddi.

A 47-year-old woman was admitted for evaluation of pain in the right upper quadrant of the abdomen. Seventeen years previously, she had undergone cholecystectomy for cholelithiasis. Endoscopic retrograde cholangiopancreatography (ERCP) showed a cystic dilatation of the terminal portion of the common bile duct (CBD) protruding into the duodenal lumen and delaying the drainage of contrast medium. In this patient the CBD and the pancreatic duct appeared to be unimportant in the formation of the cyst. Repeated changes in the radius of the cyst suggested dysfunction of the ampullary component of the sphincter of Oddi, with maintenance of normal function of the common duct component. The pathogenesis of the choledochocele in this patient is discussed in relation to dysfunction of the sphincter of Oddi. In addition, 2 criteria for the diagnosis of choledochocele by ERCP are proposed: [1] cystic dilatation of the terminal portion of the CBD protruding into the duodenal lumen, and [2] absence of the narrow segment of the CBD.

Muscular dystrophy-like disease in a thoroughbred foal.

A 1-month-old male thoroughbred foal, which had difficulty in walking, was killed and examined by histological, histochemical and ultrastructural methods. The muscles of the trunk and upper hind limbs were chiefly affected, and changes in the affected muscles resembled those in muscular dystrophy in man. The type of muscular dystrophy present in this foal and the significance of this disease in thoroughbred horses are discussed. The dystrophy in this foal resembled the limb-girdle type or myotonic dystrophy of muscular dystrophy in man.

Pathogenesis of cholesteatoma based on clinical results of anterior tympanotomy.

A new conception of the pathogenesis of cholesteatoma is proposed based on clinical results of anterior tympanotomy, a procedure developed in order to prevent recurrence of cholesteatoma following retraction pocket formation after canal-up operation. In the cholesteatomatous ear, there is always a long, thick bony septum between the epitympanum and the supratubal recess. This septum is called the anterior attic bony plate, forming the tympanic diaphragm together with the tensor tympani mucosal fold. In anterior tympanotomy, this bony plate is removed finishing intact canal wall mastoidectomy, making a new ventilating route to the eustachian tube. With this technique, the rate of recurrence was reduced from 17 to 5%. Based on these results it is proposed that the real pathogenesis of cholesteatoma is not tubal dysfunction, but the osseous tympanic diaphragm.

[Respiratory tract infections caused by Branhamella catarrhalis in outpatients with pneumoconiosis].

To investigate the occurrence of Branhamella catarrhalis respiratory tract infections in 109 outpatients with pneumoconiosis, clinical and bacteriological studies were performed during a 4-year period from April 1984 to March 1988. B. catarrhalis was isolated in 26 patients; only three of these received continuous corticosteroid treatment. The incidence of B. catarrhalis respiratory tract infections increased gradually during the years 1984-1986, but decreased for the first time in 1987 compared with the previous year. There was a seasonal variation in isolations with a peak incidence during the winter, a pattern in contrast to Haemophilus influenzae. Almost all isolates produced beta-lactamase. B. catarrhalis found in mixed culture was usually in association with H. influenzae or Streptococcus pneumoniae. The isolation rates for B. catarrhalis in sputum of patients with pneumoconiosis followed those of H. influenzae and S. pneumoniae, and almost all strains were positive for beta-lactamase, so B. catarrhalis should be admitted that it is a primary pathogen.

Effect of cold pressor test on carbon monoxide diffusing capacity in normal subjects.

We investigated changes in the pulmonary carbon monoxide diffusing capacity (DLco) during the cold pressor test (CPT) on 25 normal subjects. In 10 of them we also observed changes in circulatory parameters by a computerized dual cadmium telluride detector system, using an equilibrium radionuclide blood-pool label. DLco and DLco per unit of alveolar volume (DLco/VA) averaged in the control period were 29.4 +/- 4.1 ml/min/mm Hg, 6.1 +/- 0.8 ml/min/mm Hg/l (mean +/- SD). During the 2nd minute of CPT, DLco increased by 3.6 +/- 1.5% and DLco/VA by 5.1 +/- 1.5% (mean +/- SE). The systemic blood pressure increased by 17% (mean increase) whereas the heart rate and the stroke volume remained unchanged. The increases were small but significant (p less than 0.05, p less than 0.01, respectively). We conclude that the increase in DLco is due to cold-induced systemic vasoconstriction followed by a passive shift of blood into the pulmonary vasculature.

Hypothalamic neurons from a developmental aspect.

The development of hypothalamic neurons was examined in vivo and in transplanted grafts in rats. The neurons appeared in vivo in distinctive chronotopical schedules showing such morphological characteristics as synaptocrine or hemocrine neurons. These phenotypical properties seemed to be primed already by day 12.5 of gestation in rats, because the grafted hypothalamic primordia from 12.5-day-old embryonal rats differentiated neurons, which express these neuronal properties in the third ventricle of adult female rats. The synaptocrine neurons projected to other neurons, suggesting the establishment of synaptic contacts, and the hemocrine neurons projected to vasculatures developed in the grafts, suggesting the accomplishment of neurovascular associations.

[A histological observation of the primary teeth with enamel hypoplasia resulting from perinatal disturbance].

Two primary teeth with enamel hypoplasia from two cerebralpalsied children were subjected to histological examination. One primary tooth was B and the other was C. The location of enamel hypoplasia in these teeth was considered to coincide with the neonatal line reaching the enamel surface. The purpose of this study was to examine the location of the enamel hypoplasia in these teeth in relation to the neonatal line, and to detail the histological structure of the teeth. They were examined morphologically as well as histologically, by using a stereoscopic microscope, SEM, a light microscope and a microradiogram. The results obtained were as follows: 1. The size of the tooth crown of these teeth was smaller than the normal standard, but the form of tooth crown was normal. 2. Based on streomicroscopic observation, enamel hypoplasia was found on the labial and lingual surfaces in B, and on labial surface only in C. In B, the enamel hypoplasia on labial surface was extended widely to incisal 2/3 of the crown, while that on the lingual surface was minimum. In C, there were two spots of enamel hypoplasia located on the mesial and distal incisal edges respectively. 3. Based on SEM observation, the hypoplastic enamel was formed right after their birth. 5. Based on microradiographic observation, the neonatal lines were radiolucent, and the adjacent areas of the neonatal line did not show any poor calcification. 6. From these histological observations, it was found that the perinatal disturbance directly affected the tooth enamel formation and these enamel hypoplasias were positively caused by the cerebral palsy of these children.

Monoclonal antibodies which differentiate high- and low-affinity binding sites of interleukin-2.

Five monoclonal antibodies (L15, L20, L23, L34, and L61) against human recombinant interleukin-2 were tested for their effects on the interleukin-2 bioactivity and binding. Four of these monoclonal antibodies, L15, L20, L34, and L61, which had neutralizing activity, completely blocked interleukin-2 binding to the high-affinity receptor. On the other hand, L23, which had a very weak neutralizing activity, blocked interleukin-2 binding to the low-affinity receptor. These results suggest that there are at least two distinct binding sites on the interleukin-2 molecule; those for the high-affinity receptor and those for the low-affinity receptor. These monoclonal antibodies should be useful tools in the study of the interaction between interleukin-2 and interleukin-2 receptor.

Characterization of [3H]U46619 binding in pig aorta smooth muscle membranes.

The binding characteristics of [3H]U46619, a tritiated thromboxane A2/prostaglandin H2 mimetic, were studied in pig aorta smooth muscle membranes. Binding was fast, saturable, selective and reversible. The KD values determined by kinetics, equilibrium binding and drug competition methods were 68, 42 and 53 nM, respectively. The Bmax was 87.8 fmol/mg protein. Specific binding was competitively displaced by thromboxane A2/prostaglandin H2 antagonists. Binding was also displaced by prostaglandins D2, E2 and F2 alpha with IC50 values of 8, 21 and 12 microM, respectively. U46619 contracted the rat and pig aorta smooth muscle, but the response of the latter was slower than that of the former. The antagonists prevented the U46619-induced contraction of rat aorta with the same rank order as that for the inhibition of ligand binding in pig aorta smooth muscle membranes. These results provide evidence that the putative thromboxane A2/prostaglandin H2 receptor in vascular smooth muscle membranes can be detected by a ligand binding technique.

Phase plane (volume to volume-time function loop) display of data from radionuclide ventriculography obtained by single cardiac probe system.

To assess the left ventricular (LV) performance more sensitively, a new display method of phase plane (PP), displaying volume and volume-time function (dV/dt) in a single image, was applied to radionuclide ventriculography obtained by a single cardiac probe system. The sampling interval was 10 msec and the data acquisition time was 60 sec. The LV volume curve was smoothed by fitting a fourth order polynomial curve of Fourier's analysis. Then the dV/dt was calculated. In this single image PP display, the width of the horizontal axis indicates relative LV volume, and the height of the vertical axis indicates dV/dt. The direction of the rotation of this loop is clockwise. We classified 126 patients with various heart diseases into seven groups, according to the configuration of the loop. The most interesting finding was that the distortion of the loop during diastole was frequently seen in patients with hypertension and angina pectoris, whereas their ejection fraction was within normal limits. We concluded that the single image PP display is a sensitive method for assessing the abnormality of the LV function, not only by evaluating the conventional parameters, but also by analyzing the configuration of the volume to volume-time function loop.

The significance of stapedial reflex test in facial nerve paralysis.

The stapedial reflex of 30 patients with peripheral facial nerve paralysis was compared with the results of facial paralysis scores. The stapedial reflex was measured with an oto-reflex analyser, RS-06A. The stimulation frequencies in the stapedial reflex test were 500, 1,000 and 2,000 Hz for both ipsilateral and contralateral stimulation. The stapedial reflex test at 500 Hz for contralateral stimulation seemed to be useful for evaluating the degree of facial paralysis. With 500 Hz or 1,000 Hz contralateral stimulation, the stapedial reflex test seemed to be useful for predicting the prognosis for facial nerve paralysis. In these stimulations, cases with a positive reflex within 2 weeks showed complete recovery within 12 weeks, and cases with positive reflex within 4 weeks showed recovery within 24 weeks.

Teratogenic interaction of ethanol and hyperthermia in mice.

Alcohol and maternal hyperthermia have been implicated in human birth defects. Both ethanol and heat can induce neural tube defects (NTDs) and other developmental abnormalities in mice when large doses are given during pregnancy. To explore the teratogenic interaction of both agents, pregnant ICR mice were injected with a single dose of 25% ethanol and/or were heat-stressed in a water bath at 42 degrees C on the morning of Day 8 of gestation. Combined treatment with ethanol (0.01-0.02 ml/g) and heat (10 min), when they were given concurrently or 1 hr apart, resulted in a significant increase of resorptions and externally malformed fetuses. Skeletal malformations and visceral variations also increased significantly following a concurrent exposure to both agents. These results indicate that ethanol and heat can be synergistically teratogenic in mice when the doses of each agent are below the teratogenic threshold. It was also suggested that pretreatment with a small dose of ethanol may not enhance the teratogenicity of heat when the hyperthermic stress is strong enough and teratogenic by itself.
