Genomic expression responses to DNA-damaging agents and the regulatory role of the yeast ATR homolog Mec1p.

Eukaryotic cells respond to DNA damage by arresting the cell cycle and modulating gene expression to ensure efficient DNA repair. The human ATR kinase and its homolog in yeast, MEC1, play central roles in transducing the damage signal. To characterize the role of the Mec1 pathway in modulating the cellular response to DNA damage, we used DNA microarrays to observe genomic expression in Saccharomyces cerevisiae responding to two different DNA-damaging agents. We compared the genome-wide expression patterns of wild-type cells and mutants defective in Mec1 signaling, including mec1, dun1, and crt1 mutants, under normal growth conditions and in response to the methylating-agent methylmethane sulfonate (MMS) and ionizing radiation. Here, we present a comparative analysis of wild-type and mutant cells responding to these DNA-damaging agents, and identify specific features of the gene expression responses that are dependent on the Mec1 pathway. Among the hundreds of genes whose expression was affected by Mec1p, one set of genes appears to represent an MEC1-dependent expression signature of DNA damage. Other aspects of the genomic responses were independent of Mec1p, and likely independent of DNA damage, suggesting the pleiotropic effects of MMS and ionizing radiation. Intracellular localization and movement phenotypes of alfalfa mosaic virus movement protein mutants.

Thirteen mutations were introduced in the movement protein (MP) gene of Alfalfa mosaic virus (AMV) fused to the green fluorescent protein (GFP) gene and the mutant MP-GFP fusions were expressed transiently in tobacco protoplasts, tobacco suspension cells, and epidermal cells of tobacco leaves. In addition, the mutations were introduced in the MP gene of AMV RNA 3 and the mutant RNAs were used to infect tobacco plants. Ten mutants were affected in one or more of the following functions of MP: the formation of tubular structures on the surface of protoplasts, association with the endoplasmic reticulum (ER) of suspension cells and epidermal cells, targeting to punctate structures in the cell wall of epidermis cells, movement from transfected cells to adjacent cells in epidermis tissue, cell-to-cell movement, or long-distance movement in plants. The mutations point to functional domains of the MP and support the proposed order of events in AMV transport. Studies with several inhibitors indicate that actin or microtubule components of the cytoskeleton are not involved in tubule formation by AMV MP. Evidence was obtained that tubular structures on the surface of transfected protoplasts contain ER- or plasmalemma-derived material.

MLL and CREB bind cooperatively to the nuclear coactivator CREB-binding protein.

A fragment of the mixed-lineage leukemia (MLL) gene (Mll, HRX, ALL-1) was identified in a yeast genetic screen designed to isolate proteins that interact with the CREB-CREB-binding protein (CBP) complex. When tested for binding to CREB or CBP individually, this MLL fragment interacted directly with CBP, but not with CREB. In vitro binding experiments refined the minimal region of interaction to amino acids 2829 to 2883 of MLL, a potent transcriptional activation domain, and amino acids 581 to 687 of CBP (the CREB-binding or KIX domain). The transactivation activity of MLL was dependent on CBP, as either adenovirus E1A expression, which inhibits CBP activity, or alteration of MLL residues important for CBP interaction proved effective at inhibiting MLL-mediated transactivation. Single amino acid substitutions within the MLL activation domain revealed that five hydrophobic residues, potentially forming a hydrophobic face of an amphipathic helix, were critical for the interaction of MLL with CBP. Using purified components, we found that the MLL activation domain facilitated the binding of CBP to phosphorylated CREB. In contrast with paradigms in which factors compete for limiting quantities of CBP, these results reveal that two distinct transcription factor activation domains can cooperatively target the same motif on CBP.

Characterization of a calcium-activated chloride channel as a shared target of Th2 cytokine pathways and its potential involvement in asthma.

Interleukin (IL)-9 is a T helper (Th) 2 cytokine recently implicated as an essential factor in determining susceptibility to asthma. Transgenic mice overexpressing IL-9 exhibit many features that are characteristic of human asthma. To better understand the mechanism by which IL-9 mediates the various biologic activities in asthma, we performed suppressive subtraction hybridization with whole lung from IL-9 transgenic and control mice. Here we report the identification of mCLCA3, a calcium-activated chloride channel that was specifically induced in the lung epithelium of IL-9 transgenic mice. Expression of mCLCA3 could also be induced by intratracheal administration of IL-9 or other Th2 cytokines (IL-4, IL-13), but not by interferon-gamma. Moreover, expression of mCLCA3 was induced in the lung of antigen-exposed mice, and this induction could be suppressed by neutralizing IL-9 antibody treatment, indicating IL-9 is both necessary and sufficient to induce mCLCA3 in this experimental model of asthma. Finally, we demonstrate that hCLCA1 is the human counterpart to mCLCA3 and is also induced in vitro in human primary lung cells by Th2 cytokine treatment. Together, these data strongly implicate the involvement of mCLCA3 (in mice) and hCLCA1 (in humans) in the pathogenesis of Th2 cytokine-mediated asthmatic disorders.

Natural course of schizophrenia: 2-year follow-up study in a rural Chinese community.

BACKGROUND: A number of studies have questioned whether the natural course of schizophrenia is more favourable in "developing" than "developed" societies and whether culture is a factor in producing a favourable course. AIMS: This prospective study tests the hypothesis that the natural outcome of schizophrenia would be favourable in a Chinese rural area. METHOD: We investigated all patients with schizophrenia, including patients who had not received any treatment, among 149 231 rural community population in Xinjin County, Sichuan in 1994. Those patients who had never received treatment were followed up for two years. RESULTS: Three-quarters of patients with schizophrenia who had not been treated remained symptomatic. As the duration of illness increased, the illness became more serious. The clinical outcome of the drug-treatment group was significantly better than for patients who had not received any treatment. CONCLUSIONS: The natural clinical outcome of schizophrenia in the Chinese rural community was poor, and occupational functioning of patients with schizophrenia was comparatively better. Schizophrenia itself has a specific natural course--antipsychotic drug treatment and psychosocial treatment will produce an improvement in prognosis.

Matrix effect on the analysis of oligonucleotides by using a mass spectrometer with a sonic spray ionization source.

Matrix or impurities remaining in a DNA sample solution after various sample treatment procedures may influence a subsequent DNA analysis. In this work, several matrices were investigated concerning their effects on the analysis of oligonucleotide by using an ion-trap mass spectrometer equipped with a sonic spray ionization source. Inorganic salts of sodium chloride and magnesium chloride depressed the signal intensity by about 50% when the content of the salts was about 10 microM. dNTPs and Taq showed more severe depression on the oligonucleotide. However, Tris, or (hydroxymethyl)aminomethane, intensified the signal intensity, if its content was within an appropriate range. When the content of Tris was about 500 microM, the signal intensity was enhanced by factors of 3 and 5 for the 6-mer and the 20-mer oligonucleotides, respectively. With the existence of Tris, matrix effects from the inorganic salts, dNTPs and Taq were reduced.

[Effects of tumor necrosis factor alpha on the expression of connective tissue growth factor in hepatic stellate cells].

OBJECTIVE: To investigate the effect of tumor necrosis factor alpha (TNF-alpha) on the expression of connective tissue growth factor (CTGF) in rat hepatic stellate cells (HSC) in vitro. METHODS: HSCs were prepared from SD rats by in situ perfusion and single-step Nycodenz gradient, and were treated with TNF-alpha and TGF-beta 1. The expression of CTGF in HSC was assessed using reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: Both TNF-alpha and TGF-beta 1 could stimulate the expression of CTGF mRNA in HSC. The expression of CTGF was not found in HSC after treatment with TNF-alpha(10microgram/L) for 6 hours, but was observed after TNF-alpha treatment for 24 and 48 hours. TGF-beta1 could induce the mRNA expression for CTGF just after 6 hours of treatment at 1microgram/L concentration. CONCLUSIONS: TNF-alpha is able to induce the expression of CTGF in HSC, and this may be one of the mechanisms for TNF-alpha involving in the liver fibrosis.

Preliminary interspecific genetic maps of the populus genome constructed from RAPD markers.

We have constructed RAPD-based linkage maps for an interspecific cross between two species of the genus Populus (P. adenopoda and P. alba), based on a double pseudo-test-cross strategy. Of a total of 360 polymorphic fragments scored, 290 showed a test-cross configuration, corresponding to DNA polymorphisms heterozygous in one parent and null in the other. In the female parent, P. adenopoda, 82 markers were grouped in 19 different linkage groups (553 cM), whereas in the male parent P. alba, 197 markers established a much more complete framework map with an observed genome length of 2300 cM covering 87% of the total P. alba genome. The larger number of test-cross markers detected for the P. alba parent than for the P. adenopoda parent might be due to a higher level of heterozygosity in the former than in the latter. In this study, we detected only a small percentage (2%) of the intercross dominant markers heterozygous in both parents and segregating 3:1 in the progeny. The further focus in this mapping study should be on the identification of more intercross markers, to align the two parent-specific maps into a consensus map for mapping important genes causing species differentiation during long evolutionary divergences.

Inhibition of JAK kinase activity enhances Fas-mediated apoptosis but reduces cytotoxic activity of topoisomerase II inhibitors in U266 myeloma cells.

Our previous work demonstrated that the Janus kinase (JAK)-Stat3 pathway regulates expression of Bcl-x(L) in the U266 human multiple myeloma cell line and prevents Fas-mediated apoptosis. Inhibition of this pathway by the JAK selective kinase inhibitor AG490 or dominant-negative Stat3 protein results in down-regulation of Bcl-x(L) expression and enhanced sensitivity to Fas-mediated apoptosis. Because Bcl-x(L) has also been implicated in resistance to chemotherapeutic drugs, we investigated whether inhibition of the JAK-Stat3 pathway and subsequent reduction in Bcl-x(L) expression would also enhance cytotoxic drug activity. Contrary to this prediction, pretreatment of U266 myeloma cells with AG490, followed by exposure to topoisomerase II- inhibiting agents, antagonized drug-induced apoptosis. This effect correlated with reduced cyclin D1 expression and cell cycle arrest. The cell cycle arrest following AG490 pretreatment further correlated with reduced mitoxantrone-induced DNA double-strand breaks and reduced cell death, findings consistent with the critical requirement of DNA damage for drug cytotoxicity. These studies demonstrate that inhibition of the JAK-Stat3 pathway can result in paradoxical effects relative to cytotoxic drug response. These paradoxical responses may be explained by the findings that JAK-Stat3 signaling regulates the expression of multiple genes involved in controlling cell proliferation and apoptosis. Thus, understanding the cellular context of inhibiting signal transduction pathways is essential for the design of novel combination therapies for cancer.

[Electrochemical therapy and implanted ports treatment for unresectable carcinoma of body and tail of pancreas].

OBJECTIVE: To examine the efficacy and safety of electrochemical therapy (ECT) in treatment of patients with unresectable carcinoma of the body and tail of pancreas. METHODS: A total of 18 patients (14 men and 4 women; mean age 57.7) with unresectable carcinoma of the body and tail of pancreas were treated by ECT between May 1989 and June 1996 in our hospital (group A). Another 16 patients (12 men and 4 women; mean age 60.3) were treated by ECT plus intraperitoneal chemotherapy between July 1996 and June 1999 (group B). In group B, ECT was used during the operation. Meanwhile, 300 mg calcium leucovorin (CF), 500 mg 5-FU and 200 mg carboplatin were injected weekly into the abdominal cavity through the implanted ports. Pain was determined by using a 10-point visual analog pain scale (VAS) before and after the treatment, life quality periodically assessed according to the 5-step scale system, and tumor size measured using CT or MRI. RESULTS: All the patients were followed up for 5-30 months (mean = 10 months), showing that there were no operative deaths and such complications such as pancreatic fistula and bleeding in both groups. The tumor size in both groups was decreased to some degree. Ascites in 4 of 6 cases in group B were well controlled, whereas there was no changes in 6 cases of ascites in group A. In group A, the VAS basal score was reduced from 7.4 +/- 1.7 before to 2.66 +/- 1.05 after the treatment and the peak VAS from 9.3 +/- 0.9 to 3.71 +/- 0.96 (P < 0.05). In group B, they were decreased from 7.5 +/- 1.5 to 2.17 +/- 1.23 and 9.2 +/- 0.9 to 3.4 +/- 1.13, respectively (P < 0.01). Significant reduction of VAS score persisted for over 3 months after ECT. The satisfaction rate in group B was 37.5%, 62.5% and 25% in 7 days, 1 month and 3 months, respectively. CONCLUSIONS: ECT is a safe and effective procedure for treatment of unresectable carcinoma of the body and tail of pancreas and in relieving its pain and improving the life quality of patients. Supplementary injection of chemical drugs through drug delivery pump is helpful for control of ascites.

[Prevalence of dementia in Shanghai urban and rural area].

OBJECTIVE: To study the prevalence and epidemiological characteristics of dementia and their major types in people aged 55 years and old in urban and rural area, Shanghai. METHODS: A sample with 17,018 residents aged 55 years and above from 22 urban and 14 rural communities was drawn through a stratified multiple stage cluster sampling. A door-to-door three-phase procedure was used to investigate the prevalence of dementia. In 1997 and 1998, A brief memory testing by MMSE was administered to all participants. In phase 2, those who scored below the cutoff points and four percent of those whose score was in the normal range were interviewed to identify dementia through a set of diagnostic examinations according to the DSM-IV criteria. In phase 3, a follow-up program was conducted in 1998 for all persons who was involved in phase 2 to identify those with Alzheimer's dementia (AD) and vascular dementia (VaD) by NINCDS-ADRDA and NINDS-AIREN criteria. RESULTS: Of the targeted sample, 15,910 (94.2%) were interviewed. The overall prevalence of dementia was 3.0% in individuals aged 55 years and above, 4.32% in those aged 65 years old and 5.29% in those aged 70 years and above. The age-adjusted prevalence rates of dementia were 2.03%, 3.78% and 5.48% respectively among the age groups mentioned above. For AD the age-adjusted prevalence rates were 1.37%, 2.71% and 4.13% for individuals in above age groups. Of all types of dementia, AD was accounted 72%, followed by VaD the second. The prevalence of AD was 1.41% in man comparing to 2.78% in woman. The age-specific prevalence of AD increased with age. CONCLUSION: The prevalence of dementia was 3.0% in individuals aged 55 years in urban and rural areas in Shanghai. Alzheimer's disease was more frequent than VaD.

Percutaneous balloon aortic valvuloplasty in the treatment of congenital valvular aortic stenosis in children.

OBJECTIVE: To assess the effect of the balloon valvuloplasty for for congenital valvular aortic stenosis (AS) in children. METHODS: A total of 27 (mean age 6.09 years) children with AS accepted the treatment of percutaneous balloon aortic valvuloplasty (PBAV). The ratios of balloon/valve were 0.95 +/- 0.08 for 19 cases of typical AS and 1.00 +/- 0.11 for 8 cases of hypoplastic AS. The patients were evaluated by the gradients across aortic valves in pre- and post-PBAV and by echocardiogram during the follow-up period. RESULTS: Fifteen of 19 (78.9%) cases of typical AS had a better outcome and the gradient of the remaining 4 cases (26.7%) had increased after follow-up (delta P > 50 mm Hg). Four of 8 (50.0%) cases of hypoplastic AS had satisfactory responses and the gradient of the remaining 3 cases (75.0%) rose. There was no moderate to severe aortic insufficiency (AI). CONCLUSION: The balloon aortic valvuloplasty provides safe and significant hemodynamic and clinical improvement in pediatric patients. The outcome of PBAV for typical AS is better than for hypoplastic AS.

Percutaneous balloon angioplasty of coarctation of the aorta in children: 12-year follow-up results.

OBJECTIVE: To evaluate the efficiency of percutaneous balloon angioplasty of coarctation of the aorta in children and discuss its risk factors that can lead to poor long-term results. METHODS: From September 1987 to August 1999, 24 patients underwent 27 balloon angioplasty procedures for native or recurrent coarctation of the aorta at our institution. There were 19 patients with discrete (membranous) coarctation, 4 patients with long-segment and aortic arch hypoplasia, and 1 patient with postoperative recurrence. The balloon diameter was chosen not to exceed the diameter of the aorta proximal to the stenotic site (11.06 +/- 2.80 mm vs 11.78 +/- 3.18 mm), with the chosen diameter two to four times that of the stenotic segment (2.57 +/- 0.68 times). The patients have been followed up for half to twelve years (mean 6.2 +/- 2.8 years). Satisfactory result was defined as a reduction in the pressure gradient across the site of coarctation to < or = 20 mm Hg. RESULTS: The systolic pressure gradient across the coarctation site decreased from 48.17 +/- 14.68 mm Hg to 14.96 +/- 13.12 mm Hg (P < 0.01) and the diameter of the coarctation site increased from 4.66 +/- 2.43 mm to 8.80 +/- 3.32 mm (P < 0.01). Immediate satisfactory results were obtained in 19 patients (79%). Of the 5 patients with unsatisfactory results, 4 had aortic arch hypoplasia and 1 had membranous coarctation. Of the 19 patients with satisfactory results, 18 patients had membranous coarctation and 1 patient had recurrent postoperative coarctation. No aneurysm and other complications had occurred in any patients at the follow-ups. CONCLUSION: Percutaneous balloon angioplasty is an effective treatment alternative to surgery in most patients with recurrent postoperative or native membranous coarctation of the aorta. It should not be recommended for the coarctation of aortic arch hypoplasia.

[Changes of the expression of integrin alpha6beta1 and the activity of focal adhesion kinase in the liver sinusoidal endothelial cell in experimental fibrotic rats].

OBJECTIVE: To investigate the expression of integrin alpha(6)beta(1) and the activity of focal adhesion kinase (FAK) in liver sinusoidal endothelial cells (SECs) from experimental fibrotic rats induced by CCl(4). METHODS: By in situ collagenase perfusion and two-step Percoll gradient centrifugation, SECs were isolated and cultured from normal and CCl(4) -treated Wistar rats. The expression of integrin alpha(6)beta(1) was determined by cell-ELISA, and the activity of FAK was assessed by immunoprecipitation-tyrosine kinase assay. RESULTS: The integrin alpha(6)beta(1) was almost absent in the normal SECs and was up-regulated during the fibrotic process; SECs from experimental fibrotic rats possessed higher expression level of integrin alpha(6)beta(1) than normal SECs (P<0.05). The FAK activity in SECs from experimental fibrotic rats increased significantly as compared with the normal controls (P<0.05). CONCLUSIONS: The expression of integrin alpha(6)beta(1) on SECs and the increase of FAK in SECs may be important in the phenotype and function changes of SECs during hepatic fibrogenesis.

[Dynamic analysis of incidence and mortality of stroke and the risk factors in the communities in Shanghai, in the 1990's].

OBJECTIVE: To understand the trend of incidence, mortality of stroke and the risk factors at the community level in the 1990's. METHODS: To establish a network of disease surveillance. Both incidence and mortality of stroke were surveyed in the 35 - 74 age group during 1991 - 1999 using the method of WHO MONICA. By cluster sampling in 1992 and 1998, 500 individuals aged 35 - 59 from each female and male group were investigated respectively on the risk factor of stroke. RESULTS: The incidence of stroke was averaged 168.2 per 100 000 in the group aged 35 - 79 during 1991 to 1999 with 194.5 and 144.9 pre 100 000 respectively for the male and female group of this age. The incidence of stroke in the male group was higher than that in the female group, also with a faster speed of increase in the male group. The overall mortality of stroke was averaged 79.2 per 100 000 with 87.2 and 71.4 per 100 000 in male and female groups respectively of this age group. The incidence of stroke increased with age in both male and female groups. The average age of onset had been 66.1 year olds during 1996 - 1999, 3.5 years older than that age groups during 1991 - 1995, but did not show much difference between male group and female group. CHO, Blood Pressure, BMI, Prevalence of Hypertension, TC/HDLC, WHR, Prevalence of High-CHO, Prevalence of Low-HDL all had a tendency of increase in both male and female groups. The rates of smoking cigarettes and drinking alcohol both decreased, especially in the male group. CONCLUSION: The incidence of stroke and some relative risk factors seemed to increase in the 1990's, but the mortality of stroke had a decrease in the same period in the communities in Shanghai.

[The role of YIGSR and RGD peptides in regulating fenestrae of sinusoidal endothelial cells].

OBJECTIVE: To investigate the effects of Tye-Ile-Gly-Ser-Arg (YIGSR) and Arg-Gly-Asp (RGD) polypeptides on the fenestrae of the liver sinusoidal endothelial cells (SECs). METHODS: By in situ collagenase perfusion and two-step percoll gradient centrifugation, SECs were isolated from normal Wistar rats and cultured on collagen type I or laminin-coated coverslips. The population of SECs fenestrae and their diameters were observed using scanning electron microscopical technique; and the synthesis of collagen type IV in SECs was assessed by radio-immunoassay (RIA). RESULTS: The average number of fenestrae and their diameter in the SECs cultured on laminin-coated coverslips were significantly lower than those on collagen type I-coated coverslips. After the treatment with YIGSR pentapeptides (50 micrograms/ml) and RGD tripeptides (50 micrograms/ml) together for 48 hrs, the number and diameters of fenestrae in the SECs that grew on laminin-coated coverslips increased markedly. Meanwhile, the YIGSR and RGD peptides could significantly decrease the synthesis of collagen type IV in SECs that grew on laminin-coated coverslips. CONCLUSION: The results suggested that the YIGSR and RGD polypeptides may have an important role in regulating the morphology and function of SECs.

[The paraoxonase Gln-Arg192 polymorphism in patients with coronary heart disease in Chinese population].

OBJECTIVE: To investigate the 192 Gln-Arg polymorphism of paraoxonase (PON) gene and its relationship with serum lipids levels in patients with coronary heart disease in Chinese population. METHODS: The genotype and allele frequency of paraoxonase gene 192 Gln-Arg polymorphism was assayed by polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP). Serum lipids were measured by enzymatic kits in 118 CHD patients and 128 healthy subjects whose fasting serum TG levels were < 1.82 mmol/L and TC levels < 6.2 mmol/L from a population of Chinese Han nationality in Chengdu area. RESULTS: Both in CHD group and control group, the QR genotype of PON gene was the major one, and its frequencies were 0.578 and 0.525 respectively. No differences were found in PON gene Gln-Arg polymophism in the CHD group when compared with the control group. The frequency of R allele of PON gene in Chinese was significant higher than that in European and American Caucasians (0.51 vs. 0.26-0.31, P < 0.01) and Indian (0.51 vs. 0.17, P < 0.01), and significantly lower than that in Japanese (0.51 vs. 0.65, P < 0.01). CONCLUSION: These results suggested that the Gln-Arg192 polymorphism of the PON1 gene was not associated with coronary heart disease in Chinese population.

[Apolipoprotein C III gene Sst I polymorphism in patients with endogenous hypertriglyceridemia in Chinese population].

OBJECTIVE: To investigate the apoC III gene Sst I polymorphism and its relationship with serum lipids and apolipoproteins (apo) levels in patients with endogenous hypertriglyceridemia in Chinese population. METHODS: The genotype and allele frequency of apoC III gene Sst I polymorphism was assayed by polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP). Serum lipids were measured by enzymatic kits and apolipoproteins A I, A II, B100, C II, C III and E were measured by RID kits developed by Apolipoprotein Research Unit of WCUMS in 176 HTG patients whose fasting serum TG levels were > or = 2.26 mmol/L and in 199 healthy subjects whose fasting serum TG levels were < 1.82 mmol/L and TC levels < 6.2 mmol/L from a population of Chinese Han nationality in Chengdu area. RESULTS: In both HTG group and control group, S1 allele was the major allele and homozygous S1S1 genotype was the most frequent one. The frequency of S2 allele was significantly higher than that reported in Caucasians (0.289 vs 0.06-0.16, P < 0.05). No differences were found in apoC III gene Sst I polymophism in the HTG group when compared with the control group (0.287 vs 0.289, P > 0.05). The genotype of S2S2 was not associated with higher TG and apolipoproteins levels when compared with the genotypes of S1S1 and S1S2 (P > 0.05). CONCLUSION: These results suggest that the Sst I polymorphism of the apoC III gene was not associated with endogenoushypertriglyceridemica in Chinese population.
