[Experimental and clinical studies of an ampicillin suppository (KS-R1) in pediatrics].

Blood levels of ampicillin (ABPC) were measured in 10 childish patients with heart disease after the rectal administration of KS-R1 at doses of 125 mg and 250 mg. Average blood levels of ABPC at 15, 30 minutes, 1, 2 hours and 4 hours after the administration of KS-R1 were 6.8, 6.9, 3.1, 1.1 mcg/ml and 0.1 mcg/ml with half-life of 0.64 hours in patients of age from 1 year to 4 years 7 months old (dose level 8.9 approximately 13.9 mg/kg, average 10.5 mg/kg), and 5.2, 6.1, 3.4, 1.0 mcg/ml and 0.1 mcg/ml with half-life of 0.65 hours in patients of age from 7 years 10 months to 10 years 7 months old (dose level 8.3 approximately 13.9 mg/kg, average 9.8 mg/kg), respectively. Clinical effective rate (excellent and good) was 87% in 55 childish patients with infections. Bacteriologically, 13 strains (74%) out of 18 strains which were isolated from the patients were eradicated. No severe side effects were observed. Diarrhea was observed in 3 cases.

A novel Japanese Burkitt's lymphoma cell line, P32/Ishida, with a new variant chromosomal translocation (2;14).

A novel cultured cell line, P32/Ishida, was established from a Japanese 8-year-old boy with abdominal Burkitt's lymphoma. The P32/Ishida cells were proved to have immature B-cell phenotypes on the basis of immunological surface marker analysis: surface immunoglobulins (gamma, mu, kappa), Ia-like antigen, B1 antigen, and common acute lymphoblastic leukemia antigen were positive. A small amount of IgM, but no IgG, could be detected in both cell extract and culture supernate of P32/Ishida cells. Cytogenetic studies revealed that the P32/Ishida cells consisted of cells having three derivative karyotypes characterized by common marker chromosomes, dup(1), and 14q + due to a new variant chromosomal translocation (2;14), which has not been identified in usual Burkitt's cell lines. Epstein-Barr virus nuclear antigen and terminal deoxynucleotidyl transferase activity were confirmed to be negative.

Effect of partial cystectomy on the induction of pre-neoplastic lesions in rat bladder initiated with N-butyl-N-(4-hydroxybutyl)nitrosamine followed by bladder carcinogens and promoters.

The effect of partial cystectomy on the occurrence of pre-neoplastic lesions, papillary or nodular hyperplasia (PN hyperplasia), of the bladder in male F344 rats was studied in an experiment in which bladder carcinogens and promoters were given to the rats after initiation with BBN. The bladder carcinogens tested were N-ethyl-N-(4-hydroxybutyl)nitrosamine (EHBN) and N-4[4-(5-nitro-2-furyl)-2-thiazolyl]formamide (FANFT) and the bladder promoters were sodium saccharin, sodium cyclamate, and DL-tryptophan. Partial cystectomy significantly decreased the occurrence of PN hyperplasia in rats treated with EHBN and tended to inhibit that in rats given saccharin or tryptophan. Thus partial cystectomy inhibited rather than enhanced the induction of PN hyperplasia.

Increase of inactive form of aspartate aminotransferase in pyridoxine-deficient rat liver.

Cytosolic aspartate aminotransferase from rat liver was separated into at least 3 subforms, focused at pH 6.2, 5.9, and 5.7, by isoelectric focusing. Increase of subforms with low pI values was observed in pyridoxine-deficient rat liver. These subforms with low pI values showed low catalytic activities relative to their antigenic activities. The Km values for substrate and optimal pH values of the two main subforms were not significantly different in pyridoxine-deficient and control rat livers. Some conformational change of enzyme in the cytosol of pyridoxine-deficient rat liver was suggested by circular dichroic and fluorescent spectra. The N and C terminals of the enzyme from both pyridoxine-deficient rats and controls were shown to be alanine and glutamine, respectively.

Histopathological analysis of preneoplastic changes during N-butyl-N-(4-hydroxybutyl)-nitrosamine-induced urinary bladder carcinogenesis in rats.

Sequential microscopic alterations of the urinary bladder epithelium during carcinogenesis were examined in rats after oral administration of 0.01% or 0.05% N-butyl-N-(4-hydroxybutyl)nitrosamine (BBN). Simple hyperplasia appeared after 4 weeks of BBN administration. This regressed by 12 weeks after BBN discontinuation but reappeared focally in some areas after 20 weeks and persisted to the termination of the experiment. Preneoplastic papillary or nodular hyperplasia appeared earlier and more frequently in rats treated with 0.05% BBN than in those treated with 0.01%, but these lesions regressed gradually during a prolonged observation period after BBN was discontinued. These results suggest that 2 types of papillary or nodular hyperplasias exist, one reversible and the other irreversible. Tumors appeared earlier in rats treated with 0.05% BBN than in those with 0.01% BBN.

[Clinical evaluation of antiemetics for vomiting due to cancer chemotherapy in children].

Comparative study of antiemetic effect on vomiting due to cancer chemotherapy was performed in 62 children with various malignant diseases. Twenty-one children were treated with metoclopramide, 23 children with domperidone and remaining 18 children received methylprednisolone. Each drug was administered intravenously after administration of anticancer agents, and repeated if necessary. The most effective antiemetics was methyl-prednisolone with effective rate of 89% in comparison with 51% of domperidone and 17% of metoclopromide treated group, respectively. Methyl-prednisolone may be useful for severe vomiting due to anticancer drugs. Safer and significantly better therapeutic efficacy was observed in the group treated with domperidone than that with metoclopromide.

A novel monocytoid cultured cell line, P31/Fujioka, derived from acute monoblastic leukemia.

A novel cultured cell line, P31/Fujioka, of monocytoid nature was established from leukemic cells in the peripheral blood of a seven-year-old boy with acute monoblastic leukemia. The P31/Fujioka cells have abundant cytoplasm, an indented nucleus of monocytoid appearance, pseudopods detectable by electron microscopy and alpha-naphthyl butyrate esterase activity which is completely inhibited by NaF, but they have no peroxidase activity. Immunologically, the P31/Fujioka cells possess Fc gamma-receptor and phagocytic activity towards sensitized erythrocytes (oxEAIgG), and are reactive with various monoclonal antibodies such as OKM1, anti-Mol, FMC10, FMC12 and OKI1. Chromosome analysis revealed the presence of marker chromosome 11q--due to Nos. 7; 11 translocation and No. 9 pericentric inversion. These findings indicate that the P31/Fujioka cells are derived from the patient's monoblastic leukemia cells and show a more distinct monocyte antigen than other known monocytoid cultured cell lines, U-937 and THP-1. The absence of Epstein-Barr virus nuclear antigen of this line was confirmed.
