Ganglions and ganglionic neurons in the cat's larynx.

Localization, projections and role of ganglia and ganglionic neurons in the laryngeal framework were demonstrated in cats. Six to 8 large size ganglia containing 50 to 80 ganglionic neurons per ganglion in the paraglottic space, 4 to 6 small ganglia involving 5 to 25 cells in each ganglion dorsal to the posterior cricoarytenoid muscle, and 1 to 3 small ganglia including 15 to 25 perikarya per ganglion around the inferior laryngeal nerve were observed. Each ganglion showed spindle shaped covering fibrous capsule. Ganglionic neurons totaling 600 to 800 were oval shaped with an average diameter of 25 microns. Projections of the ganglionic neurons to the superior cervical ganglion (SCG) and nodose ganglion (NG) through the ipsilateral internal branch of the superior laryngeal nerve and to periphery were detected. From the dorsal motor nucleus of vagus, SCG and NG, the ganglionic cells received projections ipsilaterally. On immunocytochemistry many vasoactive intestinal polypeptide (VIP)-immunoreactive (ir) neurons, some neuropeptide Y (NPY)- and tyrosine hydroxylase (TH)-ir neurons and a few substance P (SP)-ir cells were recognized in ganglions. VIP-, NPY-, TH-, SP-, and calcitonin gene-related peptide (CGRP)-ir fibers were also observed in and around vessels and glands. Following denervation, VIP-, NPY- and TH-ir neurons and fibers did not change. These results prove that laryngeal ganglionic neurons have endogenic autonomic, especially cholinergic nature and innervate vessels and glands.

[Autonomic innervation of feline laryngeal glands: an immuno-electronmicroscopic study].

Autonomic innervation of feline laryngeal glands was investigated immunoelectronmicroscopically using antisera for vasoactive intestinal polypeptide (VIP), neuropeptide Y (NPY), and tyrosine hydroxylase (TH). A relatively dense population of immunoreactive (ir) VIP fibers was found in the vicinity of the basement membranes of glandular and myoepithelial cells. Some VIP -ir fibers contacted the basement membrane and some of the fibers penetrated the adjoining glandular cells intercellularly without making synaptic contacts with them. Ir-NPY and -TH fibers were less abundant than VIP-fibers and were distributed beneath the basement membranes of myoepithelial cells. These fibers were not encountered between glandular cells. The estimated ratio of ir-VIP, -TH and -NPY fibers was 20:4:1, based on the density of fibers in glandular tissues. Because this value was equal between serous and mucous glandular cell, these two tissues may receive the same pattern of autonomic innervation.

Antifungal activities of pradimicin derivatives modified at C4'-amino group.

In order to explore potent derivatives of pradimicins (PRMs), modification of their C4'-amino group was carried out. 4'-N-Cyano (1,2), 4'-deamino-4'-nitroguanidino (4), 4'-deamino-4'-ureido (7-9) and 4'-deamino-4'-thioureido (10) derivatives were synthesized by trimethylsilylation of PRMs A and C, followed by condensation with appropriate reagents. 4'-Deamino-4'-guanidino (5) and 4'-deamino-4'-amidino (6) derivatives were synthesized by catalytic hydrogenation of 4 and 2, respectively. 4'-N-Nitroso derivative 3 was prepared by treatment of PRM A with nitrous acid. Among these compounds, the 4'-N-cyano derivative of PRM C (2) exhibited in vitro and in vivo antifungal activities comparable to the parent compounds together with good water-solubility.

Pradimicins FS and FB, new pradimicin analogs: directed production, structures and biological activities.

Exogenous addition of D-serine to Actinomadura spinosa AA0851 resulted in directed production of pradimicin FS, a new D-serine analog of pradimicin S, together with pradimicin FB. Pradimicin FS was produced in higher yields by derivation of ferrous sulfate-resistant strains from strain AA0851. Pradimicin FB, a minor product, was identified as deglucosylpradimicin FL. Pradimicin FS was equivalent to pradimicin S in syncytium formation inhibition activity and in vitro and in vivo antifungal activities.

Synthesis and antifungal activities of pradimicin A derivatives modification of the alanine moiety.

Chemical modifications of the carboxyl group in the alanine moiety of pradimicin A were performed and in vitro and in vivo antifungal activities of the derivatives were examined in comparison with those of pradimicin A. The amide derivatives showed activities comparable to pradimicin A, indicating that the free carboxyl group can be modified without impairing the antifungal activity.

Cephalosporins having a heterocyclic catechol in the C3 side chain. I. Enhancement of efficacy against gram-negative bacteria.

Two groups of cephalosporins substituted with a variety of heterocyclic catechols in the C3 side chain were synthesized. One is a group of 3-(heterocyclic catechol-substituted methyl)cephalosporins and another is 3-[(E)-3-heterocyclic catechol-substituted 1-propen-1-yl]cephalosporins. Cephalosporins in the latter group showed higher in vivo efficacies than those in the former group having the same heterocyclic catechol, especially against Pseudomonas aeruginosa A9843A, although there was no significant difference between their in vitro activity. Among the latter group, the 5,6-dihydroxybenzimidazole derivative (6e) and the 2,6-dihydro-7-hydroxy-6-oxo- isoquinoline derivative (6b) showed much higher activity than ceftazidime (CAZ) and imipenem (IPM) against P. aeruginosa A9843A both in in vitro and in vivo studies.

Autologous BMT in high risk patients with CALLA-positive ALL: possible efficacy of ex vivo marrow leukemia cell purging with monoclonal antibodies and complement.

Autologous BMT (auto-BMT) has been conducted for 17 high-risk common ALL antigen (CALLA)-positive non-T cell type ALL patients. Ex vivo purging of leukemia cells from infused BM cells was performed using complement and three kinds of mouse monoclonal antibodies reactive to CALLA-positive leukemia cells: NL-1 (IgG2a), NL-22 (IgM), and HL-47 (IgM). Minimal residual leukemia cells in BM were examined by PCR method in Philadelphia chromosome (Ph1)-positive ALL cases. BCR/ABL chimeric transcript, which was positive in BM samples before purging, was shown to be absent after ex vivo purging in all three cases tested. Among four Ph1-positive cases transplanted in first CR, three cases survived in CR remission 77, 29 and 26 months after BMT, and one case died without relapse 4 months after BMT. The other four Ph1-negative cases transplanted in the first CR also remained in CR except one who relapsed. The results of minimal residual leukemia cell studies and clinical data indicate the effectiveness of our ex vivo leukemia cell purging method and auto-BMT in the early stage of CR for patients with high risk ALL.

[Analysis of abnormal alkaline phosphatase in patient with high alkaline phosphatasemia].

We describe here a 69-year-old male with high serum alkaline phosphatase (ALP) activity who was showed high alkaline phosphatasemia. High ALP level, 112.4 K.A. was found in his serum. But, except for ALP, all other laboratory data including cancer markers were within normal range in this case. An electrophoretic pattern of patient ALP isozyme without neuroaminidase digestion showed liver-type ALP, but the ALP isozyme pattern with neuraminidase digestion from Vibrio Cholerae was identified as bone-type ALP. Moreover, the patient ALP reacted with anti-bone ALP MoAb. The apparent molecular size of patient ALP was 160kDa similar to bone ALP. Furthermore, we investigated the multiformity of the sugar chain(s) of patient ALP by serious lectin affinity chromatographies. From these results, the increase of multiantennary complex type and/or bisecting GlcNAc type sugar chain(s) of ALP was shown in patient serum. These results suggested that the patient might have a distant metastasis as in a case of carcinoembryonic antigen described previously. Besides, patient urinary pyridinoline and deoxypyridinoline was high level comparing normal adult urine. From detailed bone survey, the patient was observed the bone metastasis of cancer. Thus, the measurement of ALP sugar chain subfractions may be useful for diagnosis of patient with bone metastasis of cancer.

[Clinical usefulness of 111In transferrin scintigraphy in colorectal cancer].

As assessment was made regarding the clinical value of 111In transferrin in scintigraphy on 28 lesions in 26 cases of colorectal cancer. The positive rate of colorectal cancer was high: 21 lesions out of the 28 (75%) were found to be positive. As for the location of cancer, there was a tendency for the positive rate to be high in the ascending and transverse colon. There was no obvious trend regarding Borrmann's classification, histological type, or macroscopic depth of invasion. There was a trend for cases in which the maximum diameter of the tumor was large and depth of invasion was in progress to be positive. Ten cases in which a specimen was resected were all shown to be positive by scintigraphy. Radioactivity in the tumorous regions was 4.41 +/- 2.96 times that of the non-tumorous regions. Moreover, tumorous tissue was strongly stained by the immuno-histological staining with anti-Tf.receptor antibody. From the above findings, it was considered that 111In transferrin is clinically useful in scintigraphy, since it is evident that it accumulates in the tissue of colorectal cancer.

[Analyses of ventricular arrhythmias in patients with progressive systemic scleroderma (PSS) by ambulatory electrocardiography].

Disorder of cardiac conduction and automaticity have been widely reported in patients with progressive systemic scleroderma (PSS). In the present study, ventricular arrhythmias were monitored by 24-hour ambulatory electrocardiographic recording (Holter monitor) in 30 female patients with PSS and 20 apparently healthy female controls. The following results were obtained: 1) In the waking state, 8 cases of 30 patients showed grade III, IVA or IVB of ventricular arrhythmias, according to Lown's classification. On the other hand, ventricular arrhythmias, such as grade III, IVA or IVB, were not observed in the control group. 2) In the patient group, grade IVA arrhythmias were observed during defecation, driving or light working, and grade IVB were during defecation, urination, light working or sleeping. These results suggest that it is important to examine arrhythmias by Holter monitors in patients with PSS.

[Current status of ultrasound examination of the superficial organs (breast and thyroid)].

The ultrasound examination of superficial organs began with the application of the breast and thyroid. After the development of the high resolution real-time echography, this method has also been used for the examination of the salivary gland, parathyroid and lymph nodes, and it has become an indispensable diagnostic equipment, owing to low price, high diagnostic accuracy rate, short examination time and no invasiveness. Recently, colour Doppler equipment has developed and it has altered the reliability of ultrasound. [Breast]. The real-time method brought about the evaluation of compressibility and mobility by the dynamic test, and the interruption of the borderline of the mammary gland was added to the diagnostic criteria for breast cancer. Colour Doppler imaging will become a useful method to diagnose tumours, by evaluating the vascularity, blood flow volume or flow velocity. For nonpalpable tumours, ultrasonically guided aspiration cytology and biopsy were devised and they showed good results. [Thyroid]. Thyroid echography, together with aspiration cytology, became an integral part of the diagnostic method for thyroid cancer, and the necessity of the X-ray of the thyroid has decreased. The accuracy rate for papillary carcinoma recorded more than 80%. In the screening, the detection rate of carcinoma is now 1%. The differential diagnosis between carcinoma and adenoma, which both have a follicular pattern, is a problem yet to be solved. [Parathyroid]. Hyperplasia and adenoma of the parathyroid show hypoechoic and hypervascular area. In addition, the salivary gland and lymph nodes can also be evaluated with echography.

[A case of achalasia evaluated with esophageal scintigraphy in terms of efficacy with endoscopic pneumatic dilatation].

The therapeutic effects on pneumatic dilatation were evaluated quantitatively by radionuclide transit study using 99mTc-Sn colloid in an achalasia patient. After dilatation, transit time (T3/4) with distilled water has improved from 13.5 seconds to 5.5 seconds. Radionuclide transit study using 99mTc-Sn colloid was a preferable method to evaluate the efficacy of the treatment quantitatively in an achalasia.

Resistance to phorbol ester-induced differentiation of a U-937 myeloid leukemia cell variant with a signaling defect upstream to Raf-1 kinase.

Previous studies have demonstrated that treatment of human U-937 myeloid leukemia cells with 12-O-tetradecanoylphorbol-13-acetate (TPA) is associated with growth arrest and induction of monocytic differentiation. The present work describes the isolation of a U-937 cell variant, designated TUR, which is unresponsive to the growth-inhibitory effects of this agent. The results demonstrate that, in contrast to U-937 cells, the TUR line fails to respond to TPA with induction of the c-jun, junB, c-fos, and EGR-1 early response genes. The finding that these cells also fail to exhibit adherence or induction of the tumor necrosis factor and c-fms genes further supports their resistance to TPA-induced differentiation. In contrast, TUR cells responded to 1,25-dihydroxyvitamin D3, another inducer of monocytic differentiation, with growth arrest and induction of early response gene and c-fms transcripts. TUR cells also responded to okadaic acid, an inhibitor of type 1 and 2A protein phosphatases, with similar changes in gene expression. Further characterization of TUR cells has demonstrated decreased expression of protein kinase C as compared to wild-type U-937 cells. We also demonstrate that although treatment of U-937 cells with TPA is associated with activation of the Raf-1 serine/threonine kinase, there was no detectable decrease in electrophoretic mobility of this protein in TPA-treated TUR cells. Taken together, these findings indicate that the TUR variant is defective in TPA-induced signaling events upstream to activation of Raf-1 kinase.

Phorbol ester-induced monocytic differentiation is associated with G2 delay and down-regulation of cdc25 expression.

Treatment of human U-937 myeloid leukemia cells with 12-O-tetradecanoylphorbol-13-acetate (TPA) is associated with growth arrest and induction of monocytic differentiation. The present results demonstrate that exposure of G1- or S phase-enriched U-937 cells to TPA was associated with delayed transition through G2-M and then exit from the cell cycle in the subsequent G0-G1 phase. In contrast, G2-M cells treated with TPA progressed through G1-S, were delayed in the next G2-M phase, and then entered a nonproliferative G0-G1 state. The effects of TPA on expression of cell cycle control genes demonstrated down-regulation of the cdc25 mitotic inducer during S-G2 progression. In order to investigate the role of cdc25 in the control of cell cycle progression, U-937 cells were stably transfected with an antisense cdc25 construct under control of a metallothionein-inducible promoter. Expression of this construct in G1 phase cells was associated with a significant delay in transition through S and G2-M phase. Similar results were obtained when G1 phase cells were incubated with a 14-base pair antisense cdc25 oligomer. Taken together, these results indicate that the cdc25 gene product is functionally associated with S-G2 transition of proliferating myeloid leukemia cells and that down-regulation of this gene by TPA is associated with G2-M delay.

[A case of recurrent colon cancer treated markedly effective with 5'-DFUR].

A 71-year-old male with recurrent colon cancer was admitted to our hospital with obstructive jaundice. Virchow's node metastasis was palpable in his neck. The enlarged intraabdominal lymphnodes due to recurrence and liver metastasis were demonstrated on computed tomography. Bile duct obstruction was treated by expandable metallic biliary endoprosthesis. 5'-DFUR (5'-deoxy-5-fluorouridine) was administered orally at 600 mg daily. Virchow's node metastasis completely disappeared by 46 days after administration of 5'-DFUR. The enlarged intraabdominal lymphnodes and liver metastasis disappeared on computed tomography. No significant side effects from 5'-DFUR were encountered for 18 months. 5'-DFUR appears to be a useful and safe treatment for recurrent colon cancer.
