Tissue reaction to implanted biomaterials.

Tissue reactions to the single-crystal sapphire, titanium, Co-Cr-Mo alloy, and PMMA were histologically evaluated after implantation into the subcutaneous tissue and tibia of rats. In subcutaneous tissue the reaction from early necrotic change and acute inflammation to final encapsulation by fibrous connective tissue was least pronounced around single-crystal sapphire implants. Tissue reactions of a slightly greater degree were found around titanium and Co-Cr-Mo alloy implants. The most prominent tissue reaction was observed around PMMA implants. In the tibia, single-crystal sapphire, titanium, and Co-Cr-Mo alloy implants were encapsulated by newly formed compact bone, but bone was not so evident around PMMA implants. These findings suggest that single-crystal sapphire implants have more favorable tissue compatibility than those of titanium, Co-Cr-Mo alloy, and PMMA.

Treatment of head and neck tumors by contact Nd-YAG laser surgery.

The contact Nd-YAG laser system with ceramic rods which has been developed by us was applied to animal experiments and clinical practice. It was confirmed that: 1) this method can be performed at low power of 6 to 8 W in contact laser incision of soft tissue and at 3 to 4 W in localized laser hyperthermia; 2) it permits accurate and precise incision because mis-shots of laser irradiation can be eliminated in the target tissue; 3) it causes less bleeding with minimal damage to adjacent tissue; and 4) it has remarkably high controllability. The contact Nd-YAG laser systems are very useful as one of the new modalities for surgical procedures as well as localized laser hyperthermia in head and neck tumors.

Properties of rat liver signal peptidase reconstituted into liposomes.

EDTA/KCl- or pyrophosphate-treated rough microsomes of rat liver clearly showed the co-translational cleavage of pre-human placental lactogen and translocation of the product into membrane vesicles. The signal peptidase fraction was isolated by chromatography on Sephacryl S-300 of deoxycholate-treated membranes and reconstituted into liposomes by dialysis or by the Biobeads SM-2 method. Assay of the signal peptidase activity was performed with pre-human placental lactogen synthesized by the reticulocyte lysate system programmed with human placental lactogen mRNA. The signal peptidase reconstituted into liposomes showed stable activity over the temperature range of 0 to 45 degrees C; in contrast, the detergent-solubilized signal peptidase of dog pancreatic membranes was completely inactivated at the unusually low temperature of 37 degrees C. It was shown that this inactivation was due to the presence of detergent. Signal peptidase from rat liver was insensitive to a variety of protease inhibitors, like the enzyme from dog pancreas, but differed from the latter in being inhibited by chymostatin and TPCK.

Cerebral edema associated with acute hepatic failure.

The clinicopathological findings of cerebral edema were investigated in patients with acute hepatic failure autopsied at Okayama University Hospital between 1970 and 1980 retrospectively. Nine (64%) of 14 hepatic failure cases were found to have cerebral edema during a post-mortem examination of the brain. Clinical features of the patients with cerebral edema were not significantly different from those of the patients without cerebral edema. However, general convulsions were observed more frequently in patients later found to have cerebral edema. Moreover, the length of time from deep coma to death was much shorter in the brain edema cases with cerebral herniation than without herniation.

[Syntheses of [prolyl-U-14C]alacepril and its related compounds].

In order to study the metabolic fate of alacepril, an anti-hypertensive agent, the 14C-labeled compound of alacepril and its related compounds were synthesized. [Prolyl-U-14C]alacepril was synthesized in over-all yield of 32.7-38.0% by the mixed anhydride condensation of L-phenylalanine with [prolyl-U-14C]DU-1163, which had been prepared from L-[U-14C]proline and N-(S-3-acetylthio-2-methylpropanoyloxy)succinimide. [Prolyl-U-14C]captopril and [prolyl-U-14C]DU-1227 were prepared in high yields by hydrolysis of [prolyl-U-14C]DU-1163 and [prolyl-U-14C]alacepril, respectively. [Prolyl-U-14C]captopril-cysteine was synthesized by condensation of [prolyl-U-14C]captopril with cystine S-monoxide in 55.0% yield.

Biphenomycins A and B, novel peptide antibiotics. II. Structural elucidation of biphenomycins A and B.

The structures of biphenomycins A and B, novel peptide antibiotics produced by a strain of Streptomyces, have been established as 1 and 2, respectively, on the basis of spectroscopic and chemical evidence. They are unique in that they are cyclic peptides containing a biphenyl moiety included in a 15-membered ring and show potent antibacterial activities especially against Gram-positive bacteria.

New antitumor antibiotics, FR-900405 and FR-900406. II. Production, isolation, characterization and antitumor activity.

The new antitumor antibiotics, FR-900405 and FR-900406, were isolated from the culture broth of Actinomadura pulveracea sp. nov. No. 6049. These compounds which contain sulfur in the molecule, represent a novel class of antitumor agents. FR-900405 and FR-900406 are highly active in mice against experimental tumors and exhibit antimicrobial activity against Gram-positive and Gram-negative bacteria and fungi.
