A successful GABA replacement therapy for choreo-ballistic status in a 9-year-old girl, who had been previously treated medically or surgically without beneficial results, was reported. Involuntary movements completely ceased after intravenous administration of GABA (the maximal doses: 533 mg/kg/day) of 3 weeks' duration. Chorea and ballism are assumed to be caused by hypofunction of the GABA-neurons in the basal ganglia. Administration of GABA has been considered to be ineffective for these involuntary movements, since GABA has been believed not to cross the blood-brain barrier. The present case suggests that a large amount of intravenous administration of GABA gives a chance to treat involuntary movements caused by deficiency of GABA-neurons, although the essential mechanism of intracerebral penetration of GABA remains uncertain.

A murine monoclonal antibody, 1C5, was produced by fusion of spleen cells obtained from mice immunized with CAC-1, a human cell line of adenocarcinoma derived from uterine cervix, and NS/1 myeloma cells. 1C5 can be used for the staining of routine formalin-fixed and paraffin-embedded tissue sections. 1C5-defined antigen was found to have a molecular weight of 26,000. The 1C5-defined antigen was resistant to neuraminidase and trypsin treatment, but sensitive to periodate treatment, indicating that an epitope of the 1C5-defined antigen is a carbohydrate moiety. Immunohistochemical study using immunoperoxidase staining demonstrated that 1C5 reacted with 87% of adenocarcinomas of the uterine cervix, 39% of endometrial carcinomas of the uterus, 100% of ovarian mucinous cystadenocarcinomas, 43% of ovarian serous cystadenocarcinomas, 45% of adenocarcinomas of the colon, and 40% of gastric adenocarcinomas, thus showing the broad reactivity to adenocarcinoma cells of various origins. However, 1C5 did not show any reactivity to ectocervix epithelium, cervical intraepithelial neoplasia, or squamous cell carcinoma of the uterine cervix. In addition, adenocarcinoma of the uterine cervix exhibited strong cytoplasmic reactivity with 1C5, whereas endometrial carcinoma of the uterus showed the luminal reactivity. 1C5 also reacts with 95% ethanol-fixed malignant cells in cervical smears.
The distinctive theta rhythm that appears at the frontal midline during the performance of mental tasks has been designated as frontal midline theta (Fm theta). Fm theta shows individual differences and seems to be related to certain personality traits. In several studies, it has been indicated that low platelet monoamine oxidase (MAO) activity is also associated with certain personality traits. In the present study, we found a negative correlation between the appearance of Fm theta and platelet MAO activity. Subjects with marked extroversion show a high amount of Fm theta and low MAO activity. It is therefore suggested that Fm theta, an electrophysiological marker, may be useful in the investigation of monoamine functions in the central nervous system (CNS) by way of platelet MAO activity, a biochemical marker.
A new anion exchange resin with an imidazolium salt on a epoxide polymer skeleton was synthesized. This white powder material was odorless and tasteless. The in vitro sodium cholate binding of this resin was much more potent than that of cholestyramine. The hypocholesterolemic activity of this resin in cholesterol-fed rabbits proved to be 4.3 times more potent than that of cholestyramine. These results suggest that effective reduction of plasma cholesterol may be achieved with lower doses of this resin.
A method for quantitative analysis of hemoglobin color to estimate the oxygen saturation was developed. The method uses an image-input and -processing system composed of a 3-tube video camera and a digital image analyzer. Using the system connected to a microscope, facilitated diffusion of oxygen in hemoglobin solutions was observed and analyzed in a position-sensitive way. The results confirmed its applicability to this study and gave information about the diffusion mechanism expressed by the empirical formula J = kY, where J is the flux of oxygen, Y is the oxygen saturation of hemoglobin, and k is a constant.
Young exercised rats with a diminished weight gain had a decrease in high density lipoprotein (HDL) cholesterol and phospholipid levels in the plasma and augmented free cholesterol and phosphatide in the aorta. When the weight gain in the trained rats paralleled the gain in non-exercised animals, the values of these lipids were not altered. The levels of aortic free cholesterol in the non-exercised and exercised groups were inversely associated with concentrations of HDL-cholesterol, but were not related to the activities of lecithin cholesterol acyltransferase. In addition, the total cholesterol and phospholipid contents in the aorta negatively correlated with HDL-cholesterol concentrations. We propose that in young exercised rats with a diminished weight gain, the removal of aortic lipids is hampered due to a reduction in HDL-cholesterol.
FK506, a new immunosuppressive agent, was given intramuscularly to rats for 12 days, starting on the day of type II collagen immunization. FK506 in doses of 0.32 mg/kg or more suppressed arthritis and also suppressed humoral and skin test response to type II collagen. FK506 suppressed arthritis only when given during the afferent limbs of immune response (0-4 days), whereas the drug was only marginally effective when treatment was started during the efferent limbs of immune response (7-11 days). FK506-induced immunosuppression continued and/or was maintained throughout the experiments (50 days). These rats immunized with type II collagen and treated with FK506 failed to develop arthritis even following a secondary immunization 50 days later but were fully capable of developing experimental allergic encephalomyelitis. This result suggest that FK506-treated rats develop specific unresponsiveness toward the type II collagen. It is concluded that FK506 is a strong immunosuppressive drug on collagen-induced arthritis.
We investigated the effect of a new immunosuppressant, FK506, on the development of experimental allergic encephalomyelitis (EAE) in rats. EAE developed in 100% of rats immunized with myelin basic protein (MBP) in complete Freund's adjuvant. FK506 in doses of 1.0 mg/kg/day or more prevented the clinical signs of EAE for at least 50 days, when administered intramuscularly 5 days a week for 2 weeks starting on the day of immunization (days 0-4 and days 7-11), and a similar result was obtained, when the compound was given for 5 days (days 0-4). FK506, however, showed a significant but weak effectiveness when started from 7 days after immunization. Delayed-type hypersensitivity (DTH) to MBP developed before EAE, and anti-MBP antibody levels increased. Both humoral and cellular immune response to MBP were completely suppressed in rats treated with FK506. From these results, it is presumed that immunosuppression of cell-mediated immunity and/or humoral immunity by the treatment of FK506 actually causes the decreased incidence noted in the experiment for the development of EAE.
A survivor of the Hiroshima atomic bomb experienced triple cancer of the urogenital organs after a long history of heavy smoking. The cancers comprised a clear cell carcinoma of the right kidney, transitional cell carcinomas of the urinary bladder, left ureter and left renal pelvis, and adenocarcinoma of the prostate.
Peri-implant tissues of the single-crystal sapphire implant connected with neighbouring teeth by a metal bridge-work were examined clinically, radiographically, and histologically in ten monkeys. Professional tooth cleaning was performed during the study. At 3-12 months after insertion, most of the implants were firmly connected to the surrounding tissues and peri-implant gingiva was regarded as healthy, based on various periodontal parameter scores. Destructive changes of the peri-implant bone were not found radiographically. Histologically, peri-implant gingiva was revealed to show similar structure to that of the gingiva around natural teeth. Direct bone-implant interface was observed at 3 months after insertion, while a thin loose fibrous connective tissue layer was present between bone and implant at 6 and 12 months. Such different interrelationship between bone and implant might be attributable to the difference in distribution of functional stress.

Effects of indomethacin, a potent inhibitor of prostaglandin (PG) synthesis, on the fever induced by recombinant human interleukin 1-alpha (rhIL 1-alpha) was studied in conscious rabbits. Intracerebroventricularly administered rhIL 1-alpha induced a dose-dependent increase in colonic temperature that was prominently suppressed by pretreatment with indomethacin given either intracerebroventricularly or subcutaneously. On the other hand, fever induced by intravenous administration of rhIL 1-alpha was not completely suppressed by either subcutaneous or intracerebroventricular indomethacin; a small rise in colonic temperature persisted at approximately 45 min after rhIL 1-alpha injection. This rise in colonic temperature was suppressed when indomethacin was given both intracerebroventricularly and subcutaneously. It is suggested that PGs synthesized in the central nervous system contribute to the IL 1 fever and that part of IL 1-alpha given peripherally is also transmitted into the central nervous system to contribute to IL 1 fever.
Temperature within the brain and the esophagus and at the tympanum were obtained in a 12-yr-old male in a series of experiments that began 8 days after surgery for implantation of a drainage catheter. Fanning the face did reduce tympanic temperature but not temperature in the brain; brain temperatures followed esophageal temperatures. In long-term monitoring, temperature in the lateral ventricle was 0.5 degree C above esophageal temperature and 0.2 degree C below that in white matter 1 cm above, with the offsets fixed throughout the overnight cycle. All temperatures went through similar excursions when the face was excluded from fanning applied to the body. These observations highlight the fact that in humans the defense against hyperthermia takes advantage of cooling distributed over the entire skin surface.

A large surplus portion of the mitral leaflets appears to be involved in the systolic anterior motion (SAM) of the mitral valve in hypertrophic cardiomyopathy (HCM). To understand the exact features and the mechanism of the surplus leaflet, we studies the mitral valve and its ring quantitatively with two-dimensional echocardiography in 17 patients with HCM (5 with large SAM, 6 with small SAM and 6 without SAM) and 10 normal subjects. In the large SAM group, the surplus length of the anterior mitral leaflet (AML) increased but that of the posterior mitral leaflet (PML) did not increase when compared to the other 3 groups. In the large SAM group, the length of AML was greater than in the normal group and the length of PML was greater than in the other 3 groups. The mid-systolic anteroposterior diameter of the mitral ring did not differ among the 4 groups. The mid-systolic position of the coaptation was more anterior in the large SAM group than in the other 3 groups. In conclusion, the surplus size of AML is actually increased in the patients with HCM and large SAM, which is caused by the elongation of both AML and PML with anteriorly displaced coaptation and with no dilation of the mitral ring.
