[Diagnosis and therapy of epididymitis].

140 patients with acute epididymitis were observed between 1979 and 1983 in the Department of Urology of Bundeswehrkrankenhaus Hamburg. The diagnostic measures and therapy were done by standardized criteria. When the first therapeutic regimen was finished 75 of 140 patients (54%) were reinvestigated and treated when necessary. In acute epididymitis infiltration of the spermatic cord with local anaesthesia, suspension and cooling of the testis and broad-spectrum antibiotics for several days are recommended. Effective treatment depends on precisely selected diagnostic parameters which should concentrate on bacteriological and immunochemical investigation of urine and semen and include clinical and radiological examination of the lower urinary tract.

Response of spinal cord oxygen tension to aortic occlusion.

In order to study the factors influencing spinal cord injury resulting from occlusion of the descending aorta, the local surface PO2 (sPO2) was measured in the lumbar region (L4-5) of the exposed spinal cord after laminectomy in the pig. This is the area supplied by the arteria radicularis magna anterior (ARMA). The following aspects were considered: duration of occlusion, blood supply by the ARMA, distribution and width of spinal cord collaterals. The animals were divided into two groups. In group I (n = 7) the descending aorta was occluded immediately below the left subclavian artery; in group II (n = 7) the abdominal aorta was occluded immediately superior to the ARMA. Occlusion induced a rapid sPO2 decrease in all animals, from 29 to 3 mm Hg in group I (-14 mm Hg/min) and from 28 to 8 mm Hg in group II (-12 mm Hg/min). After 45 minutes of occlusion the blood flow through the aorta was released giving a pronounced sPO2 overshoot (36 mm Hg) about 10 minutes later. After 20 minutes there had been a return to initial sPO2 values. The results of this study confirm the hypothesis that spinal cord injury during occlusion of the descending aorta is primarily due to ischemic hypoxia/anoxia.

Dislocated type-A spermatogonia in human seminiferous tubules.

Type-A spermatogonia can be found in different locations throughout the germinal epithelium of human seminiferous tubules. Generally they represent the population of germ cells adjacent to the basal lamina, but under special conditions they may be located in the adluminal compartment. This aberrant location is found not only in the terminal segments but also in the seminiferous tubules of men older than 65 years of age, and in cases of intratubular seminoma. Furthermore, in both cases, type-A spermatogonia may occupy an intermediate position between basal and intraluminal locations, without showing any signs of cytological damage.

[Chlamydia infections in the gynecology outpatient clinic].

Chlamydia trachomatis is a common agens of non-specific genitourinary inflammatory conditions. We examined 269 female outpatients of the clinic. For 16 per cent of the patients Chlamydia trachomatis could be isolated. In these positive cases we found the following distribution: 32.5 per cent in the cervix and urethra, 60.5 per cent only in the cervix and 7 per cent only in the urethra. The clinical symptoms and importance of the disease are discussed.

Three-dimensional analysis of dendritic spines. I. Quantitative observations related to dendritic spine and synaptic morphology in cerebral and cerebellar cortices.

A total of 212 dendritic spines (108 from the visual and 104 from cerebellar cortices of the mouse) were analyzed in serial sections. Dendritic spines (DS) and synaptic active zones (SAZ) were classified according to their shape, and the following quantitative data were measured: DS stalk and bulb diameters, DS length and volume, number of cisterns of the spine apparatus, DS and SAZ surface areas and their mutual proportions. Quantitative relationships between the spine apparatus and the size of DS and SAZ, between the volume and surface area of DS and between the size of DS and the size of SAZ were studied. Thin, mushroom-shaped and stubby DS with simple (circular or oval), complex (perforated, annulate or horseshoe-shaped) and multifocal SAZ were found on terminal branches of pyramidal cell apical dendrites and club-shaped DS with simple (circular or oval) SAZ on spiny branchlets of Purkinje cells. Statistically significant differences were found between all values measured on various DS types in the visual cortex. Linear dependencies of the DS surface area on DS volume and of the SAZ surface area on the DS surface area were established. Only a limited area of DS plasma membrane (7-10%) was occupied by SAZ. This finding indicates a possible functional importance of the SAZ/DS (and possibly also of the total SAZ/total postsynaptic membrane) surface ratio.

Inverted duplication in the genome of the temperate Streptomyces phage SH3.

The DNA of the temperate Streptomyces phage SH3 contains 100 base-pair long inverted repeats separated by a 940 base-pair long segment of DNA as revealed by electronmicroscopic analysis of snapback structures formed after rapid intrastrand reannealing of denatured DNA. The inverted repeat structure was found preferentially at map unit 22 of the circular physical map, in rare cases also in other positions, suggesting a movable character of this genetic element.

Expression of the replication region of phage lambda DNA cloned into pBR322 in E. coli minicells.

Replication region of bacteriophage lambda DNA was cloned into pBR322 plasmid by the use of two restriction enzymes--PstI and HindIII. The restriction analysis of four obtained plasmids revealed that lambda DNA was cloned in both orientations. Recombinant plasmids were transferred to the minicell-producing strain of E. coli and synthesis of the plasmid-mediated proteins was analysed by polyacrylamide-gel electrophoresis. All four recombinant plasmids produced lambda DNA replication proteins pO and pP as well as some proteins specific for pBR322. The orientation of cloned fragment did not affect the synthesis of lambda DNA replication proteins.

Interaction of bacteriophage T7 with Hind-endonuclease-producing Haemophilus influenzae Rd cells.

In order to test the influence of the restriction endonucleases of Haemophilus species on in vivo DNA restriction of phage T7 and its close relative T3 as well as to determine whether the anti-DNA restriction effect of the T7 and T3 ocr+ gene is also expressed in Haemophilus cells, we studied the interaction of T7 and T3 with Haemophilus influenzae serotypes Rd and Rc and Haemophilus parainfluenzae. The results show that T7 and T3 are unable to infect these bacterial species because they do not absorb to the cells.

Telethermography in the diagnostics and management of malignant melanomas.

Telethermography is a harmless, noninvasive method of examining some patients for some conditions. There are more important differential criteria than thermography for the diagnosis of malignant melanomas, but in some cases the proof of a surrounding difference in temperature may make a contribution to the diagnosis of malignant melanoma, especially if it gives decisive information as to the otherwise clinically inapparent extent of the malignancy locally, and thus guides the surgeon to excise widely enough. Also, thermography of the whole body is complementary to established techniques of examination used in evaluation of patients with malignant melanomas. By making visible hypo- or hyperthermic areas, it helps to detect metastases and confirms or denies suspicious findings obtained by other methods of examination.

Genetic study of plasmid-associated zonal resistance to lincomycin in Streptococcus pyogenes.

The phenomenon of zonal resistance to lincomycin, which is characteristic of most clinical isolates with lincomycin resistance in Streptococcus pyogenes, has been studied. These strains grow within a defined concentration range of lincomycin (approximately 60 to 200 microgram/ml), or at lincomycin concentrations below the minimal inhibitory concentration for susceptible strains. It is shown that the zonal growth phenomenon is a stable phenotype and results from induction of resistance only within the zonal concentration range of lincomycin. These strains also possess inducible resistance to erythromycin which is nonzonal in character. One-step mutations to constitutive resistance have been isolated which are of two types: constitutive for lincomycin or for erythromycin, but not for both. Those strains with constitutive erythromycin resistance retain their zonal resistance for lincomycin. Mutants doubly constitutive for both lincomycin and erythromycin can be obtained by a second mutational step from either of the singly constitutive mutants. Satellite deoxyribonucleic acid has been shown to be present in the zonal resistant strains. A plasmid, pSM10419, of 14.9 megadaltons, has been isolated from one of the doubly constitutive mutants and used to jointly transform Streptococcus sanguis strain Challis to constitutive resistance to both lincomycin and erythromycin. From this, a multicopy plasmid of reduced size, pSM10 (5.4 megadaltons), which retains its resistance phenotype, has been isolated and mapped with restriction endonucleases HindIII (three sites), EcoRI (one site), KpnI (one site), and HpaI (one site). The staphylococcal plasmid pC221 (2.9 megadaltons; chloramphenicol resistant) has been fused to pSM10 at the EcoRI site resulting in a chimeric plasmid, pSM10221 (8.3 megadaltons), which retains resistance to chloramphenicol, erythromycin, and lincomycin. pSM10 is therefore suggestive as an effective cloning vehicle for the genus Streptococcus.
