Production of monospecific antisera against antigenic componentsof Mycobacterium bovis (BCG).

Monospecific antisera against four different antigenic components of Mycobacterium bovis (BCG) were produced by immunization of rabbits with precipitates cut out of gels after crossed immunoelectrophoresis (CIE). An important feature is to use a semipurified mycobacterial antigenic preparation, or a cross-reactive antigen, to obtain a simpler pattern in CIE so that a clean precipitate, free from contaminating antigens, can be obtained for immunization. The findings with one particular antigen are shown, and the use of such antisera is discussed.

mu-chain disease in a case of chronic lymphocytic leukaemia and malignant histiocytoma. I. Clinical aspects.

A 27-year-old woman with proteinuria and normal serum creatinine for 10 years developed chronic lymphocytic leukaemia (CLL) and after further 2 years during treatment with prednisone and chlorambucil a malignant histiocytoma appeared. Free mu-chains were demonstrated in serum, but it is possible that mu-chain disease (mu-CD) and Bence-Jones proteinuria had persisted unrecognized for 12 years, since vacuolated plasma cells were found when the primary bone marrow smear was re-evaluated and no glomerular disorders were found. The clinical findings from the now 11 known cases of mu-CD are reveiwed and the immunochemical findings are reported ain part II (Axelsen et al 1976). The patient's mother died of acute leukaemia at the age of 24. The mother's sister has mammary carcinoma and several members of the mother's line apparently have increased tendency to infections. Of 27 family members, none exhibited immunoglobulin defects, but 10 members of the mother's line had B- as well as T-lymphocytosis and 5 members had leukocytosis.

Mu-chain disease in a case of chronic lymphocytic leukaemia and malignant histiocytoma. II. Immunochemical studies.

Agarose gel electrophoresis of serum from the eleventh case of mu-chain disease (mu-CD) showed two distinct abnormal bands due to free light chains and kappa-complexes with other serum proteins. This caused diagnostic difficulties on conventional immunoelectrophoresis, and crossed immunoelectrophoresis with intermediate gel is demonstrated as a superior tool for investigation of mu-CD. A simple rocket immunoelectrophoresis method foe mu-CD screening is also shown. The mu-CD protein had alpha-2-mobility and a part of the protein carried kappa-determinants probably due to secondary binding. Preparative ultracentrifugation and size chromatography on serum demonstrated that the mu-CD protein was present in serum in several sizes, the smallest being a dimer of mu-chains. Due to scarcity of material the abnormal protein could not be characterized further.

Interactions between ricinus agglutinin and human IgM and IgG.

Ricinus agglutinin, purified until homogeneous, precipitates serum glycoproteins with terminal nonreducing galactose residues including IgM and IgG. Almost 100% of IgM reacted with anti-IgM and ricinus agglutinin in quantitative precipitin tests. In similar tests, almost 100% of polyclonal IgG was precipitated by excess anti-IgG, whereas only about 10% reacted with ricinus agglutinin. In quantitative precipitin analyses and affinity chromatography experiments with insolubilized ricinus agglutinin and isolated monoclonal IgG1 and IgG3 proteins, only IgG3 proteins reacted with ricinus agglutinin.

Protein A reactivity of two distinct groups of human monoclonal IgM.

Fifteen human monoclonal IgM proteins could be divided into two groups based on their ability to inhibit binding of a labelled human monoclonal IgM (IgM-Se) to Staphylococcus aureus Cowan 1 (IgM-Se assay); seven were positive and eight were negative. We found complete agreement between positive reaction in the IgM-Se assay, the presence of protein A reactivity in gel diffusion experiments, and high titres in an indirect haemagglutination test detects antibodies to protein A-IgG complexes. The protein A precipitation patterns of IgM and autologous IgG were not identical.

BCG antibody profiles in tuberculoid and lepromatous leprosy.

In sera from 12 patients with polar tuberculoid leprosy, 12 with subpolar tuberculoid leprosy, and 16 with lepromatous leprosy were demonstrated a total number of 125 anti-BCG precipitins by means of crossed immunoelectrophoresis with intermediate gel. Up to 14 different precipitins were found in individual sera, and the complexity in antibody response was higher than previously realized. The specificity of 69% of the antibodies was defined, and these antibodies were titrated in three arbitrary titer units. A highly significant difference (P < 0.002) was found in antibody response between the tuberculoid and the lepromatous group. Due to simplicity, sensitivity, and high resolution, the method used is a promising tool for providing exact data to be used as guidelines for purification of important individual mycobacterial antigens. The need for reference antisera is emphasized.
