Tyrosine hydroxylase was detected immunohistochemically in 3 human fetuses aged 5 1/2-6, 9-10 and 13-14 weeks. In the youngest fetus only the developing sympathetic chains were stained. By 9-10 weeks sympathetic terminals, adrenal medulla, carotid bodies, locus ceruleus, substantia nigra and basal ganglia showed positive reactions. In the oldest fetus a dense sympathetic innervation was present in the ductus venosus. Catecholaminergic neuronal systems, including their terminal axons, are well developed by 2 1/2 months of human gestation. PAP immunohistochemistry has the advantage of being applicable to archival formalin-fixed, paraffin-embedded tissue.
A reproducible animal model for studying pheochromocytoma is described. Rat pheochromocytoma cells (PC-12 line) maintained in tissue culture and injected subcutaneously into rats produce a functioning tumor at the side of inoculation. Animals harboring these tumors develop alterations characteristic of pheochromocytoma including weight loss, hypertension and a contracted blood volume. This model provides an easily reproducible system for studying the pathophysiologic features of pheochromocytoma.
Twenty-eight patients with Parkinson disease (PD) were treated with bromocriptine for at least 2 years (mean, 2.8 years; range, 2 to 5 years). All of them had first been treated with levodopa (alone or combined with carbidopa, as Sinemet) for 7.4 years (range, 1 to 10 years). At the time bromocriptine was started, all were showing increasing disability. In these patients, attempts to increase levodopa resulted in adverse effects, and attempts to decrease levodopa resulted in increased parkinsonism. Bromocriptine (mean daily dose, 56 mg) was added to levodopa and resulted in improvement of at least one stage (Hoehn and Yahr scale) in 21 of the patients. After 2 years, five of these patients continue to maintain this improvement. The remaining patients, although there has been deterioration, maintain some of their original improvement. Bromocriptine, when added to levodopa, results in improvement that is maintained, in part, for at least 2 years. The ratio of bromocriptine to levodopa has to be periodically readjusted.

The treatment of intermittent claudication by physical re-training of a patient with lower limb arteriopathy is not new (Alpert 1969, Cachovan 1976, Kindler 1965, Schoop 1964). It gives an effective improvement of walking distance and of physical performance capacity. Most programs used need the patient to be kept in an hospital and thus entail a significant social cost. We have set up a program for the retraining of patients that are not hospitalized and have checked the short and long term action achieved. The walking distance, the maximum performance in specifical exercises, the walking exercises on a tread mill, are studied during the clearance X133, the total leg blood flow before and after an arterial occlusive hyperhemic test is studied simultaneously.

Tyrosine hydroxylase antigen was localized immunohistochemically in sympathetic neurons from human autopsy tissue. The reaction persists in paraffin-embedded tissue, and the method is applicable to archival specimens. Increased amounts in this antigen per cell may partially compensate for decreased numbers of sympathetic neurons in familial dysautonomia.
