Inhibition of cortical acetylcholine release and cognitive performance by histamine H3 receptor activation in rats.

1. The effects of histamine and agents at histamine receptors on spontaneous and 100 mM K(+)-evoked release of acetylcholine, measured by microdialysis from the cortex of freely moving, rats, and on cognitive tests are described. 2. Local administration of histamine (0.1-100 microM) failed to affect spontaneous but inhibited 100 mM K(+)-stimulated release of acetylcholine up to about 50%. The H3 receptor agonists (R)-alpha-methylhistamine (RAMH) (0.1-10 microM), imetit (0.01-10 microM) and immepip (0.01-10 microM) mimicked the effect of histamine. 3. Neither 2-thiazolylethylamine (TEA), an agonist showing some selectivity for H1 receptors, nor the H2 receptor agonist, dimaprit, modified 100 mM K(+)-evoked release of acetylcholine. 4. The inhibitory effect of 100 microM histamine was completely prevented by the highly selective histamine H3 receptor antagonist, clobenpropit but was resistant to antagonism by triprolidine and cimetidine, antagonists at histamine H1 and H2 but not H3 receptors. 5. The H3 receptor-induced inhibition of K(+)-evoked release of acetylcholine was fully sensitive to tetrodotoxin (TTX). 6. The effects of intraperitoneal (i.p.) injection of imetit (5 mg kg-1) and RAMH (5 mg kg-1) were tested on acetylcholine release and short term memory paradigms. Both drugs reduced 100 mM K(+)-evoked release of cortical acetylcholine, and impaired object recognition and a passive avoidance response. 7. These observations provide the first evidence of a regulatory role of histamine H3 receptors on cortical acetylcholine release in vivo. Moreover, they suggest a role for histamine in learning and memory and may have implications for the treatment of degenerative disorders associated with impaired cholinergic function.

A home-based exercise program for nondisabled older adults.

OBJECTIVES: This paper describes a videotaped, home-based, strength training program, titled Strong-for-Life and reports on its effectiveness in improving muscle strength, psychological well-being, and health status in a sample of older persons. DESIGN AND SETTING: We enrolled 102 nondisabled, community-dwelling older people aged 66 to 87, identified from the Medicare beneficiary list, into a randomized, controlled trial. MEASUREMENTS: Effectiveness was based on change in isokinetic upper and lower extremity muscle strength, psychologic well-being, and health status. RESULTS: Results revealed several statistically significant short-term benefits after 12 to 15 weeks of exercise, especially for men. Younger older adults demonstrated a 10% improvement in knee extensor strength relative to control subjects. Older male exercisers achieved significant differences relative to controls in perceived anger, tension, and overall social functioning. Male exercisers, in general, achieved significant improvement in perceived vigor. Women did not report psychological benefits following participation in the program. CONCLUSION: Study results reveal that the Strong for Life program, designed to be widely disseminated to the nondisabled older population, has many short-term positive benefits.

Representation of mental imagery functions.

A questionnaire was administered to 250 undergraduates to study their conceptions about the efficacy of mental images in thinking. Analysis showed that subjects rated differently the usefulness of visual imagery according to the kind of content rather than the mental process involved.

[Ultrasound in endodontics: S.E.M. verification].

After giving a brief illustration of the complex morphology of the root canal system, the Authors discuss the use of ultrasound in Endodontics. Following a critical evaluation of the literature and of the mechanisms of ultrasonic instruments, they illustrate their experience through a wide selection of S.E.M. photographs of teeth treated in vitro with sodium hypochlorite at various concentrations and for different lengths of time. They demonstrate the synergistic action between ultrasound and irrigation solutions, and conclude that the best results are with 2.5% hypochlorite for 1 minute.

Backfiltration (BKF) in single needle dialysis.

Backfiltration (Bkf) is a potentially very dangerous phenomenon in haemodialysis, when chemical or biological contamination is present in the dialysate. It occurs when the dialysate pressure within the filter exceeds the pressure in the blood compartment, i.e. when Pbo < Pdi + pi o (where Pbo = the hydrostatic blood pressure at the filter outlet, Pdi = hydrostatic dialysate pressure at the filter inlet, pi o = oncotic pressure at filter outlet).
