Thymostimulin in malignant pleural effusions.

Twelve patients were admitted to the trial: 1 primary pleural tumour (mesothelioma), 7 metastases of breast carcinoma, 1 lung epidermoid, 1 lung adenocarcinoma, 1 uterine carcinoma, and 1 ovarian carcinoma. Thymostimulin (TS) was injected intrapleurally at the mean dose of 140 mg (2 mg/kg). The mesothelioma was the only resistant case. Complete remission was obtained in 4 patients after 3 weeks and in 4 within 8 weeks of treatment. This preliminary trial has shown that intrapleural treatment of neoplastic pleural effusion with a thymic hormone preparation can induce rapid and efficient palliation without subjecting the patients to distressing or toxic procedures.

The drug-immune system interaction in cancer therapy.

The interaction between drugs and the immune system during cancer treatment is reviewed. Special attention is given to drug hypersensitivity. The selective activity of drugs on lymphocytes, the induction of new cell antigens, and the optimal schedule of treatment with immunointerfering drugs are discussed.

Human polypeptidic hormone-like substances in microorganisms.

The presence of factors cross-reacting with polypeptidic human hormones (chorionic and pituitary gonadotropins, prolactin, ACTH and gastrin) has been studied with immune staining methods and agglutination tests in schizomycetes and protozoa of different origin. The bacteria strains freshly isolated from tumor patients do not react with the sera, whereas P. maltophilia ATCC 13637, some E. coli strains namely ATCC 12795, K12, 113/3, 1047, a free-living alga (O. malhamensis ATCC 11532) and a protozoon L. enrietti ATCC 30035 cross-react with the sera against hCG and its beta subunit. The last mentioned three bacterial strains (but neither the former one nor the eucaryotic microorganisms) react with rabbit sera raised against human pituitary gonadotropins, and a minority of this population react even with antibodies against human prolactin. B. mycoides ATCC 4004 reacts only with the latter antiserum. None of the examined microorganisms combines with antisera against other polypeptidic human hormones such as ACTH or gastrin. The bacterial production of hCG-like factor(s) by P. maltophilia is fairly high in Brain Heart Infusion Agar, disappears in cultures carried out in glucose-inorganic salt medium either plain or supplemented with growth factors or hCG beta-subunits, and is lost in tetracycline or chloramphenicol resistant mutans. The role played by joint sharing of antigens in conditioning the interaction between different organisms as well as the immune interfering properties of hCG are discussed.

Cardiotoxic effects of antitumor agents.

Cardiotoxic effects related to treatment with some antitumor agents (chiefly anthracycline antibiotics, cylophosphamide, corticosteroids, 5-fluorouracil, and radiation therapy), their association, and contemporaneous administration of other (potentially) cardiotoxic drugs are reviewed. Experimental models, clinical findings, and diagnostic methods are briefly described. Pathogenetic factors and interactions of antitumor drugs with biomembranes, DNA, and immune reactions are discussed.

Autoantibodies and spleen cell-mediated cytotoxicity in adriamycin-induced myocardiopathy in rabbits.

Spleen cells from rabbits affected by adriamycin-induced cardiomyopathy were specifically cytotoxic for rabbit embryonic heart cells in microcytotoxicity tests. The interactions between "sensitized" spleen cells and rabbit embryonic heart cells were observed by scanning electron microscopy. Sera from these rabbits were shown by immunofluoresence to contain antibodies reactive with rabbit embryonic heart, kidney, and muscle cells. Cytotoxic antibodies were not detected. These sera contained factors that enhanced the cytotoxicity mediated by "sensitized" spleen cells and made normal spleen cells capable of destroying targets. The detection of these factors and of cytotoxins was dependent on the method of target-effector cells treatment by serum.

Antiviral activity of a pyrazino-pyrazine derivative.

The 2,3-dihydroxy-6-bromo-pyrazino-[2,3-beta]-pyrazine is a substance selected during the antiviral screening of pyrazino-pyrazine derivatives. The compound shows antiviral activity in vitro against measles, NDV, some influenza viruses and against herpes simplex and zoster, infectious canine hepatitis and vaccinia viruses. It had no effect on ECHO 9 virus. Therapeutic trials showed activity also on herpetic keratoconjunctivitis experimentally induced in rabbits.
