Monoclonal antibody to human beta interferon: characterization and application.

Seven stable mouse hybridomas secreting monoclonal antibodies to human fibroblast beta interferon (IFN-beta) were isolated, all seven of which belonged to IgGl subclass and kappa type. While neutralizing the antiviral activity of human fibroblast IFN-beta, they failed to neutralized both that of human IFN-alpha and human IFN-gamma. These monoclonal antibodies neutralized the antiviral activity of human fibroblast IFN-beta but not that of human IFN-alpha and IFN-gamma. Two of the seven monoclonal antibodies, YSB-1 and YSB-2, showed particularly high neutralization titers in the ascitic fluid. Monoclonal antibodies were purified from cultures of hybridoma grown in a serum-free medium. The purified monoclonal antibodies, YSB-1(5 micrograms/ml) and YSB-2(1 microgram/ml), neutralized IFN-beta from 10EU/ml to 1EU/ml. Human fibroblast IFN-beta was purified to 3.5 X 10(7) IU/mg protein (1.0 X 10(8) IU/mg protein in the peak fraction) by the monoclonal antibody (YSB-2) affinity column chromatography, whereas human recombinant IFN-beta obtained from E.coli was also purified to 6.5 X 10(7) IU/mg protein (1.1 X 10(8) IU/mg protein in the peak fraction) by the monoclonal antibody (YSB-1) affinity column chromatography.

Malignant lymphoma of the oral cavity: clinicopathologic analysis of 20 cases.

Twenty cases of malignant lymphomas (ML) of the oral cavity were reviewed in the light of recent histologic classification. They occurred in 12 male and 8 female patients, with the age range of 11-80 years (median 51 years of age). Eighty percent of patients were stages IE and IIE. Histologically, 15 cases (75%) were ordinary non-Hodgkin's lymphoma (NHL) with diffuse large cell type being the most common. The remaining 5 cases, undefinable by classification scheme for ordinary NHL, were designated as malignant histiocytosis of the oral cavity (3 cases) and peripheral T-cell lymphoma (2 cases), respectively. There were no cases of follicular lymphoma. The present cases comprised 45% of high grade, 40% of intermediate grade, and 15% of low grade malignancy. A follow-up study showed that the stage of diseage, histologic classification, and frequency of mitosis correlated well with survival.

Histological and morphological observations of the paravestibular canaliculus.

The paravestibular canaliculus was studied by light microscopy in serial sections of the temporal bones from otosclerotic patients who underwent fenestration or stapes surgery. In all examined 23 specimens (13 cases), the endolymphatic duct and sac were observed to be normally developed with no pathological findings. The paravestibular canaliculus was found in 14 of the specimens (60.9%). Its course was traced from the proximal to the distal area in 12 specimens, and it appeared to merge into the distal portion of the endolymphatic sac in 7 of them. Twelve of the paravestibular canaliculi contained one vein, and 3 contained several veins. No artery was found. The paravestibular canaliculus was observed to originate from small vascular channels around the vestibule in the otic capsule, lateral to and near the internal aperture of the vestibular aqueduct.

Influence of ptaquiloside on the development of newborn mice.

We attempted to induce neurological disorders in mice by a single administration of various doses of ptaquiloside (PT) within 24 h after birth. Animals which survived for a long period after treatment showed a reduction in body weight, but did not develop any gait disturbance. On the other hand, in treated animals which died before beginning to walk, karyorrhexis in the external granular layer of the cerebellum and karyorrhexis and rosette formation in the neuroblastic layer of the retina were present. In addition, treated animals showed karyorrhexis and great reduction in the number of the cells in the thymus, suggesting cytotoxic and cytolytic effects of alkylating agents on lymphocytes in this organ.

Effect of gamma-aminobutyric acidA receptor agonists and antagonists on the release of enkephalin-containing peptides from dog adrenal gland.

Chromaffin cells of the adrenal medulla are known to store and release catecholamines, Met5-enkephalin (ME)-like peptides and gamma-aminobutyric acid (GABA). The present study documents that stimulation of GABAA receptors located on chromaffin cell membranes of canine adrenal glands, eliciting depolarization of chromaffin cell membranes, modulates the responsiveness of chromaffin cells to splanchnic nerve stimulation. 4,5,6,7-Tetrahydroisoxazolo[5,4-c]pyridin-3-ol (0.143 mmol/2 ml/min), a selective GABAA receptor agonist infused into the aortic pouch, increases the release of ME-like peptides and catecholamines into the adrenal effluent blood. Prior infusion into the aortic pouch of the GABAA receptor blocker, bicuculline (0.05 mmol/2 ml/min), prevents the 4,5,6,7-tetrahydroisoxazolo[5,4-c]pyridin-3-ol-elicited release of both substances. A stoichiometric relationship exists between the release of both substances; 1 nmol/ml of plasma of catecholamines was coreleased with 2 pmol/ml of plasma of ME-like peptides. The chromatographic profile on a Sephadex G-75 column indicates that, after injection of 4,5,6,7-tetrahydroisoxazolo[5,4-c]pyridin-3-ol, various MW forms of ME-like peptides are released into the adrenal effluent blood. A similar profile for the release of ME-like peptides was obtained when electrical stimulation (10 V/6 Hz) of the splanchnic nerve was used as a stimulus. These data suggest that direct stimulation of GABAA receptors causes depolarization of chromaffin cell membranes by a burst of Cl- channel opening and triggers neurotransmitter release.(ABSTRACT TRUNCATED AT 250 WORDS)

[Studies on antibacterial activity of flomoxef and its distribution to serum and intraperitoneal exudate in surgery].

1. Bacteria detected in cases of acute abdominal ailments encountered in the first department of surgery at Hiroshima university and its related facilities during the recent 4 and a half years were reviewed and the antibacterial activities of flomoxef (FMOX, 6315-S) against these isolates were investigated. As a result, this antibiotic was found to show very high activities against Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Klebsiella pneumoniae and Proteus spp. 2. One gram of FMOX dissolved in 100 ml of physiological saline was administered by intravenous drip infusion during operation and its concentrations in serum and intraperitoneal exudate were determined. The intraperitoneal exudate concentration exceeded the serum concentration in 2 hours after the completion of the drip infusion and, reached high enough concentration to be effective against susceptible bacteria with MICs about 12.5 micrograms/ml. The above results suggested that the new cephem antibiotic FMOX for injection should be sufficiently effective against acute peritonitis.

Induction of tumors in ACI rats given a diet containing ptaquiloside, a bracken carcinogen.

Fifteen female ACI rats initially 5 weeks old were each given a diet containing 0.027-0.08% ptaquiloside [(PT) CAS: 87625-62-5], a carcinogen in bracken, throughout the 210-day experimental period. A control group of 20 female ACI rats was given basal diet without PT. Both ileal and urinary bladder tumors developed in all rats in the experimental group. The ileal tumors were multiple and mostly developed in the distal 10 cm of the ileum. These ileal tumors were identified histologically as epithelial tumors, such as adenomas and adenocarcinomas, and also as nonepithelial malignant tumors, malignant fibrous histiocytomas. The urinary bladder tumors were transitional cell carcinomas, keratinizing squamous cell carcinomas, and sarcomas. Papillomas of the urinary bladder were found in 4 rats in the control group. These results show that, like bracken diet, PT induces tumors in both the ileum and urinary bladder.

[Erythrocyte calcium and pH levels during postoperative tetany following radical operations for thyroid cancer].

To investigate the mechanism of postoperative tetany, erythrocyte calcium and pH were consecutively measured in 15 patients who underwent total removal and subsequent autotransplantation of parathyroid glands at the surgery for thyroid cancer. Of these, 8 patients presented tetany signs, but the other 7 patients had no symptoms. In the non-tetanized group, the concentration of erythrocyte calcium decreased by 33.2% and 46.0% on the 3rd and 5th postoperative day, respectively. On the other hand, it increased by 49.8% and 34.6%, respectively, in the tetanized group, showing a significantly different concentration between two groups. No significant differences were observed, however, in the serum calcium level at any time between two groups. The erythrocyte pH as well as the plasma pH in the tetanized group was significantly higher on the 3rd postoperative day, than that in the non-tetanized group. These results suggest the possibility that the elevated cellular calcium content is more closely linked with postoperative tetany than the serum calcium level, and that the elevated cellular pH as well as plasma pH is another causative factor in evoking tetany.

Suppression by beta-lactam antibiotics of thromboxane A2 generation and arachidonic acid release in rabbit platelets in vitro.

High concentrations of latamoxef and some other beta-lactam antibiotics suppressed thromboxane A2 generation as determined from the thromboxane B2 level in the in vitro aggregation of rabbit platelets in agonist-induced stimulations. In aggregation induced with a low concentration of collagen, the suppression of thromboxane B2 generation correlated well with the suppressions of aggregation and serotonin (5-HT) release; collagen produced thromboxane A2-dependent aggregation and 5-HT release as judged from the inhibitory action of indomethacin. Latamoxef also suppressed thromboxane B2 generation when platelet stimulation was induced by thrombin or platelet activating factor at concentrations at which it did not affect either aggregation or 5-HT release. However, latamoxef did not affect platelet responses including thromboxane B2 generation induced by exogenous arachidonic acid or calcium ionophore, A23187. Beta-lactam antibiotics also interfered with arachidonic acid release from the membrane phospholipids of platelets which had been prelabelled with [3H]arachidonic acid and aggregated with collagen. These results suggest that in the in vitro aggregation of platelets, beta-lactam antibiotics interfere with some of the receptor-stimulated processes which lead to arachidonic acid release from the membranes and this, in turn, suppresses thromboxane B2 generation.

Effect of kallikrein-kinin system on calcium-vitamin D3 metabolism.

Kallikrein has been reported to stimulate callus formation and cell proliferation. It is well-known that vitamin D3 play an role on the metabolism of calcium. However, the relation between vitamin D3 and kallikrein are still poorly understood. We have studied the effect of kallikrein on calcium and vitamin D3 with canine kidney. Mongrel dogs were anesthetized with sodium pentobarbital and prepared by the method of Nakanishi et al. Drugs were infused into renal artery for 60 minutes under three different serum calcium concentrations (Exp. I, Exp. II, Exp. III). Blood samples were collected at the midpoint of the 60 minutes urine collection period. Plasma vitamin D3 concentrations were measured by the metabolites of 25-hydroxyvitamin D3 and 1 alpha,25-dihydroxyvitamin D3. 25-hydroxyvitamin D3 was estimated by competitive protein binding assay and 1 alpha,25-dihydroxyvitamin D3 was determined by radioreceptor assay after separated using HPLC. Electrolytes of sodium, potassium, chloride, calcium and phosphorus in serum and urine, plasma cyclic AMP, glomerular filtration rate, renal blood flow and blood pressure were also measured respectively. Plasma 1 alpha,25-dihydroxyvitamin D3 concentration was found to be decreased by the infusion of kallikrein (0.02 KU/kg/min) in three experimental conditions, especially in Exp. II. Bradykinin (0.02 microgram/kg/min) also caused to kallikrein-like changes of vitamin D3. It is suggested that kallikrein-kinin system is related to inhibit the mechanism of vitamin D3 activation system on kidney.

Reexamination of histological findings of ileal sarcomas induced in rats given diet containing bracken fern.

The ileal sarcomas induced in rats given diet containing bracken fern were examined microscopically and electron microscopically. These ileal sarcomas, which were induced at high incidence (80%-44%), have previously been identified as spindle cell fibrosarcomas. However, studies showed that they should be diagnosed as malignant fibrous histiocytomas (MFH) being classified into the following four histological types; fibrous, pleomorphic, giant cell and myxoid type. No types of sarcomas other than MFH were found in the ileum.
