Hydrogenated soybean oil (HSO) as a vehicle for the chronic and controlled administration of testosterone in the orchidectomized rat.

The use of a novel vehicle, hydrogenated soybean oil (HSO) for the chronic and controlled release of testosterone (T) in the orchidectomized rat is described. At 60 degrees C, HSO is a liquid in which T is readily soluble. When cooled at approximately 43 degrees C, HSO can be injected into experimental animals without the need of special equipment. At body temperature, the oil forms a solid depot from which T is released over prolonged periods of time. Our studies indicate that a single injection of 5 mg T in HSO in the orchidectomized rat, maintains sex accessory gland weights (AGW) above castrate levels, decreases serum concentrations of LH and maintains serum concentrations of T within the intact range for 10 days after injection. In contrast, these parameters of functional androgenicity are maintained for only three days when T is administered at the same dosage in a conventional oil vehicle. A dose of 15 mg T in HSO maintained serum T concentrations and AGW within intact range for 55 days after injection, but was unable to maintain LH suppression in these castrated animals beyond 14 days after injection.

[Unusual course in 2 patients with testicular teratoma during vinblastine-cyclophosphamide chemotherapy].

Two patients with metastatic embryonal testicular cancer achieved complete remission receiving a vinblastine-cyclophosphamide combination chemotherapy. After a previous treatment with vinblastine-bleomycin one patient had developed progressive disease, the other one had attained partial remission only. Because of our own experiences and because of the results in single agent chemotherapy this well tolerated vinblastine-cyclophosphamide combination is proposed as maintenance therapy for testicular teratoma. Other maintenance therapies according to the literature and the problem of maintenance therapy in general are discussed.

[Death in utero of a fetus in multiple pregnancies (author's transl)].

Six intrauterine deaths which occurred in the course of our survey of 160 multiple pregnancies are reported here. The way of handling them obstetrically is discussed, taking note of the facts given in the literature. When prevention is possible it rests on consecutive clinical and paraclinical observation. Ultrasound tomography is of the greatest possible value to diagnose poor intrauterine growth in one fetus. When one has died and this has been confirmed, routine Caesarean section seems to be indicated to allow the remaining twin to escape, as much from the risk of chronic fetal distress as from the added risks that follow on the retention of a dead fetus.

Sperm-egg interaction: evidence for boar sperm plasma membrane receptors for porcine zona pellucida.

Freshly ejaculated, noncapacitated boar sperm bind rapidly and in large numbers to pig egg zona pellucida in vitro. In the present study, the number of sperm bound decreased sharply when sperm motility was lowered by energy poisons or by reducing the temperature. Highly motile sperm from humans, guinea pigs, and rats, added at concentrations ten times higher than control sperm, did not bind to the porcine zona. At the same high concentration, a small number of hamster and bull sperm bound to the zona. Binding of boar sperm to the zona pellucida was blocked almost completely by diluted whole antiserum to sperm plasma membranes and by univalent (Fab) antibody to these membranes. When antibody to sperm plasma membrane was first absorbed with plasma membrane vesicles, sperm binding was not inhibited. These results provide direct evidence for the existence of sperm plasma membrane receptors for the zona pellucida of the pig.

On the presence of bridges linking the inner and outer acrosomal membranes of boar spermatozoa.

The presence of structures bridging the inner and outer acrosomal membranes of the equatorial segment of boar spermatozoa was clearly demonstrated in cells that have undergone a variety of treatment procedures to displace the electron-dense contents of the acrosome. En-face sections show bridges to be punctate and not linearly extensive as might be suggested by sections perpendicular to the flat plane of the head. About 4.5 x 10(5) bridges, each measuring 7 nm across and spaced 7 nm apart, are arrayed hexagonally in the equatorial segment, but bridges are not present within the principal segment of the acrosome. Short-term treatment with trypsin partially digests the bridges, but does not disrupt the spacing or strict parallel configuration of equatorial segment membranes. However, short-term treatment with pronase digests most bridges and effectively disrupts the typical configuration of the equatorial segment. Freeze-fracture of the cytoplasmic face of the acrosomal membranes of the equatorial segment reveals a pattern throughout the phospholipid layer of the membrane which is similar to the pattern of bridges present in en-face thin sections of the equatorial segments. The data suggest that numerous bridges link the inner and outer acrosomal membranes of the equatorial segment of the acrosome and they play a major, if not an exclusive, role in maintaining the close spacing and parallel arrangement of the membranes in this portion of the acrosome.

Restriction and modification in Bacillus subtilis: identification of a gene in the temperate phage SP beta coding for a BsuR specific modification methyltransferase.

A gene coding for a modifying DNA-methyltransferase which methylates the central C in the BsuR recognition sequence 5'GGCC was identified in the genome of the temperature Bacillus subtilis phage SP beta. This gene is expressed only after induction of the prophage by either mitomycin C or UV. The presence of active methyltransferase in induced cells leads to modification of BsuR recognition sites in SP beta DNA as well as in heterologous DNA.

Characterization and localization of adenylyl cyclase in membrane vesicles and intact boar and human spermatozoa.

The enzymic properties of adenylyl cyclase in purified membrane vesicles from human and boar spermatozoa are described. Plasma membrane vesicles, which appear to be right-side-out, show a marked increase in activity in the presence of the detergent Triton X-100, manganous ion and alkaline pH. Electron-microscope cytochemical assays indicated the presence of adenylyl cyclase in boar and human sperm plasma membranes and also within the axoneme of intact human spermatozoa. The significance and precautions in the evaluation of the cytochemical data are discussed.

Renal lesions in the hypertensive syndromes of pregnancy: immunomorphological and ultrastructural studies in 114 cases.

One hundred and fourteen women presenting during pregnancy with an abnormally high blood pressure and/or proteinuria had a renal biopsy usually on the 8th day following delivery. The pathological specimens were examined by light and/or electron and/or immunofluorescence microscopy. Forty-one cases were studied with all three techniques. The patients could be allocated to six groups on the basis of clinical criteria. The first two groups (52 patients) showed the typical clinical and pathological features of classical preeclampsia. The remaining 62 women (four groups) had isolated hypertension, and, of these, 42 had a renal pathological pattern similar to that of preeclampsia. These 42 patients also had persistent hyperuricemia. Thus in pregnancy, hypertension and persistently elevated uric acid levels are indicative of glomerular lesions of "pregnancy induced nephropathy".

Morphological observations on the binding of boar sperm to porcine zona pellucida.

Scanning electron microscopy demonstrated that freshly ejaculated boar sperm bind in large numbers to the cumulus-free zona pellucida of porcine oocytes. Sperm were bound head-first although some portions of the flagellum occasionally made contact with the zona. Scanning microscopy also showed that sperm did not undergo an acrosome reaction or penetrate the zona after binding. Binding of boar sperm was visibly inhibited by pretreatment of the sperm by Fab antibodies to boar sperm plasma membranes. The forgoing indicate that specific receptors for the zona pellucida are present and exposed on the surface of non-capacitated boar sperm. In addition, when soluble components which were extracted from sperm by sedimentation at high speed, were preincubated with oocytes, sperm did not bind. Minor surface changes were noted after these treatments, but the zona remained intact and was not penetrated by sperm. Either this treatment inactivated sperm receptors or the outer layer of the zona, which may be the only portion of the zona containing receptors, was removed. Since binding of the sperm to the zona pellucida involved the sperm plasma membrane, we suggest that this membrane is intact when the sperm reaches the zona. Surface changes accompanying capacitation are apparently not necessary for species-specific sperm-egg binding in vitro in the pig.

Direct evidence for formation of hybrid vesicles by fusion of plasma and outer acrosomal membranes during the acrosome reaction in boar spermatozoa.

A variety of treatment procedures was utilized to identify the origin and composition of the vesicles formed during the acrosome reaction of boar spermatozoa. Whether the acrosome reaction occurred spontaneously or was induced chemically the vesicles were hybrid vesicles composed of roughly equal proportions of plasma and outer acrosomal membranes.

Contraceptive effects of intravaginal application of acrosin and hyaluronidase inhibitors in rabbit.

Eight single-drug and combinations of two-drug treatments were used to study the contraceptive efficacy of the intravaginal application of acrosin inhibitors (TLCK; NPGB) and hyaluronidase inhibitors (Compound 53D/k; Phosphorylated hesperidin) in the rabbit. Drug concentrations were selected so that they did not inhibit rabbit sperm motility upon incubation at 37 degrees C for 30 minutes. In the first series, the drugs were added to semen before artificial insemination; in the second series, the drugs were administered intravaginally before artificial insemination with untreated rabbit semen. The data show that these drugs are highly effective intravaginal contraceptives in the rabbit. The data suggest that this is a specific inhibitory effect on fertilization since the drug concentrations used did not affect sperm motility.

Morphologic characteristics of the chemically induced acrosome reaction in human spermatozoa.

The morphologic changes accompanying the acrosome reaction in human spermatozoa, as it is induced by the antibiotic A23187 and calcium ions, are described. The reaction is shown to be similar to that observed in other species when the reaction occurs spontaneously or is induced by physiologic fluids. The reaction in human spermatozoa differs from the chemically induced reaction in other species in that plasma membrane microfilaments, prominent in the boar, and tubular-like elements prominent in boar, rabbit, and monkey sperm, are not observed. Motility remains high when human spermatozoa are treated with A23187 and calcium and it is possible that these agents may be useful in the study of certain causes of infertility.
