A characterization study of a cell line (BS/BEK) from bovine embryo kidney.

A cell line (BS/BEK) which was obtained from bovine embryo kidney tissue, when studied at its 140th passage level it showed the following properties: 1. An epithelial-like morphology, possessing a heteroploid karyotype with a modal chromosome number ranging between 70 and 75 chromosomes. 2. It failed to produce tumors in mice and in hamster. 3. It was shown to be ready susceptible to the replication of several viral agents originated from a variety of animal species. 4. It was not contaminated by mycoplasma or other bacterial spp.

Protection tests in pigs vaccinated with the lapinized Chinese strain of hog cholera virus (HCV) previously adapted in a minipig kidney (MPK) cell line, to challenge infection with virulent HCV.

Pigs which had been vaccinated with the Lapinized Chinese strain of Hog Cholera Virus previously adapted in a minipig cell line cultures (MPK-LC-HCV), resultet to be protected when they were subjected to challenge infection with virulent Hog Cholera Virus (HCV) 6 or 11 months later. The challenge virus was never isolated from any of the vaccinated pigs. The MPK-LC-HCV vaccine induced a significant rise of the antibody titer to the HCV in pigs kept under field conditions.

Intra- and paraprosthetic valve regurgitation. A color Doppler echocardiographic study.

We prospectively followed a cohort of 64 patients bearing an aortic or mitral prosthetic valve (mean follow-up 5.2 +/- 3.2 months) in order to evaluate if color-coded two-dimensional Doppler echocardiography (CFD) could provide some major advantages with respect to pulsed (PW) and continuous wave (CW) Doppler in the diagnostic accuracy of detection of intra-, and paraprosthetic leaks. During follow-up 4 cases of pathologic prosthetic regurgitation ensued and were all correctly and easily identified by CFD while one of them was missed both by PW and CW Doppler. Based on our results we conclude that CFD is the best noninvasive tool actually available for the correct identification of prosthetic valvular regurgitation because it can provide useful accessory information difficult to obtain with other echocardiographic techniques.

MTT colorimetric assay for testing macrophage cytotoxic activity in vitro.

The MTT (3-(4,5-dimethylthiazol-2-yl)2,5-diphenyl tetrazolium bromide) cleavage assay, originally described by Mosmann (1983, J. Immunol. Methods 65, 55) for measuring cell survival/proliferation, has been used successfully to quantitate macrophage-mediated cytotoxicity. Peritoneal macrophages, from control or pyran copolymer MVE2-treated mice and from TU5 and L929 murine cell lines, were used as effectors and targets respectively in a 48 h cytotoxicity test, at three effector: target ratios (10:1, 5:1, 2:1). The amount of MTT reduced by cells to its blue formazan derivative during an additional 4 h of culture was quantified spectrophotometrically at 570 nm using an ELISA reader. A linear relationship between the formazan generated and the number of viable TU5 and L929 cells was demonstrated, together with time-dependent growth characteristics for these cells. The formazan produced by macrophages was independent of their functional state and did not interfere with the target cell signal. MVE2-activated macrophages strongly inhibited the survival/growth of target cells in a dose-related way, whereas the cytotoxic activity of control macrophages was very low. Finally, the MTT method compared favorably with the 3H-TdR uptake method in evaluating macrophage cytotoxicity, and both of them were more sensitive than the 3H-TdR release assay. The MTT cleavage method is a useful alternative to radioisotopic methods for quantitating macrophage cytotoxicity for actively growing in vitro targets. Its main advantages are: (a) sensitivity and reproducibility; (b) elimination of the need for radioactive compounds; (c) ease with which it can be performed and quantified; (d) rapidity.

Whole mouse nitroxide free radical pharmacokinetics by low frequency electron paramagnetic resonance.

The in vivo uptake distribution and reduction of the oxygen-sensitive nitroxide spin label PCA in the mouse monitored by low frequency electron paramagnetic resonance (EPR) spectroscopy are reported. Spectra were obtained from the head and liver regions of pentobarbital anesthetized mice during different circulatory and ventilatory conditions. Identical clearances were found in these regions during normoxia. Moderate hypoxia (10% O2-90% N2) did not significantly affect the spin label reduction rate.

Cerebral cytochrome-C-oxidase copper band quantification in perfluorocarbon exchange transfused cats.

Rapid scanning near infrared spectroscopy (730-960 nm) was utilized to determine cat brain cytochrome-c-oxidase after blood-perfluorocarbon (FC-43) exchange. Spectra were carried out before, during, and after the exchange transfusion on cats with preserved somatosensory evoked potentials and microsphere determined cerebral blood flow (CBF). Carbon dioxide (8%) did not produce an increase in CBF. Difference spectra between the hypercapnic state (8% CO2-92% O2) and post mortem, demonstrated the presence of a broad absorption band centered around 820-845 nm which could be attributed to the oxidized low potential copper ion of cytochrome-c-oxidase. We were unable to further oxidize this band by adding carbon dioxide to the inspired gas mixture. This may be due to the near maximal CBF levels present in this preparation. The spectral data support the hypothesis that near infrared spectroscopy can quantify changes in the redox state of cytochrome-c-oxidase.

Genetic differences in cystic fibrosis patients with and without pancreatic insufficiency. An Italian collaborative study.

To determine the number and frequency of mutations that occur at the cystic fibrosis locus (CF), we have examined the allele and haplotype frequencies of eight polymorphic DNA markers linked to CF in 163 Italian patients who were sub-divided according to their clinical presentations. The distribution of haplotypes for the tightly linked polymorphisms KM.19 and XV-2c differ significantly between patients with and those without pancreatic insufficiency. The haplotype found most commonly in CF chromosomes occurs much more frequently in pancreatic insufficient than in pancreatic sufficient patients. Among the 19 pancreatic sufficient patients, 6 (31.6%) show at least one copy of the rare KM.19 = 1, XV-2c = 2 haplotype, as against 16 of 138 patients (11.6%) with pancreatic insufficiency. In addition, only 5 pancreatic sufficient patients (26.3%) are homozygous for the common 2.1 haplotype, as compared with 88 patients (63.8%) with pancreatic insufficiency. These findings support the hypothesis of allelic heterogeneity at a single locus in CF and suggest that different mutations underlie the presence or absence of pancreatic insufficiency in this disorders.

Prenatal diagnosis and linkage disequilibrium with cystic fibrosis for markers surrounding D7S8.

Three polymorphic DNA markers surrounding the D7S8 locus were tested for their usefulness in the diagnosis of cystic fibrosis (CF) by linkage analysis. The markers correspond to the loci D7S424 and D7S426. These polymorphisms were studied by centers in the U.S., the United Kingdom, the Netherlands, and Italy, using samples from populations throughout Europe and North America. The additional information provided by these probes increased the heterogeneity of the region from 50% to 58% and was essential for a completely informative diagnosis in one family. A very high degree of linkage disequilibrium was found between these markers, which span a distance of approximately 250 kb. In addition, linkage disequilibrium with CF was noted. Significant heterogeneity of linkage disequilibrium was found among the populations, both for the marker-marker pairs and between the markers and CF.

A study of some pathogenetic aspects of bovine viral diarrhea virus infection.

The cytopathic (CP) TVM-2 strain of bovine viral diarrhea virus (BVDV) induced in calves a severe disease, characterized by the clinical picture which is usually reported for the acute primary infection observed under natural conditions. In contrast, the calves inoculated with a different biotype of BVDV, the non-cytopathic (NCP) New York-1 strain, remained clinically normal with the only evidence of virus replication in these calves being the recovery of the virus from their pharyngeal swabbings and blood and also the detection of specific neutralizing antibody in their serums. When calves were immunosuppressed with dexamethasone (DMS), they underwent an overt systemic disease of such a severity that in most of the cases it ended with the death of the animals. This result was obtained with either the CP and the NCP strain of BVDV. Finally, the mixed infection that was obtained in the calves with the CP and the NCP BVDV did not result in any particular unexpected pathological situation. It was speculated that the immunosuppressive activity of BVDV could be a property peculiar to certain isolates of the virus.

A serological comparison of some animal herpesviruses.

Bovine herpesvirus 1 (BHV-1) isolates (Cooper-type strain 4975 and Oxford) were compared in neutralization tests with the bovine herpesvirus 4 (BHV-4) isolate (85/16 TV) and the herpesviruses of red deer (D2839/1) and goats (E/CH). Hyperimmune antiserum was prepared in rabbits against the plaque-selected viruses and endpoint and kinetic neutralization test were made. BHV-4 was clearly different from the other four viruses. The closely-related BHV-1 strains were also related in these tests to the red deer herpesvirus. The Oxford strain seemed rather closer antigenically than the Cooper-type strain to the red deer herpesvirus. Antiserum to the caprine herpesvirus failed to neutralize either BHV-1 strain or red deer virus, but antiserum to the Cooper-type and red deer herpesviruses did neutralize caprine virus to a limited extent.

Immunosuppressive activity of 4'-iodo-4'-deoxy-doxorubicin on humoral and cell mediated immune responses in mice: comparison with doxorubicin.

4'-Iodo-4'-deoxy-doxorubicin (I-DX) is a new halogenated doxorubicin (DX) derivative, selected for clinical testing in view of a number of pharmacotoxicological advantages vis-à-vis the parental compound. It was thus of interest to compare the immunological effects of I-DX to those of DX. The compounds were injected i.v. into mice at equal doses in terms of acute toxicity, over a variety of treatment schedules. When tested in single doses for anti SRBC primary and secondary antibody response, I-DX, unlike DX, was not immunodepressive; on the contrary it often caused a moderate amplification of the response. Following a multiple treatment schedule, I-DX was as depressive as DX only when administered after the antigen, any other treatment regimen, before or concomitant with the antigen, being uneffective. I-DX was completely uneffective and DX only slightly active in delaying skin allograft rejection and in reducing DTH reactivity to SRBC. I-DX was more inhibitory than DX for lymphoid cells in vitro, and both drugs reduced spleen cellularity without modifying the relative percentage of B and T cells after administration in vivo. However, the splenic reactivity to LPS ex vivo, unlike that to ConA, was restored earlier in I-DX than in DX-treated mice.

Hepato-atrial anastomosis as emergency treatment for traumatic rupture of suprahepatic inferior vena cava and hepatic veins.

The case of a 21-year-old patient operated on as an emergency for traumatic rupture of the suprahepatic inferior vena cava (IVC) and hepatic vein (HVs) is presented. Since anatomic reconstruction of the ruptured vessels was considered unfeasible, a direct anastomosis was instituted between the right atrium and the liver capsule around the disrupted IVC and HVs, using extracorporeal circulation, deep hypothermia and circulatory arrest. Hemorrhage was easily controlled and the patient survived. As far as we know, this operation, originally developed for the treatment of the Budd-Chiari syndrome, has never been used before as a life saving procedure under the circumstances described.

Extracapsular cataract surgery and intraocular lens implantation in glaucomatous eyes that had a filtering bleb operation.

We studied 112 eyes of 80 glaucomatous patients who had filtering operations and also developed cataracts. Extracapsular cataract extraction (ECCE) was performed in all eyes. One hundred four (104) eyes had posterior chamber intraocular lenses (IOLs) implanted and eight had ECCE without IOLs; the latter were excluded from this clinical investigation. We evaluated intraocular pressure (IOP) and visual acuity and examined the visual fields during a follow-up period of eight months. We compared these results with the preoperative measurements. This study demonstrates that ECCE and posterior chamber IOL implantation is the optimal solution for glaucomatous eyes that have had a filtering bleb operation to obtain a good functional result and IOP stabilization.

Redox changes in cat brain cytochrome-c oxidase after blood-fluorocarbon exchange.

Rapid scanning near-infrared spectroscopy (730-960 nm) was utilized to determine cat brain cytochrome-c oxidase copper band by blood-perfluorochemical emulsion (Oxypherol) exchange. Spectra were carried out before, during, and after the exchange transfusion on animals with preserved somatosensory-evoked potentials and microsphere-determined cerebral blood flow. Remaining hemoglobin (less than 4% of control) was converted to carboxyhemoglobin that does not absorb in this spectral region. Difference spectra, between an hypercapnic status (8% CO2-92% O2) and postmortem, demonstrated the presence of a broad absorption band centered around 820-845 nm that could be attributed to the oxidized low potential copper ion (CuA) of cytochrome-c oxidase. However, we were unable to further oxidize this band by adding CO2 to the inspired gas mixture, but this inconsistency may be due to the near-maximal cerebral blood flow levels present in this preparation. Cytochrome oxidation by CO2 is normally attributed to increased O2 delivery to the tissue, secondary to an increased cerebral perfusion. We were unable to induce further increases in cerebral blood flow. In contrast, the cytochrome band could be reduced both by lowering fractional O2 concentration and by inducing circulatory arrest. The spectral data support the hypothesis that it is possible to quantify the cytochrome-c oxidase copper band in the near-infrared spectral region.

Regional distribution of cystic fibrosis linked DNA haplotypes in Italy, a collaborative study.

In view of the reported variations of cystic fibrosis (CF) associated haplotypes among populations, a detailed analysis of the distribution of two tightly linked DNA markers (Xv-2c and KM.19) in 18 Italian regions was performed. Haplotypes were determined for 405 CF chromosomes carrying the mutation, and showed significant heterogeneity between the North, Center, and South of Italy and the island of Sardinia. KM.19/PstI Restriction Fragment Length Polymorphism (RFLP) showed significant heterogeneity in these four areas, and was statistically correlated with the geographic distribution of the disease, while Xv-2c/TaqI polymorphism, like the haplotypes of normal chromosomes, was more uniformly distributed over the same four areas. Correlation coefficients between the markers and the mutation have been used to obtain additional indications on the physical distance between the markers and the fibrosis locus.

A new dentinal bonding agent and microfilled resin system: a 2-year clinical report.

The purpose of this investigation was to study the clinical results obtained with the Gluma dentinal bonding system and Pekalux microfilled composite resin. Sixty esthetic restorations were placed by three different operators. Two years of clinical observation revealed good results. Alpha scores were given to 50 restorations for marginal integrity, 52 for color stability, and 54 for anatomic form. Bravo scores were given to eight restorations for marginal integrity, six for color stability, and four for anatomic form. Only two restorations had to be replaced.

Prevention of cystic fibrosis in Italian families by DNA studies.

We performed a feasibility study of prenatal diagnosis in the Italian population by DNA analysis utilizing the probes: metD, metH, J3.11, KM.19 and XV-2c. With these probes, 118 out of 126 families (93.7%) were fully informative. We also tested part of the families with new additional tightly linked probes, E.9 and W3D1.4. With all the probes feasibility was complete in 44 out of 45 families (97.8%). In particular with a set of only 3 tightly linked probes, KM.19 + XV-2c + W3D1.4, 71% of the families were fully informative. We carried out 8 prenatal diagnoses, 4 of which with the polymerase chain reaction (PCR) for KM.19. These data show that these probes are highly informative and can be used for feasibility studies and prenatal diagnosis of CF in the Italian population.
