Dichloroisoprenaline (1 mg), injected intramuscularly into the rat, retarded the elimination of water and retained sodium, depressing both glomerular filtration rate and renal blood flow. A dose of 4 mg had a similar but more intense action. These actions of dichloroisoprenaline resembled those of 2 or 4 mug of (+/-)-isoprenaline given subcutaneously. Dichloroisoprenaline and (+/-)-isoprenaline summed in salt retaining effect but competed in renal vascular action. 2-Isopropylamino-1-naphth-2'-ylethanol hydrochloride (nethalide, 1 mg intramuscularly) was without action during water diuresis. A dose of 4 mg inhibited solely the excretion of water. Nethalide (1 mg) completely antagonized the antidiuretic and vascular actions of 2 mug of (+/-)-isoprenaline; antagonism of the salt retention caused by (+/-)-isoprenaline was less complete. A dose of 4 mg of nethalide antagonized both the salt retaining and the vascular actions of 4 mug of (+/-)-isoprenaline.
The actions of a closed-ring [spironolactone, SC-9420, beta-(7alpha-acetylthio-17beta-hydroxy-3-oxoandrost-4-en-17alpha-yl)propionic acid lactone] and of an open-ring [SC-11480, potassium beta-(17beta-hydroxy-3-oxoandrosta-1,4-dien-17alpha-yl)propionate] spirolactone have been examined on the cat perfused kidney, in concentrations ranging from 5 to 20 mg per 150 ml. of blood. Both compounds had weak aldosterone-like effects and caused antidiuresis with retention of sodium, potassium and chloride. The urinary changes they induced summed with those of aldosterone, without competition. Neither spironolactone nor SC-11480 affected renal blood flow or glomerular filtration rate. Both compounds antagonized the reduction in renal vascular resistance caused by aldosterone in these preparations.

Intravenous injections of adrenaline, but not those of noradrenaline, caused the appearance of a substance which resembled isoprenaline in R(F) value and pharmacological activity in blood withdrawn from the lower abdominal aortae of chloralosed cats rested after acute adrenalectomy and induction of lasting block in autonomic ganglia with hexamethonium. The formation of the isoprenaline-like compound was prevented by pretreatment with pyrogallol but not by pretreatment with reserpine, cocaine, phentolamine, dibenamine, harmaline, or iproniazid. The liver proved a main site of the origin of this isoprenaline-like substance.
