Ultracytochemistry of glycogen particles in human erythroblasts. Semiquantitative observation.

We examined the fine structural distribution of glycogen particles in the cells of the erythrocyte series from thirty individuals with or without hematological disorders using the periodic acid-thiocarbo hydrazide-silver proteinate (PA-TCH-SP) technique, which is known to extend PAS staining to the ultrastructural level. In diseases exhibiting dyserythropoiesis, such as refractory anemia with excess of blasts (RAEB) and juvenile chronic myelocytic leukemia (JCML), glycogen particles remarkably increased. In diseases showing hypererythropoiesis such as iron deficiency anemia, hemolytic anemia, or in cells obtained from very small premature infants, an increase in glycogen particles was observed in most cases. Compared to the PAS staining technique, which also utilizes the periodic acid reaction, the PA-TCH-SP technique appears to be more sensitive for visualizing glycogen particles in PAS negative cells, in addition to its capability of demonstrating the distribution of periodate reactive substance at the fine structural level.

Genetic variants of protease inhibitors against fungal protease and alpha-chymotrypsin from hemolymph of the silkworm, Bombyx mori.

Many electrophoretic variants of hemolymph inhibitors of proteases from Aspergillus melleus and pancreatic alpha-chymotrypsin were found using 126 silkworm strains. Six inhibitors of the fungal protease were detected and eight of chymotrypsin; the distribution of inhibitors among Japanese, Chinese, and European races was investigated. Comparison of electrophoretic patterns from F1 hybrids and parents showed that the offspring produce inhibitors of both parental types. Segregation in F2 and backcrossing suggest that the expression of each inhibitor is controlled in most cases by a pair of alleles which are responsible for strong and null bands. Two bands of fungal protease inhibitors C and D were controlled by codominant alleles. These results suggest that polymorphism of hemolymph protease inhibitors in the silkworm would be a useful experimental system for the study of the genetic control of protease inhibitors.

Spondyloepiphyseal dysplasia tarda with progressive arthropathy.

The clinical and roentgenographic manifestations of six cases suffering from spondyloepiphyseal dysplasia tarda with progressive arthropathy are presented. The roentgenographic features consist of generalized platyspondyly and varying degrees of epiphyseal involvement with conspicuous enlargement of both ends of the short tubular bones of the hands. This disorder appears at an earlier age, and is more crippling than the usual form of spondyloepiphyseal dysplasia tarda. It is suggested that orthopaedic measures are essential for all patients with this disorder, as profound disability may be expected by progressive involvement of the major joints.

Membranous lipodystrophy. A case report.

The case is described of a 35-year-old housewife diagnosed as having membranous lipodystrophy (as described by Nasu et al. in 1970 and called lipomembranous polycystic osteodysplasia by Hakola in 1972). The main symptom of this patient was a slowly progressive dementia. Skeletal symptoms were not seen. The computerized tomogram of the brain showed calcification of bilateral basal ganglia and the plain roentgenograms of the bones revealed cystic radiolucent areas at the distal end of the bones of the patient's extremities. Histological examination of the curretted material from the right talus revealed fa 'membranocystic' pattern. The fatty tissue curetted from the cyst of the talus and the lysosomal enzymes of the white blood cells were biochemically normal. A possible relationship between this disease entity and connective disorders is considered.

Characterization of hemopoietic precursor cells in juvenile-type chronic myelocytic leukemia.

In order to study the pathogenesis of juvenile-type chronic myelocytic leukemia (CML), we examined the colony-forming capacity and colony composition in the bone marrow (BM) and peripheral blood (PB) of three children with juvenile-type CML. Large numbers of granulocytes and macrophage colonies were formed by BM and PB cells. Whole agar culture staining revealed that especially macrophage colonies increased in comparison with normal controls. After removal of carbonyl iron-laden cells with a magnet or deprivation of cells adherent to glass from BM cells, the number of macrophage colonies markedly reduced in comparison with the number of colonies formed by untreated BM cells, suggesting that some of the macrophage colony-forming cells (M-CFC) may have phagocytic and/or adherent activity. Radiation sensitivity and thymidine suicide rate of these M-CFC were not different from those of granulocyte colony-forming cells (G-CFC). The predominance of M-CFC in juvenile-type CML may be one of the reflections of fetal-type myelopoiesis since M-CFC are predominant in cord blood and PB in the neonatal period. Moreover, considerable numbers of erythroid-colony-forming units (CFU-E) were present in PB of all patients. It may be concluded that juvenile-type CML is a panmyelopathy with the predominance of M-CFC.

Properties of protease inhibitors from the haemolymph of silkworms, Bombyx mori, Antheraea pernyi and Philosamia cynthia ricini.

1. Effects of inhibitors in the haemolymph from three silkworms on proteases from the alimentary canal of respective insects were studied as well as those on bovine trypsin and alpha-chymotrypsin. 2. In Bombyx haemolymph, three inhibitor fractions were separated by gel filtration on Sephadex G-75. These fractions differed in the specificity of inhibition for different proteases and in thermal stability. 3. In Antheraea haemolymph, a relatively different elution profile was observed compared to that of Bombyx. Philosamia haemolymph showed the intermediate type of elution pattern. 4. Inhibitors from the haemolymph of Antheraea and Philosamia were heat stable. 5. Distinct electrophoretic patterns of inhibitors were observed in three silkworms.

Comparison of three alkaline proteases from digestive fluid of the silkworm, Bombyx mori L.

1. Digestive fluid proteases of the silkworm, 6B1-3, were separated, partially purified and their properties were compared. 2. These proteases were different in the substrate specificity, effect of inhibitors, Km and influence of Mn2+. 3. Hydrolyzing ability for natural substrates was comparatively high in 6B1, whereas the hydrolysis of synthetic substrates of trypsin by 6B1 was lower than that by 6B2 or 3. 4. The protease activity was sensitive to DFP and PMSF. The soybean trypsin inhibitor differentially affected three proteases. Silkworm haemolymph strongly inhibited the protease activity of 6B2 and 3, but scarcely affected 6B1.
