Fibrinogen from human plasma: preparation by precipitation with heavy-metal coordination complex.

Potassium tetrathiocyanato-(S)mercurate II [K(2)Hg(SCN)(4)] is used in a mild and rapid procedure for the isolation of human fibrinogen from fresh plasma. The final product, 94 to 99 percent of which is coagulable by thrombin, represents an average yield of 80 percent and is stable in solution. It is free of plasmin, streptokinase-activatable plasminogen, and coagulation factors II, V, VIII, X and XIII. Sedimentation analysis reveals a single peak with a sedimentation coefficient equal to 7.0S at infinite dilution. Immunodiffusion on cellulose acetate results in two precipitin lines with rabbit antiserum to whole human serum. The fibrinogen precipitates are unusual in that they are flocculent and readily redissolve.

Alloreactivity. I. Effects of age and thymic hormone treatment on cell-mediated immunity in C57B1/6NNia mice.

C57B1/6NNia mice 1, 12, and 24 months old showed loss of cellular-mediated cytotoxicity with aging. Treatment of the three age groups with different thymic hormone preparations effected their cellular mediated cytotoxicity differently. When cytotoxicity of the thymic hormone treated groups was compared to that of the physiological saline treated group, 1-month-old mice treated with serum thymic factor (FTS) at 1 microgram/mouse and 10 ng/mouse had significantly higher activity, and lower to similar activities at 12 and 24 months; TP5 (active fragment of thymopoietin) at 1 microgram and 10 ng caused significantly higher activity in 1-month-old mice, and lower to higher and significantly lower to similar activity at 12 and 24 months, respectively; TM4 (an analogue of TP5) at 1 ng showed significantly depressed activity in 1-month-old mice, and significantly enhanced activity in 12- and 24-month-old mice; thymosin at 10 micrograms and 1 microgram had slightly lower, but not significant, depression at 1 month, similar activities at 12 months and significantly depressed to higher activity at 24 months. Unimmunized control mice showed significant protection in the 12-month-old mice in comparison to 1- and 24-month-old mice. Different hormone preparations showed age- and dose-dependent effects on the ability of spleen cells to kill P815 mastocytoma. Partial restoration of cytotoxicity was observed in 24-month-old mice treated with FTS, TP5 and thymosin fraction V.

Effects of various treatments on the quantitative recovery of endotoxin from water-soluble metalworking fluids.

Three extraction methods were compared for their effectiveness in the quantitative removal of endotoxin from unused and used bulk water-soluble metalworking fluid (MWF) samples. Soluble, synthetic, and semisynthetic fluids were studied. The three modes of extraction consisted of (1) pyrogen-free water (PFW); (2) PFW and Tween 20 (polyoxyethylene sorbitan monolaurate); and (3) PFW, Tween 20, and sonication. Results suggest that vigorous recovery methods yield higher amounts of endotoxin from MWF samples than mild recovery methods in PFW alone. Additional studies are required to aid in the understanding of the factors that significantly affect endotoxin extraction yields from these fluids.

Clinical management of the organ donor.

Critical care nurses play a vital role in the clinical management of potential organ donors. Knowledge of the physiologic traits a donor will exhibit and the clinical interventions necessary to prevent circulatory collapse are essential in providing care to the organ donor. Maintaining hemodynamic stability in the donor optimizes organ function and improves the chances of successful results in transplant recipients.

Image processing algorithms for digital mammography: a pictorial essay.

Digital mammography systems allow manipulation of fine differences in image contrast by means of image processing algorithms. Different display algorithms have advantages and disadvantages for the specific tasks required in breast imaging-diagnosis and screening. Manual intensity windowing can produce digital mammograms very similar to standard screen-film mammograms but is limited by its operator dependence. Histogram-based intensity windowing improves the conspicuity of the lesion edge, but there is loss of detail outside the dense parts of the image. Mixture-model intensity windowing enhances the visibility of lesion borders against the fatty background, but the mixed parenchymal densities abutting the lesion may be lost. Contrast-limited adaptive histogram equalization can also provide subtle edge information but might degrade performance in the screening setting by enhancing the visibility of nuisance information. Unsharp masking enhances the sharpness of the borders of mass lesions, but this algorithm may make even an indistinct mass appear more circumscribed. Peripheral equalization displays lesion details well and preserves the peripheral information in the surrounding breast, but there may be flattening of image contrast in the nonperipheral portions of the image. Trex processing allows visualization of both lesion detail and breast edge information but reduces image contrast.

Developmentally disabled children can be adopted.

People have often thought that developmentally disabled children were hard to place and poor risks in adoption. However, this research shows that a large number of children of all ages and disabilities were placed for adoption with few disruptions. In light of these findings, agencies may well put more effort into identifying developmentally disabled children as suitable candidates for adoption services.

Relationship between foster care and adoption units serving developmentally disabled children.

There were few differences between developmentally disabled foster children and those who were adopted. The foster children as a group were slightly older and more likely to have intact biological families who continued to maintain contact with them. More differences were found in the agencies' structures and practices. The major problem areas identified during the site visits appeared to be within the child welfare system itself, specifically at the interface between foster care and adoption. Bridging mechanisms were needed between units to temper the differences in perception, values, role expectations, and decision-making criteria used by the specialized staffs. When bridging mechanisms were in place, specialization of function was related to increased effectiveness in placing developmentally disabled children for adoption.
