Selecting and designing cell lines for improved physiological characteristics.

We have developed several approaches to create cell lines with improved characteristics in cell culture. In some cases it has been possible to isolate natural variants with useful properties. Cholesterol independent variants of the mouse NSO myeloma cell line were isolated by cloning in a selective medium. A glutamine independent variant of a hyridoma was isolated by continuous (chemostat) culture under glutamine limited conditions in the presence of glutamate. Choline independent cells were isolated from a choline limited chemostat. In an alternative approach to modifying cell behaviour, we have used recombinant DNA techniques to introduce the glutamine synthetase (GS) gene to a hybridoma. This resulted in glutamine independence and increased productivity.

In vitro evaluation of continuous mixed venous oxygen saturation and hematocrit monitors.

Four venous hemoglobin oxygen saturation monitors, two of which provide hemoglobin monitoring, were evaluated and compared to a performance standard (control) during cardiopulmonary bypass (CPB). The CDI 100 Hematocrit/Saturation Monitor, CDI 400, Cobe Hematocrit/Saturation Monitor, and the Baxter Bentley OxySat values were compared to the Radiometer Copenhagen OSM3 Hemoximeter and the Damon International Equipment Company Microbore Centrifuge Hematocrit. The Bentley OxySat meter was found to be the most accurate venous oxygen saturation monitor, with the CDI 100 and the Cobe Hct/Sat monitors correlating closely with the control. Only the CDI 400 failed to correlate with the control. The hematocrit monitors, although useful as trending devices, demonstrated inaccuracies in measurements requiring further refinement.

Spontaneously arising immortal cell line of rat retinal pigmented epithelial cells.

A continuous cell line of rat retinal pigment epithelium (RPE), named BPEI-1, has been established and characterized. Sheets of pure RPE cells, uncontaminated by choroidal or neural retinal cell types, were isolated from eyes of 7-day-old Long Evans rats and established in primary culture. The primary RPE cells became extensively spread and grew slowly for approximately 1 month, at which time a colony of small rapidly dividing cells spontaneously appeared. Following trypsinization, most of the typical primary RPE cells did not survive and were quickly outnumbered by the smaller cells, which gave rise to a cell line that was grown continuously for several hundred generations. When growing at the maximal rate in media containing 20% FBS (doubling time 18 h), the cells were fibroblastic and nearly devoid of pigment, but were capable of morphologic transition back to a pigmented, epithelioid form when cultured under low serum conditions. Evidence that these cells originated from RPE included specific immunolabeling with antibodies to cellular retinaldehyde binding protein and cytokeratin, negative GFAP immunoreactivity, and demonstration of avid phagocytosis of isolated rod outer segments by these cells. Partial characterization of choroidal cells eliminated the latter cells as possible contaminants which could have given rise to the cell line. The BPEI-1 cell line, and other rat RPE cell lines currently being developed from pigmented normal (LE, RCS rdy+p+) and retinal dystrophic (RCS p+) rats should facilitate biochemical and molecular biological approaches to study of RPE cell function in health and disease.

Clinical cardiopulmonary bypass without systemic anticoagulation.

Heparin-coated perfusion systems have been used to perform cardiopulmonary bypass with decreased systemic heparin dosages. We report the case of a 60-year-old man with end-stage liver disease and severe aortic stenosis who underwent an aortic valve replacement without systemic anticoagulation, employing a heparin-coated perfusion system. In patients with a contraindication to systemic anticoagulation, cardiopulmonary bypass with a heparin-coated system without systemic anticoagulation may be a viable alternative.

Effectiveness of the C-Sharp: reducing ergonomics problems at VDTs.

The use of visual display terminals (VDTs) has been associated with complaints of visual fatigue and body discomfort. In an effort to eliminate some of these problems, an ergonomic emphasis has been placed on the design of computer workstations. A new device, the C-Sharp, has been developed to help reduce some of the ergonomics problems related to long-term VDT usage. The C-Sharp was designed to alleviate visual strain and temporary myopia by reducing the amount of muscular work associated with accommodation and convergence to near targets. It was also designed to eliminate glare. The present study is an ergonomics evaluation to determine whether the C-Sharp meets accepted standards and guidelines. Specifically, the objectives of the research are to determine the effects of the C-Sharp on operator reading and search performance, perceived comfort, body posture, and visual acuity. The C-Sharp is compared with mesh-glare filter and no-glare filter device conditions. Subjects were blocked into three groups based on age and type of vision correction (with or without bifocals). Results indicate that the C-Sharp meets the recommendations of the Americal National Standard for human factors engineering of visual display terminal workstations. No differences in objective performance were found between the three glare device conditions. The C-Sharp allowed bifocal wearers to keep their necks in natural postures rather than tilted backwards. Post-session far visual acuity worsened regardless of the device condition.

In vitro propagation of human ocular surface epithelial cells for transplantation.

PURPOSE: To examine the possibility that ocular surface epithelial cells might be grown in culture for use as grafts. METHODS: The proliferative capacity of epithelial cells cultured from the conjunctiva, limbus, and central cornea of normal human eyes was compared. Single cells disaggregated from approximately 1 mm2 biopsy specimens were serially cocultured with lethally irradiated mouse 3T3 fibroblasts. To study the cells' ability to reform a stratified epithelium, confluent limbal cultures were released as an intact cell sheet with the enzyme Dispase and transplanted to a dermal connective tissue bed in nude mice. Attachment and differentiation properties of the reconstituted epithelium were examined immunohistochemically. RESULTS: Central corneal epithelial cells could not be propagated; they senesced in first or second passage. In contrast, limbal epithelial cells exhibited a substantial (i.e., mean of 23 population doublings) and conjunctival cells a moderate (i.e., mean of 11 population doublings) proliferative capacity. Within 4 days of transplantation to the nude mouse dermis, cultured limbal epithelial cells formed an epithelium 5-6 cell layers thick. The epithelium adhered firmly to the graft bed, and deposition of the basement membrane and anchoring fibril protein collagens IV and VII and laminin was detectable immunohistochemically. The transplanted epithelium displayed limbuslike compartmental expression of keratins K3, K13, and K19, and of the enzyme enolase. CONCLUSIONS: These results support the concept that corneal epithelial stem cells are located in the limbus and indicate that cultured autologous limbal cells may function as grafts to permanently restore the corneal epithelium after severe ocular surface injury.

Tumors produced in rats with a single dosage of 1,2-dimethylhydrazine.

To evaluate possible nephroblastoma induction in young Sprague-Dawley male rats by 1,2-dimethylhydrazine (DMH), agents including inhibitors and stimulators of the carcinogenesis were tested concurrently in 2 experiments. In series A, rats, 27 days of age, were fed the following as supplements in a basal diet at the final wt% given: hydralazine (0.035%), disulfiram (250 ppm), ferrous sulfate heptahydrate (0.55%; 0.11 g% Fe), isotretinoin (240 ppm), dehydroepiandrosterone (0.30%) in addition to selenium (2 ppm; drinker). At day 15, DMH was injected s.c. at 108 mg base/kg; duration on the diets: 51 weeks. Series B comprised 33 day-old males which were partially hepatectomized (control and indomethacin at 10 mg/l by drinker) or bilaterally gonadectomized for comparison vs sham-operated, and intact groups on s.c. injection of estradiol benzoate (15 micrograms/kg), progesterone (30 mg/kg) and diallyl sulfide (100 mg/kg), the respective controls receiving the peanut oil vehicle. Treatments were begun 8 days post-operative and 17 days later, the single dosage of DMH as in the above was injected. The oil solutions were administered at the specified weekly levels for a total of 52 injections, 2 doses being introduced per week for the 1st 3 weeks. Colon adenocarcinomas comprised the main tumors and occurred in about 15-50% of the rats with total frequencies in the respective control ranges except for decrements with the disulfiram- and iron-fed groups. Renal changes were more involved with series B and nephroblastomas of the left kidney occurred in one animal each of the estradiol benzoate- and diallyl sulfide-injected groups. Of interest, bilateral nephroblastomas were present in one of the saline-injected controls which was gonadectomized. Under the conditions explored, concurrent treatment with DMH inhibitors or synergists had a minimal effect on nephroblastoma induction.

Human nasal glandular secretion of novel antioxidant activity: cholinergic control.

Exposure of the respiratory mucosa to oxygen-enriched air contributes to the generation of the lung damage in both adult respiratory distress syndrome and bronchopulmonary dysplasia. Recent work has identified the nasal submucosal gland as the source of diverse molecules important in mucous membrane host defense. We searched for the presence of antioxidant activity in nasal glandular secretions, the absence of which could possibly predispose to oxygen-induced injury. Employing a low molecular weight preparation of nasal secretions (a pooled concentrate passed over a 10,000-dalton molecular sieve), antioxidant activity capable of inhibiting both horseradish peroxidase and Fenton reagent reactions was discovered. The following lines of evidence suggest that submucosal glands are the source of this activity. (1) Antioxidant activity present in resting, baseline nasal washings is significantly increased after cholinergic stimulation either in response to topical methacholine or induced by a gustatory reflex. (2) Application of atropine reduced the antioxidant activity to baseline levels after either of the cholinergic stimuli. (3) Levels of antioxidant activity correlated very closely with the secretion of lactoferrin, a recognized product secreted solely from the serous cell of the submucosal gland. The antioxidant activity is due to novel, previously unrecognized molecules. This activity is found in nasal secretions containing molecules less than 10,000 daltons, is unaffected by N-ethyl maleimide (which inactivates glutathione, another low molecular weight antioxidant), is not associated with the capacity to reduce cytochrome c (as seen with ascorbic acid), and resides in the water soluble pool of secretions (in contrast to vitamin E, another putative antioxidant).(ABSTRACT TRUNCATED AT 250 WORDS)

Informational masking and auditory attention.

Informational masking is broadly defined as a degradation of auditory detection or discrimination of a signal embedded in a context of other similar sounds; it is not related to energetic masking caused by physical interactions between signal and masker. In this paper, we report a systematic release from informational masking of a target tone in a nine-tone rapid auditory sequence as the target is increasingly isolated in frequency or intensity from the remaining sequence components. Improved target-tone frequency difference limens as isolation increases are interpreted as a reflection of increasingly focused auditory attention. The change from diffuse to highly focused attention is gradual over the frequency and intensity ranges examined, with each 1-dB increment in target intensity relative to the remaining components producing performance improvements equivalent to those produced by a 2% increase in frequency isolation. The results are modeled as bands of attention in the frequency and intensity domains. For attention directed by frequency isolation, there is a strong correspondence with auditory filters predicted by the power spectrum model of masking. These data also support the existence of an attention band of intensity, with a bandwidth of about 5-7 dB at the moderate levels used in this experiment.

Uric acid is a major antioxidant in human nasal airway secretions.

Airway mucosal surfaces are potentially subjected to a variety of oxidant stresses. Airway submucosal glands secrete a variety of compounds that may protect the airways from injury. Cholinergically induced nasal submucosal gland secretion has recently been found to contain a low molecular weight nasal antioxidant. In this report, the isolation and identification of this nasal secretory antioxidant are described. Concentrated, cholinergically induced human nasal secretions were fractionated through a 10-kDa sieve and subjected to DEAE anion-exchange chromatography. Fractions containing antioxidant activity were subjected to gel filtration with Bio-Gel P-2 gel (resolution range, 200-2000 Da). The resultant antioxidant fractions were then desalted by gel filtration over the same column equilibrated in HPLC-grade water, yielding only a single peak with antioxidant activity. The absorption spectrum of the purified antioxidant revealed peaks at 238 and 292 nm at pH 7. These peaks shifted to 230 and 280 nm in 0.1 M HCl and 226 and 296 nm in 0.1 M NaOH. Sodium borohydride reduction of the antioxidant had no effect on the UV absorption, whereas platinum-catalyzed hydrogenation ablated all absorption peaks. Uric acid had identical absorption peaks and showed the same chromatographic behavior as the nasal antioxidant activity on both gel filtration and DEAE columns. Uricase (which degrades uric acid) metabolized both uric acid and the purified antioxidant. Uric acid was shown to have antioxidant activity at concentrations greater than 1.5 microM. These data indicate that nasal secretions contain uric acid that serves as an antioxidant.

Cis-acting elements involved in the regulated expression of a human HSP70 gene.

Expression of reporter genes under the control of upstream sequences of a human hsp70 gene was examined in microinjected Xenopus oocytes, transfected monkey COS and human HeLa cells. The genes were strictly heat-regulated in all three cell systems. A 69 nucleotide long segment of hsp70 5' non-transcribed sequence that included at least one functional heat shock regulation sequence was sufficient for heat-controlled expression in Xenopus and monkey cells but not in human HeLa cells. An additional segment of about 200 nucleotides in length was required for optimal activity. This segment contains two heat shock regulation elements, each of which appears to contribute to the overall activity in heat-treated human cells. Upstream non-transcribed sequences of the human hsp70 gene are capable of conferring heat regulation on a heterologous promoter. The potential roles in transcription regulation of a bending center in the TATA box region, a CCAAT-like sequence and some of many potential Sp1 binding sites in the hsp70 5' non-transcribed region were investigated.

Determinants for the DNase I-hypersensitive chromatin structure 5' to a human HSP70 gene.

A DNase I hypersensitive site was detected in chromatin formed over a human hsp70 gene segment after amplification in COS7 cells. Deletion mutant analysis was used to evaluate the sequence requirements for this chromatin structure. Determinants sufficient to form the hypersensitive site are contained in a 280 base-pair sequence corresponding approximately to the region that is hypersensitive. Deletion of sequences from either end of this region resulted in reduced hypersensitivity, suggesting that multiple genetic elements contribute to the formation of this chromatin structure. As has been reported for other heat shock genes, the hypersensitive chromatin structure is present prior to heat treatment and does not change in intensity or position after heat shock, in spite of the fact that hsp70 gene expression is completely dependent on heat induction. Sequence requirements for hypersensitivity were generally similar to those for heat-induced gene expression when mutant plasmids were tested at low copy number (e.g. in HeLa cells or in COS cells without amplification); however, deletion of sequences between -223 and -162 with respect to the start of transcription abolished the hypersensitive site but had no effect on gene expression. A barrier to exonuclease III digestion was detected within this region (near an imperfect inverted repeat sequence centered at position -202), suggesting that proteins are tightly bound to the DNA at this location.

Alcohol detoxification and withdrawal seizures: clinical support for a kindling hypothesis.

There has been speculation that a kindling model may have applicability to alcohol withdrawal syndromes and seizures, suggesting that repeated alcohol withdrawals may lead to increased severity of subsequent withdrawals. We evaluated historical and clinical variables of a group of male alcoholics with (n = 25) and without (n = 25) alcohol withdrawal seizures. We found that the number of detoxifications appeared to be an important variable in the predisposition to withdrawal seizures. The withdrawal seizure group had 12 of 25 (48%) patients with 5 or more previous detoxifications, compared to only 3 of 25 (12%) of the control group. A relationship between alcohol use history and withdrawal seizures was not supported by the data. These findings support the concept that previous alcohol withdrawals may "kindle" more serious subsequent withdrawal symptomatology, ultimately culminating in withdrawal seizures.

Auditory event related potentials in violent and nonviolent prisoners.

The neurophysiologic substrates of violence and aggression have been extensively studied. Although the EEG is often normal in violent persons any abnormalities are generally nonspecific in nature. Evoked potentials have been infrequently used to study such behavior disorders. Long latency auditory event-related potentials (AEPs) were studied in prison inmates incarcerated for unmotivated violent crimes or violence during the commission of other crimes, and in prisoners jailed for non-violent offenses. Habitually violent inmates were compared with an equal number of non-violent prisoners and neurologically and audiologically intact controls. AEPs were recorded to an "oddball" paradigm from vertex and left and right temporal electrodes. None of the prisoners had latencies more than 3 standard deviations beyond the normal group mean. N1 and P2 components were longer in latency and lower in amplitude in prisoners than in controls, but this was not statistically significant. Violent and non-violent individuals did not differ in these measures. P3 was significantly prolonged in latency in violent inmates, but not in those who had committed a violent act. All components were longer in latency on the right in violent prisoners, while amplitudes were lower but not significantly so. This asymmetry was not present in non-violent prisoners or control subjects. Caution is needed in identifying differences between criminals and the general population, and in seeking markers of violent behavior.(ABSTRACT TRUNCATED AT 250 WORDS)

Esterification of 8,11,14-eicosatrienoate and arachidonate into alkylacyl- and diacylglycerophosphocholine by vascular endothelial cells.

Agonist-stimulated phospholipases release arachidonate, but not 8,11,14-eicosatrienoate, from human endothelial cells. One source of the arachidonic acid is deacylation of 1-alkyl-2-arachidonoyl-glycerophosphocholine, with subsequent conversion of some of the resultant lysophospholipid to platelet-activating factor. This study has compared the distribution of incorporated 8,11,14-[14C]eicosatrienoate in alkylacyl-GPC and diacyl-GPC with that of [14C]arachidonate synthesized endogenously by desaturation of the 8,11,14-[14C]eicosatrienoate. Cells were incubated for 24 or 48 hr with 8,11,14-[14C]eicosatrienoate, and the resultant mixture of 14C-fatty acids in the cellular lipids was characterized by gas chromatography. The choline phospholipids were then separated, hydrolyzed with phospholipase C and derivatized to diradylbenzoates. Gas chromatographic analysis indicated extensive incorporation of [14C]eicosatrienoate, as well as [14C]arachidonate, into alkylacyl-GPC. Although the ratio of esterified [14C]arachidonate to [14C]eicosatrienoate was greater in alkylacyl-GPC than in diacyl-GPC, the enrichment with [14C]arachidonate was far less than the ratio of arachidonate/eicosatrienoate released from these cells. These results thus support the hypothesis that the acyl specificity of polyunsaturated fatty acid release is provided by the agonist-stimulated phospholipase A2 rather than the composition of the alkylacyl-GPC.
