Binding of netropsin to a DNA triple helix.

The interaction of netropsin, a minor groove binding drug, with T-A-T triple helix and A-T double helix was studied using circular dichroism spectroscopy and thermal denaturation. The triple helix was made by an oligonucleotide (dA)12-x-(dT)12-x-(dT)12, where x is a hexaethylene glycol chain bridged between the 3' phosphate of one strand and the 5' phosphate of the following strand. This oligonucleotide is able to fold back on itself to form a very stable triplex. Changing the conditions allows the same oligonucleotide in a duplex form with a (dT)12 dangling arm. Circular dichroism spectroscopy demonstrates that netropsin can bind to the triple helical structure. Spectral analysis shows that the bound drug exhibits a conformation and an environment similar in double-stranded and in triple-stranded structure. However, the binding constant to the triple-stranded structure is found smaller than the binding constant to the double-stranded one. Thermal denaturation experiments demonstrate that netropsin destabilizes the triplex whereas it stabilizes the duplex.

[A comparative study of two similar groups of 364 children treated for rheumatic fever according to identical protocols (1955--1961, 1965--1972) (author's transl)].

From 1955 through 1972, all children admitted to two after-care centers for rheumatic fever were examined by the same physicians with the same diagnostic criteria and treated according to the same rules. A comparaison between two groups of children who were examined ten years apart has confirmed the changes which took place in the course of rheumatic fever during this period. The incidence of recidives recorded at admission has dropped considerably. Multiple recidives, too, have grown rarer. The incidence of the forms with cardiopathy at admission has dropped. This is, for the most part, due to the reduction in the number of recidives. The severity of cardiopathies has abated, too, and this goes irrespective of the number of previous attacks. The results of treatment in children with cardiopathy are identical in both groups. Treatment methods have not changed from the beginning to the ending of the study; there is a good ground for thinking that the advances observed are due principally to the secondary prevention and related, therefore, to improvements in the health education level, which leads to a recidive prevention better done than in the past.

Successful maintenance immunotherapy by BCG of non-Hodgkin's malignant lymphomas: results of a controlled trial.

Forty-eight patients less than 65 years old, suffering non-Hodgkin's malignant lymphomas with one or more criteria of poor prognosis, were priorly placed in complete remission by an association of chemo + radio + chemotherapy. They were then randomized to receive either BCG in weekly scarifications for 3 years or no further treatment. Five patients were excluded (four after pathological reassessment); 43 are thus evaluable. There were 13 relapses out of 23 control patients and three relapses out of 20 treated patients (0.01 less than P less than 0.025). This difference is more distinct for clinical stages I and II (0.01 less than P less than 0.025) than for stages III and IV (not significant). Eight patients died in the control group whereas there was only one death in the treated group (0.025 less than P less than 0.05). These results indicate than BCG is useful in maintaining remission in about 40% of the cases and also in increasing their survival, especially in patients with clinical stages I or II.

[Carbon assimilation and taxonomic study of Bacillus subtillis and B. licheniformis].

All 14 strains of B. subtilis can use the following 17 sources of carbon and energy: D-glucose, D-mannose, D-glucosamine, salicin, D-ribose, maltose, sucrose, cellobiose, trehalose, arbutin, starch, mannitol, glycerol, glycerate, pyruvate, fumarate, and L-proline. All 15 strains of B. licheniformis can use the following 41 sources of carbon and energy: D-glucose, D-galactose, D-mannose, D-fructose, D-glucosamine, alpha-methyl-D-glucoside, beta-methyl-D-glucoside, salicin, D-gluconate, saccharate, D-xylose, L-arabinose, L-rhamnose, D-ribose, maltose, sucrose, cellobiose, melibiose, trehalose, arbutin, raffinose, starch, inulin, mannitol, D-sorbitol, glycerol, glycerate, citrate, L-malate, D-malate, mucate, pyruvate, fumarate, alpha-L-alanine, alpha-D-alanine, asparagine, L-glutamate, L-arginine, DL-ornithine, L-proline, and 4-amino-n-butyrate. The 29 strains form two distinct groups. Group A includes the 15 strains of B. licheniformis and 2 strains of B. subtilis; group B is formed of 11 strains of B. subtilis; the remaining strain of B. subtilis belongs to neither group. Bacillus licheniformis is a more homogeneous species than B. subtilis. The percentage of guanine + cytosine in the DNA of all 29 strains was determined. In the 14 strains of B. subtilis the average is 46.3% +/- 1.5. In the 15 strains of B. licheniformis the average is 46.4% +/- 0.9.

Total body irradiation (T.B.I.). Preliminary results of a new technique in 30 patients with hematologic malignancy.

The authors present the preliminary results of a new technique of total body irradiation (T.B.I.) used in 30 patients with hematological malignancy. The schedule proposed is easy to use and reproducible. The clinical and hematologic tolerance is good even for patients having had previous cyclic polychemotherapy and/or large-field irradiation. Low-grade non-Hodgkin lymphomas previously nontreated appear to be the best indication. Further investigations have to be performed particularly to specify the interest of combining T.B.I. and chemotherapy or T.B.I. and radiotherapy in the treatment of non-Hodgkin lymphomas.

Circadian Rhythm

[Double blind randomized trial of metopimazine: for postoperative nausea and vomiting after cholecystectomy].

Eighty-four patients submitted to cholecystectomy were randomly assigned in a double-blind study either to an intravenous dose of 10 mg of metopimazine, three times D-1, for two days or to an identically administered placebo. A definite superiority of metopimazine as an anti-emetic drug was demonstrated. General acceptance was excellent, but arterial pressure might be looked over closely when metopimazine was administered intravenously immediately after a general anesthesia.

[Induction chemotherapy in high risk breast cancer. Results of a prospective therapeutic study (author's transl)].

The action of an induction chemotherapy was studied in 54 patients with extensive localized tumour. An association of doxorubicin (adriamycin), vincristine and methotrexate were used in five days courses every three weeks. Three to four courses were applied before radiotherapy (minimum of two months of treatment). This chemotherapy gave very good immediate objective responses (87% of the cases), especially for breast tumors which follow an acute course. No adverse effects on the later sequences of the treatment program were observed. The follow up period is too short to establish long term efficacy. Nevertheless, it now seems of value to apply the chemotherapy from the onset of the disease: on one hand it represents a preparatory factor which is probably useful in reinforcing the action of loco-regional treatment; on the other hand it represents the first treatment for these high risk breast cancers, whose prognosis is mainly determined by metastasis.

Malignant lymphoplasmacytoid lymphomas. Clinical and evolutive data.

A series of 31 cases of malignant lymphoplasmacytoid lymphomas (excluding Waldenström disease) is analyzed. Two-thirds of the patients initially had localizations elsewhere than in the lymph nodes and presented clinical stage I or II. The median survival is around 4 years and is particularly favorable for stage I and II patients who have received an association of radiotherapy and systematic chemotherapy; the estimated "cure rate" for these patients is around 80%.

[Isolation of bacteria that use that use nitric oxide as a respiratory electron acceptor under anaerobiosis].

Ten bacteria of the genus Bacillus were isolated from pasteurized soils, in anaerobiosis and at 32 degrees C, on peptone broth containing 0.5% KNO2. They are Gram variable rods producing oval spores. They are oxidase positive and have catalase. They grow, in anaerobiosis, on NO-3, NO-2, N2O, and NO as respiratory electron acceptors. These compounds are reduced to N2.

Anaerobiosis

[Immunoblastic lymphosarcomas. Clinicopathological aspects and course. Retrospective analysis of a series of 50 patients (author's transl)].

The immunoblastosarcoma (IB) was preceeded by another pathological state in almost one third of patients. The clinical picture was often very typical, with large inflammatory node involvement. In one third of cases, the initial site of the tumor was not nodal, but in Waldeyer's ring or a non-lymphoid organ. Analysis of topographic extension indicates a weak propensity to dissemination. The tissues which are the more frequent site of secondary invasion were the lung parenchyma and the subcutaneous tissue. The course was rapid, almost always fatal. Only a combination of radiotherapy and chemotherapy would seem capable of eradicating the tumour. On the basis of all these characteristics, the IB has a special place alongside other non-Hodgkin lymphomas and they may be individualised histologically. These cases are in general in agreement with the little data published in the literature.

Ocular toxoplasmosis and Hodgkin's disease: report of two cases.

Isolated ocular toxoplasmosis developed in two patients with Hodgkin's disease. The diagnosis was made by ophthalmoscopy of the fundus, serologic tests, and evolution. This raises the possibility that Toxoplasma infection and the localization of retinochoroiditic lesions on the macula could be the results of the immunosuppression associated with Hodgkin's disease.

BCG in the immunotherapy of non-Hodgkin's malignant lymphomas: preliminary results of a controlled trial.

Thirty-three patients exhibiting non-Hodgkin's malignant lymphoma with one or more criteria of poor prognosis and in whom complete remission had been induced by an association of chemo + radio + chemotherapy were randomized to either receive BCG or no further treatment. The present results indicate that BCG may be useful to maintain remission in about one-third of the patients, but these results are not yet significant.

[Denitrification by Bacillus licheniformis].

The denitrifying capacity of 15 strains of Bacillus licheniformis was evaluated. In general, N2 production by the cultures on complex media containing NO3- is irregular and quite slow and three of the strains never produce gas. Bacillus licheniformis grows rapidly in anaerobiosis on peptone medium containing NO3- which is reduced to NO2-. None of the strains grow in peptone medium with NO2- or N2O as the respiratory substrate, nor do they grow under an atmosphere of 10% NO-90% N2. Denitrification was studied in cell suspensions using gas chromatography. N2O production from NO3- or NO2- is always weak at best; nitric oxide is reduced to N2O at an appreciable rate. All the strains synthesize nitrate reductase A in anaerobiosis when NO3- is present. In cell extracts, nitrite reductase activity is always negligible or nil with tetramethyl-p-phenylenediamine as an electron donor.

Anaerobiosis

[Physiological study and taxonomy of Alcaligenes species: A denitrificans, A. odorans and A faecalis].

We have studied 43 strains of the species Alcaligenes dentrificans, A. odorans, and A. faecalis. Twenty-five of them were isolated by enrichment culture on minimal medium containing an organic acid (L-malate, succinate, tartrate, adipate, or itaconate) and N2O as a respiratory electron acceptor. These constitute a single phenon with the A. dentrificans strain type and 9 other strains isolated from clinical specimens. However, strain 4 differs from the other 34 strains in 12 nutritional characters, in its ability to effect a meta cleavage of diphenols, and by the absence of tetrathionate reductase. The percentages of G + C are the following: strains isolated from soil, 66.4 +/- 1.1; collection strains, 67.0 +/- 1.3. The 5 strains of A. odorans differ from the 34 strains of A. denitrificans (not including strain 4) in their inability to denitrify nitrate and use D-saccharate, adipate, pimelate, suberate, beta-hydroxy-beta-methylglutarate meso-tartrate, azelate, and itaconate. Their percentage of G + C is much lower: 56.1 +/- 0.4. From the nutritional point of view the 3 strains of A. faecalis resemble A. dentrificans. However, they differ from the latter by their inability to grow anaerobically on NO3-, NO2-, N2O, and by a slightly lower percentage of G+ C: 64.3 +/- 0.0. The 43 strains synthesize poly-beta-hydroxybutyric acid. None of them is chemolithotrophic.

Alcaligenes

[Induction chemotherapy of non-Hodgkin malignant lymphomas. Results of a controlled trial comparing two quadruple associations (author's transl)].

Two types of chemotherapy have been compared in a randomized trial to treat non-Hodgkin malignant lymphomas. 66 patients were included in this study, but only 40 were evaluable after a histologic review of all cases. Treatment were an association of cyclophosphamide, vincristine, prednisone and doxorubicine or VM 26. Patients received only one induction chemotherapy course during 15--20 days. Results were evaluated immediately at the end of the course. Side-effects were mild. Efficacy was about the same with the two protocols: 35 patients out of 40 experienced a remission of more than 50% (among them 8 experienced a complete remission). These results are better than previous one obtained with cyclophosphamide, vincristine and prednisone only. It is concluded that this type of treatment is well tolerated, quickly efficient and useful before treating patients with radiotherapy or long-term chemotherapy.
