Purification and characterisation of bovine WC1+ gammadelta T lymphocytes from peripheral blood.

In order to isolate and characterise resting WC1+ gammadelta T cells from cattle, we developed a protocol for purifying these cells by negative selection from peripheral blood. The purification method included five steps: separation of mononuclear cells on lymphoprep, depletion of monocytes by adherence to plasma-coated gelatin, enriching T cells on a nylon wool column, depleting CD2+ T cells by sheep red blood cells (SRBC), and finally depleting CD4+ and CD8+ T cells by the magnetic cell sorting technique (MACS). This procedure proved efficient and reproducible, and the purity of the isolated WC1+ gammadelta T cells was more than 97% as analysed by flow cytometry (FACS). Cytokines and costimulatory molecules mRNA expression was assessed by the reverse transcriptase polymerase chain reaction (RT-PCR) technique in freshly isolated resting WC1+ T cells. We found that purified uncultured WC1+ T cells express TNF-alpha, CD28, CTLA-4 and IL-2R alpha mRNA transcripts but do not express those for IL-2, IL-4, IL-6, IL-10 and IFN-gamma. The expression of CD28 and CTLA-4 transcripts on bovine WC1+ T cells indicates that these genes are evolutionarily conserved.

Dissociating mental transformations and visuo-spatial storage in working memory: evidence from representational neglect.

A study is reported of visuo-spatial working memory in two individuals suffering from a cognitive deficit known as unilateral spatial neglect, and seven healthy control participants. Both patients have difficulties reporting details on the left side of imaged representations, and one has an additional difficulty with perceptual input to the left of his body midline. All participants were asked to report the location and identity of objects presented in novel 2 x 2 arrays that were either present throughout or were described orally by the experimenter, with no visual input. On half of the trials, the report was to be made from the opposite perspective, requiring 180 degree mental rotation of the mentally represented array. The patients show an impaired ability to report details from the presented or the imagined left, but had no difficulty with mental rotation. Results point to a clear separation between the processes of perception and those of visuo-spatial working memory. Results also suggest that the patients might be suffering from damage to the system used for holding visuo-spatial representations rather than a difficulty with attending to elements of that representation.

Mouse hypersensitivity pneumonitis: depletion of NK cells abrogates the spontaneous regression phase and leads to massive fibrosis.

The contribution of natural killer cells (NK) in the progression of mouse hypersensitivity pneumonitis induced by repeated intranasal instillations with the thermophilic actinomycete Faeni rectivirgula was examined. These instillations determined a very large increase in the lung index (ca. twofold at 3 weeks), used as a measure of inflammation. In addition, this instillation was associated with a tenfold increase in the number of cells recovered in the bronchoalveolar lavage (BAL) at 3 weeks and thereafter. Most of these cells were macrophages, whereas T lymphocyte numbers increased at 6 weeks and thereafter. The instillations were also associated with a substantial fibrotic response in the lungs, as seen by large increases in hydroxyproline levels in the lungs. This fibrotic response, however, diminished after 6 weeks of instillations. Similarly, examination of histological preparations of lungs of challenged mice showed that F. rectivirgula induced inflammatory infiltrates of macrophages, lymphocytes, and neutrophils. The severity of the lesions were reduced in mice given more than 6 weeks of the actinomycete challenge. The involvement of NK cells on the development of this pulmonary pathology was determined by infusing F. rectivirgula-challenged mice with anti-NK 1.1 antibody. The depletion protocol was validated by verifying that such treatments effectively blocked lung NK cell activity. Such NK cell-depleted mice responded to the F. rectivirgula challenge with an increased lung index at 9 and 12 weeks, compared to mice challenged with F. rectivirgula and given control antibody. NK cell-depleted mice also responded to the actinomycete with a superior cellular recruitment in the BAL, with this increase mostly mediated by macrophages. Similarly, NK cell-depleted mice developed a fibrotic response that was much higher than that seen in control challenged mice, at 6, 9, and 12 weeks after initiation of the transnasal instillations. This was corroborated by scoring the severity of the histopathological lesions, which showed that NK cell-depleted mice had more severe lesions than challenged control mice. The importance of NK cells was confirmed by demonstrating that mice given anti-NK 1.1, challenged with F. rectivirgula and reconstituted with Percoll gradient-enriched lung NK cells had hydroxyproline levels at 9 and 12 weeks that were comparable to that seen in intact mice, as well as a histopathological score similar to control challenged mice. Overall, this suggests that in the course of a pulmonary inflammatory response, NK cells exert a suppressive effect on cellular recruitment in the BAL, contribute to down-regulating the inflammatory response, and are involved in blocking the appearance of fibrosis.

Cellular and cytokine profiles in spontaneous regression phase of hypersensitivity pneumonitis.

The phase of spontaneous regression of hypersensitivity pneumonitis was evaluated using a mouse model. C57BL/6 mice were instilled intranasally with 150 micrograms of the thermophilic actinomycete Faeni rectivirgula 3 days a week so as to establish a mouse model of farmer's lungs. It was shown that instillation of mice for a period of more than 6 weeks was associated with a significant decrease in the lung inflammation, suggestive of the so-called spontaneous regression phase seen in this pathology. Indeed, the lung index was seen to decrease after more than 6 weeks of treatment (2.2 after 6 weeks vs. 1.7 at 12 weeks, p < .01). There was also a significant decrease in lung hydroxyproline levels in animals given 12 weeks of treatment (175 micrograms/lung) compared to 6-week-treated animals (212 micrograms/lung, p < .05). Treated mice did not show a significant decrease in the alveolitis after 9 weeks of treatment. Also, there was no evidence that there was a decrease in bronchoalveolar lavage macrophage or T lymphocyte activity in mice given more than 9 weeks of F. rectivirgula treatment, as judged by O2- release and antigen-driven proliferation. Conversely, it was shown that NK cell activity in the lung digest of mice given 9 to 12 weeks of instillation was significantly higher than that seen in mice given 6 weeks of treatment. Analysis of the lung cell cytokine profile seen after ConA mitogenesis showed that after 6 weeks of F. rectivirgula treatments, nonparenchymal cells secreted high levels of tumor necrosis factor alpha (TNF alpha) and granulocyte macrophage colony-stimulating-factor (GM-CSF), whereas similar cells from the lungs of mice given 9-12 weeks of treatment secreted larger amounts of interferon-gamma (IFN gamma) and interleukin-2 (IL-2). Overall, these results suggest that the spontaneous regression phase is associated with changes in NK cell activity and lung cell lymphokine profile.

[Clinico-biological evaluation of malnutrition].

Malnutrition is frequently observed at hospital, concerning 30 to 50% of hospitalised patients. Increased length of stay and cost of care has made of this problem a major economic stake. Indeed, malnutrition diagnosis and prevention has become, since 2001, one of major french government's priority. Malnutrition results from unbalanced nutritional requirement and in-take. It associates both weight, proteic and functional loss. Its diagnosis and evaluation of its gravity are first clinical and need body mass index determination. For biological diagnosis, nutritional markers have to be very sensitive to nutritional state. Nutritional profile can be made, associating two nutritional markers (albumin and transthyretin) and one inflammation protein (alpha 1-acid-glycoprotein). Various clinical, biological or clinico-biological indexes can also be calculated. Altogether, both indexes and nutrition profile are excellent tools for (1) diagnosis and classification of malnutrition state, moderate or severe, (2) prognostic and (3) evaluation of nutritional supplementation efficiency.

[Dysalbuminemia].

Albumin is the major circulating protein. It plays a fundamental role maintaining intra-vascular oncotic pressure and carrying many endogenous and exogenous substances. Variations of plasma albumin levels can be physiologic or pathologic and both qualitative and quantitative (more frequent) disorders are regrouped under the name "dysalbuminemia". Although hypoalbuminemia are frequent, analbuminemia exists but is a rare disease. Qualitative disorders, mainly bisalbuminemia, are benign. Detected fortuitously on sera protein electrophoresis, bisalbuminemia could be genetically transmitted, it will then be permanent, or acquired and then be transient. This article proposes to review main kind of dysalbuminemia usually encountered in clinical biology laboratories.

Value of an immunofluorescence assay for the detection of Pneumocystis carinii in Bronchoalveolar lavage.

An indirect immunofluorescence monoclonal antibody assay was found to have higher sensitivity than usual stains for the detection of Pneumocystis carinii, particularly in bronchoalveolar lavage fluids in which there are only a few parasites, as in HIV-patients or in HIV+ patients with prophylaxis or treatment. For patients without any therapy, when different stains give conflicting results, decisions on therapeutic approaches to be used should consider the patient's clinical and biological status. Prospective studies are necessary to evaluate the predictive value of low parasitism in asymptomatic immunosuppressed patients.
