Factors from Trypanosoma cruzi interacting with AP-1 sequences.

Interaction between factors from Trypanosoma cruzi extracts and AP-1 sequences was studied by electrophoretic mobility shift assays. Using a double-stranded probe carrying the AP-1 sequence from the SV40 promoter, three specific complexes designated A, B, and C were detected. Complexes A and C were formed when using single-stranded probes. The relative amount of complex B, specific for double-stranded DNA, increased as a function of probe length. Complexes were stabilized by cross-linking with UVC irradiation and resolved on denaturing SDS-PAGE. Complex A generated bands of 60- and 39 kDa; complex B produced two bands of 46- and 43 kDa; and complex C generated one band of 43 kDa. The AP-1 binding activity was much higher in purified nuclear preparations than in soluble fractions, and was detected in crude extracts from the three forms of the parasite. The binding signal, however, was much stronger in amastigote and trypomastigote than in the epimastigote forms. Specific binding was increased by oxidative stress. Antibodies raised against peptides corresponding to conserved domains of mammalian c-Jun and c-Fos detected bands of 40- and 60 kDa, respectively, in a nuclear epimastigote preparation.

[Variability and correlations of missing and decayed teeth in Sicillian adolescent grouped by age and sex].

Overall and individual tooth morbidity were assessed in relation to mouth section, age and sex in 1,761 poor Sicilian adolescents aged 14 to 20 yr with decayed teeth or teeth extracted for decay. Statistical assessment of the findings in the upper and lower arch was undertaken in a search for related differences (increases or decreases) in the number of decayed or extracted teeth in each arch. The dependence on differences between one area and another on the variables considered was also examined in terms of their statistical significance. No sex-based difference emerged. The frequency of decay or extraction was usually a function of position. Both the upper and lower right or left 6th teeth (i.e. those in the middle position) were most commonly affected in males and females. Use was thus made of Vianelli's index in determination of the statistical asymmetry of teeth decayed or extracted on either side of the 6th.

[The effect of instrumentation on original apical foramen shape using steel vs niti rotatory: computerized analysis].

BACKGROUND: A fundamental requirement to obtain a correct endodontic preparation depends on the respect, during cleaning and shaping, of the original morphology of the apical foramen, (AF), so that the filling material will form a complete seal. METHODS: In our previous studies, in order to verify if this presumption was respected using rotary NiTi instruments with cutting tip, we instrumented extracted teeth characterised by a different degree of root curvatures. Using a standardized system to replace the sample, the original shape of the apical foramen of each tooth was recorded using a computerised technique and then compared to the shape after instrumentation. The data showed differences on AF shapes depending on the degree of root canal curvatures. In fact, the teeth with straight canals showed the least alterations on the original AF shape; on the contrary, the teeth with curved canals showed apical foramen enlargement or transportation. Therefore, in the current study we aimed to compare the in vitro effects of stainless steel, (Flexofile, Dentsply Maillefer, Baillaigues, Switzerland), vs NiTi safe cutting tip (Quantec SC, (Tycom Dental, Irvine CA, U:S.A.), vs NiTi non cutting tip, (Quantec LX, Tycom Dental, Irvine CA, U:S.A.), instrumentation on original apical foramen shape. We used NiTi instruments according to standard technique suggested by Tycom, and hand steel files, to instrument the apical third, according to the Crown-Down technique. Working length was fixed at -0.5 from AF. RESULTS AND CONCLUSIONS: Our results suggested that in the presence of accentuated canal curves rotary Niti, with cutting tip, cause significantly more enlargement of the AF area. At the same time, we also observed that NiTi rotary files, with both cutting and non-cutting tip, cause eccentric enlargement of AF in curved canals. Therefore, NiTi engine-driven instruments should be used carefully in the presence of accentuated canal curves to avoid enlargement or transportation of AF, probably because rotary NiTi files may slip out of operative control, leaving a mark on the foramen shape. In the presence of severe curves, we suggest modifying the operative sequences by alternating rotary NiTi with hand NiTi or stainless steel instruments, especially in the preparation of apical third.

[Request of dental care and periodontal treatment needs in a population sample of Palermo].

BACKGROUND: The organisation of periodontal health prevention programmes must take into account the public level of awareness of the periodontal diseases. The aim of the present investigation was to assess the relationships between the request of dental care and the periodontal treatment needs in a population sample of Palermo. METHODS: The study population consisted of 554 out-patients, consecutively examined during 18 months (January 1999 to June 2000) at the Palermo University Department of Oral Sciences. All participants answered a written questionnaire concerning the cause for the request of dental care and periodontal symptoms. Periodontal treatment needs were assessed using the Community Periodontal Index of Treatment Needs (CPITN). RESULTS: Statistical analysis (Chi-Square Test, Fisher's Exact Test) showed significant discrepancies between the request of dental care and the periodontal treatment needs, poor awareness of periodontal diseases and low perception of their symptoms by the patients. CONCLUSIONS: These findings showed that the awareness of periodontal diseases by the population examined is poor. This could be due either to the lack or failure of public education programmes on periodontal health. Yet, it could be related to an inadequate dentists' awareness of the periodontal diseases. Such findings should be considered in the planning of periodontal health programmes in relation to the population of Palermo.

Transbuccal tablets of carbamazepine: formulation, release and absorption pattern.

Tranbsuccal drug administration is an attractive method, as it has several advantages especially with respect to peroral delivery. Here we report: i) the aptitude of carbamazepine (CBZ) to penetrate porcine buccal mucosa and reconstituted human oral (RHO) epithelium; ii) three different tablet formulations for transbuccal administration; iii) the drug release rate from tablets. CBZ permeation through the buccal mucosa was investigated by using two different bi-compartmental open models: Franz cells for porcine buccal mucosa and Transwell diffusion cells system for RHO epithelium. Results, expressed as drug flux (Js) and permeability coefficients (Kp), indicated that CBZ well penetrates the membrane and arrives in the acceptor phase. Js and Kp resulted 7x10(-2) mg/cm2h and 0.23 cm/h for in vitro experiments and 1.81 x 10(-2) mg/cm2h and 4.57 x 10(-2) cm/h for ex vivo experiments. The flux is extensively affected by the membrane thickness. The CBZ release from three different formulations of tablets, prepared with loaded microspheres, loaded matrices, and conventional compressed physical mixture of components was studied. Using the new formulated "non-conventional" tablets prolonged drug release was obtained. Loaded matrix tablets discharged CBZ faster than microsphere tablets (17% and 12% in about 2.5 h respectively). Results indicate the possibility of administering CBZ on buccal mucosa.

[S.E.M. examination of different grades of curette wear].

After succeeding working trials, a wear test was conducted to evaluate different curettes, by using S.E.M. analysis and replica technique. The obtained results underline the importance of structural characteristics of curettes to maintain standard rates of cutting effectiveness.

[Effect on periodontal tissue of a "cushioned" bristle brush. (S.E.M. analysis)].

The Authors have made in vivo verification by means of replica technique observed with SEM any possible traumatic effects and gingival modification which a bristle toothbrush (Sanogyl, Berna) has on the soft tissues. The toothbrush has particular features which make it possible to perform an almost perfect dental hygiene of the papillar area, it does not produce any secondary negative effects on the gingival soft tissues.

[So-called Pindborg's tumor: calcifying odontogenic epithelial tumor].

A case of Pindborg's tumour or calcifying odontogenous epithelial tumour in the included +3 of a young girl is presented. The histogenetic explanation given by the first workers to the earliest cases--origin in residues of the enamel organic matrix--is accepted as the most probable, though it is pointed out that the small number of reported cases make doubt and uncertainty inevitable. An interesting point about the case is that its clinical course was followed for several years after surgery.
