Characterization of [35S]lanthionine ketimine specific binding to bovine brain membranes.

[35S]Lanthionine ketimine binds specifically and with high affinity to bovine brain membranes. This binding has been studied in detail. It is reversible, not occurring at an uptake site or at a metabolizing enzyme and depending only weakly from ionic strength; it is affected by thiol reagents. [35S]Lanthionine ketimine specific binding is displaced only by other ketimines and by catecholamines, but not by more selective adrenergic ligands; binding parameters are reported. [3H]Adrenaline but not [3H]dihydroalprenolol is partially displaced by lanthionine ketimine. With bovine brain preparations a significant stimulation of basal adenylate cyclase activity by lanthionine ketimine is observed.

RNA binding properties and evolutionary conservation of the Xenopus multifinger protein Xfin.

Xfin is a Xenopus zinc finger protein which is expressed in the cytoplasm of the oocyte and throughout embryogenesis, as well as in the cytoplasm of some specific and highly differentiated cell types (De Lucchini et al., Mech. Dev. 36, 31-40, 1991). In this paper we present a characterization of some structural features of the protein and of its nucleic acid binding properties. We found that Xfin is a phosphoprotein, is present in the soluble fraction of the cytoplasm, and is actively phosphorylated in cytosolic extracts. Several putative phosphorylation sites are present in the cDNA-derived protein sequence, mostly located at specific positions within the Zn-fingers. In an in vitro assay a fusion protein containing part of the finger region of Xfin exhibits specific binding to a poly (G) RNA homopolymer, while it does not bind DNA. The RNA binding activity of the protein is significantly enhanced by phosphorylation. A putative Xfin homolog, which appears to be evolutionarily conserved with regard to size, cytoplasmic expression and antigenic specificity, is present in representatives of five Vertebrate classes. Taken together, these results may suggest that, by virtue of its RNA binding activity modulated through phosphorylation, Xfin could serve some evolutionarily conserved function in post-transcriptional regulation processes.

NADPH-diaphorase reactivity in nerves supplying the rat anococcygeus muscle.

NADPH-diaphorase histochemistry was used to establish the nature of neurons and nerve fibers in the rat anococcygeus muscle. NADPH-diaphorase-reactive nerve cell bodies were found in ganglia in the caudal dorsal fascia of the muscle and within the muscle itself. The reactive nerve cells had a relatively smooth surface and a single axon. They formed bundles of positive nerve fibers which entered the muscle and branched further to form a network extending towards the cranial end. In addition, bundles of NADPH-diaphorase-reactive fibers of extrinsic origin joined the loose plexus of local bundles and microganglia. The NADPH-diaphorase-reactive neurons supplying the rat anococcygeus muscle provide a neuroanatomical basis for the non-adrenergic non-cholinergic relaxation of the muscle mediated by nitric oxide.

DNA-protein cross-links in welders: molecular implications.

A new method for detecting DNA-protein cross-links involving selective precipitation of DNA containing cross-linked proteins by K(+)-sodium dodecyl sulfate was utilized in the peripheral WBC of 21 male metal arc welders and in 26 male controls of similar age and racial characteristics who were not exposed to welding fumes. DNA was quantitated by Hoechst fluorescence. Although the concentration of nickel and chromium in the peripheral blood did not differ between subjects in the two groups, one-fourth of the welders had levels of DNA-protein cross-links that were above the upper limit of the controls. Mean cross-link values were 1.85 +/- 1.14% (SD) among the welders and 1.17 +/- 0.46% among the controls, a 58% statistically significant difference (P = 0.01). Thus, many welders appeared to be burdened with an excess of DNA-protein cross-links, suggesting exposure to cross-linking agents and, possibly, a detectable biological effect of potential genotoxic consequences.

Nickel induces increased oxidants in intact cultured mammalian cells as detected by dichlorofluorescein fluorescence.

Exposure of intact cultured Chinese hamster ovary (CHO) cells to water-soluble nickel (Ni) salts and to relatively water-insoluble crystalline nickel subsulfide (Ni3S2) resulted in an increased formation of the fluorescent oxidized compound, dichlorofluorescein (DCF) from the parent nonfluorescent compound, 2,7-dichlorofluorescin diacetate. This fluorescent product was also formed in vitro following oxidation with relatively strong oxidants such as hydrogen peroxide in the presence of peroxidase, suggesting that Ni increased the concentration of hydrogen peroxide in intact cells. However, formation of other strong oxidants such as hydroperoxides is possible since they have also been shown to cause the oxidation of the nonfluorescent dichlorofluorescin to the fluorescent product DCF in vitro. Localization of the oxidized fluorescent DCF in intact cells was also examined by fluorescence microscopy. Both Ni3S2 and NiCl2 appeared to increase the degree of fluorescence in intact CHO cells around the nuclear membranes. This increase in fluorescence was greater in the presence of relatively water-insoluble Ni3S2 than water-soluble NiCl2. These results add to the emerging concept that Ni-induced genotoxicity may be mediated by oxygen radical intermediates.

Structure of the tertiary component of the myenteric plexus in the guinea-pig small intestine.

The tertiary component of the myenteric plexus consists of interlacing fine nerve fibre bundles that run between its principal ganglia and connecting nerve strands. It was revealed by zinc iodide-osmium impregnation and substance P immunohistochemistry at the light-microscope level. The plexus was situated against the inner face of the longitudinal muscle and was present along the length of the small intestine at a density that did not vary markedly from proximal to distal. Nerve bundles did not appear to be present in the longitudinal muscle as judged by light microscopy, although numerous fibre bundles were encountered within the circular muscle layer. At the ultrastructural level, nerve fibre bundles of the tertiary plexus were found in grooves formed by the innermost layer of longitudinal smooth muscle cells. In the distal parts of the small intestine, some of these nerve fibre bundles occasionally penetrated the longitudinal muscle coat. Vesiculated profiles in nerve fibre bundles of the tertiary plexus contained variable proportions of small clear and large granular vesicles; they often approached to within 50-200 nm of the longitudinal smooth muscle cells. Fibroblast-like cells lay between strands of the tertiary plexus and the circular muscle but were never intercalated between nerve fibre varicosities and the longitudinal muscle. These anatomical relationships are consistent with the tertiary plexus being the major site of neurotransmission to the longitudinal muscle of the guinea-pig small intestine.

Development and utilization of a new simple assay for DNA-protein crosslinks as a biomarker of exposure to welding fumes.

Stereological and morphometric analysis of dermal fibroblasts before and after bone marrow transplantation in a case of mucopolysaccharidosis I Scheie phenotype.

Bone marrow transplantation (BMT) has been used therapeutically in several types of mucopolysaccharidoses (MPS) and other inherited metabolic disorders. Fibroblasts are severely affected in MPS due to the intralysosomal accumulation of glycosaminoglycans. We report a stereological and morphometric study at light and electron microscopy levels of dermal fibroblasts before and 21 months after BMT in a young girl with MPS I Scheie phenotype (MPS I-S). Dermal fibroblasts showed remarkable morphological changes although their density, expressed as number of fibroblasts per unit volume of dermis (number density), was not modified in the post-BMT samples as compared to pre-BMT ones. Stereological and morphometric parameters referring to cell characteristics of post-BMT fibroblasts (nuclear and cell surface densities, and both nucleus/cell and cell/nucleus volume densities) showed significant differences when compared with pre-BMT fibroblasts, and non-significant differences regarding control cells. On the other hand, quantitative parameters of the lysosomal compartment from post-BMT fibroblasts showed intermediate values between pre-BMT and control fibroblasts. These results, at cellular level, are in agreement with previous biochemical and clinical results, and clearly showed a progressive course to a non-pathological state. All parameters estimated may be considered useful tools in evaluating the success of BMT. These parameters provide quantitative data which can be statistically compared, showing the changes due to the reduction of storage material after BMT. Cell/nucleus volume density is especially interesting since not only is it easy to estimate, even by automatic procedures, but it could also constitute a numerical expression of skin anatomopathological analyses performed post-BMT.

Macrolide antibiotics as antiinflammatory agents: roxithromycin in an unexpected role.

The antiinflammatory activity of a new 14-membered macrolide antibiotic, roxithromycin, was evaluated in various rat models including carrageenan- and poly-L-arginine-induced hind-paw oedema, croton oil inflamed ear assay and polyester sponge granuloma. When administered orally to animals, roxithromycin displayed an atypical profile in the assays utilized, including: (1) marked antioedema activity similar to that of indomethacin in poly-L-arginine assay, (2) significant inhibition of lambda-carrageenan hind-paw oedema and croton-oil-induced inflammation in the ear, although indomethacin was more effective, and (3) failure to reduce the development of granuloma induced by implanted polyester sponges, while indomethacin significantly reduced the chronic inflammatory reaction. Based on these results, it is concluded that roxithromycin is active in reducing the acute inflammatory reaction in rat models through mechanisms different from conventional nonsteroidal antiinflammatory agents such as indomethacin. Therefore, roxithromycin may have a favorable impact on skin inflammatory reactions accompanying microbial infections.

Psychosocial factors and clinical evolution in HIV-1 infection: a longitudinal study.

The aim of the study was to assess the relationship between the initial psychosocial situation and the probability of later symptom development in HIV-1 infection. One hundred HIV-1 seropositive subjects, 79 in Stage III (LAS) and 21 in Stage II (asymptomatic), were examined both immunologically (CD4+, Skin Test) and psychologically (test battery). Follow-up at 6 and 12 months involved clinical and immunological reassessment of subjects, who were then classified as fully symptomatic (S, Stage IV) or unchanged (U). The two groups were compared through ANOVA on initial psychosocial measures, while stepwise logistic multiple regression was employed to assess the predictive value of psychosocial measures on clinical and immunological evolution. Psychosocial measures most clearly showing an association with clinical evolution were Denial/Repression attitudes (negatively) and Fighting Spirit (positively), whereas aspects of Hardiness and Social Support showed an effect in interaction with initial CD4+ levels. No stable results were obtained on immunologic evolution. The two groups (U and S) did not show significant differences on other independent variables, with the exception of age.

Mutagenic responses of nickel oxides and nickel sulfides in Chinese hamster V79 cell lines at the xanthine-guanine phosphoribosyl transferase locus.

Mutagenesis of several insoluble nickel compounds--crystalline nickel sulfide NiS, nickel subsulfide Ni3S2, nickel oxides (black and green) and soluble NiCl2 was studied in three Chinese hamster cell lines--at the hprt gene of the well-defined V79 cell line, and at gpt in two transgenic derivative cell lines G12 and G10. The transgenic cell line G12 responded very strongly to the insoluble Ni compounds, such that the gpt mutagenesis was at least 20 times higher than the spontaneous mutagenesis and in some experiments was even higher. In contrast the response of the G10 cells was much lower--the mutant frequencies only increased 2-3 times over the controls. In V79 cells, NiS and NiO (black) did not induce a mutagenic response at hprt. Soluble NiCl2 also exhibited no mutagenic activity in V79 cells and induced considerably lower activity than the insoluble compounds in the transgenic G12 cells. Following vitamin E pretreatment of G12 cells for 24 h prior to nickel exposure, increased cell survival was observed for several insoluble Ni compounds whereas vitamin E had no effect on NiCl2 cytotoxicity. With vitamin E pretreatment, significantly lower mutagenic responses in G12 cells were also noted for some insoluble Ni compounds, while no such effect was observed for NiCl2.

The effects of pertussis toxin on vasoconstrictor and vasodilator agents in the pithed rat may not be an indicator of G-protein receptor coupling.

1. In the present study, rats were treated with pertussis toxin (8.4 micrograms, i.v.) to determine whether pertussis toxin-sensitive G-proteins were linked to receptors that mediate effects in the cardiovascular system. 2. In the pithed rat the pressor responses to the alpha-adrenoceptor agonists noradrenaline and (to a lesser extent) methoxamine were attenuated by pertussis toxin treatment; the tachycardia produced by noradrenaline was unaffected. The pressor response to serotonin was also attenuated by pertussis toxin treatment. These observations are consistent with known effects of pertussis toxin on these receptors. 3. The vasodepressor responses to the muscarinic receptor agonist carbachol and to adenosine were reduced by pertussis toxin, suggesting that these events may have been mediated by pertussis toxin-sensitive G-proteins. However, the depressor responses to the beta 2-adrenoceptor agonist salbutamol and to the receptor-independent vasodilator drugs sodium nitroprusside and hydralazine were also reduced by pertussis toxin. These latter effects suggest that caution must be used in interpreting the effects of pertussis toxin since the mechanism of action of these drugs as elucidated in vitro is thought not to involve pertussis toxin-sensitive G-proteins.

Evidence that M1 muscarinic receptors enhance noradrenaline release in mouse atria by activating protein kinase C.

1. The M1 selective muscarinic agonist, McNeil A 343, enhanced the electrically evoked release of noradrenaline from postganglionic sympathetic nerves in mouse atria. This has been found previously to be due to activation of muscarinic receptors of the M1 subtype, probably located on sympathetic nerve terminals. The present study investigated the signal transduction mechanisms involved in the release-enhancing effects of McNeil A 343. The release of noradrenaline from mouse atria was assessed by measuring the electrically-induced (3 Hz, 60 s) outflow of radioactivity from atria which had been pre-incubated with [3H]-noradrenaline. 2. 8-Bromo cyclic AMP in the presence of IBMX was used to enhance maximally S-I noradrenaline release through cyclic AMP-dependent mechanisms. However, the facilitatory effect of McNeil A 343 (10 microM) was not different from the effect in the absence of these drugs, suggesting that McNeil A 343 enhances noradrenaline release independently of the cyclic AMP system. Furthermore, the release-enhancing effect of McNeil A 343 (10 microM) on noradrenaline release was also not altered by the 5-lipoxygenase inhibitor, BW A4C. 3. The facilitatory effect of McNeil A 343 was not altered in the presence of drugs (trifluoperazine, W7, and calmidazolium) which inhibit calmodulin-dependent processes, suggesting that the mechanisms of action of McNeil A 343 does not depend on calmodulin. 4. It was considered likely that the facilitatory effect of McNeil A 343 on noradrenaline release may be due to activation of protein kinase C, since activators of protein kinase C enhance noradrenaline release. The facilitatory effect of McNeil A 343 was abolished by the non-selective protein kinase C inhibitor,K-252a. To investigate further the involvement of protein kinase C, mouse atria were chronically incubated (9-O h) with the protein kinase C activator, 4 beta-phorbol dibutyrate (1.0 microM) in order to down-regulate protein kinase C activity. In protein kinase C-down-regulated atria, the facilitatory effect of McNeil A 343 (30 microM) was abolished. Incubation with 4 alpha-phorbol dibutyrate which does not affect protein kinase C did not reduce the facilitatory effect of McNeil A 343. This provides evidence that activation of protein kinase C is involved in the signal transduction process of McNeil A 343.

Preoperative irradiation and fluorouracil chemotherapy for locally advanced rectosigmoid carcinoma: phase I-II study.

From June 1988 to July 1991, 20 patients with locally advanced rectal or rectosigmoid cancer were treated prospectively with a strategy of combining preoperative irradiation and fluorouracil chemotherapy before surgical resection. The preoperative radiation dose was 5,000 cGy, and fluorouracil chemotherapy was administered on the first and last 3 days of irradiation in an intravenous bolus dose of 500 mg/m2. In a median follow-up of 25 months, the local regional failure rate was 10%. The 3-year actuarial overall survival and disease-free survival were 92% and 82%, respectively. Twenty percent of the surgical specimens showed no residual tumor, and only 10% showed positive lymph nodes. Significant leukopenia occurred in 10% of patients. Preoperative irradiation and fluorouracil chemotherapy increased surgical downstaging and improved local regional control. The overall toxicity was acceptable. The results of this particular multimodality approach was encouraging and warrant further investigation in phase III trials.

Effects of occupational status on the mental health of young Italian men and women. A cross-sectional study.

The study was carried out on 548 subjects (307 women, 244 men) between the ages of 21 and 26, who had all obtained secondary school certificates. The survey was conducted in two contrasting areas, one showing a high rate of unemployment, the other a rate below the national average. The sample was divided into three groups according to occupational status: employed persons, students and persons seeking their first employment. The subjects were given five questionnaires: a demographic form, the General Health Questionnaire, the Beck Depression Inventory, the Beck Hopelessness Scale and the Rosenberg Self-Esteem Scale. Our findings revealed that unemployed women experience greater psychological distress while men are more subject to marked depressive states. There were no significant differences between the two areas surveyed.

Fish oil derivatives as a prophylaxis of recurrent miscarriage associated with antiphospholipid antibodies (APL): a pilot study.

Since 1989, 22 patients with persistent antiphospholipid syndrome (PAPS) associated with recurrent miscarriage (defined as three or more miscarriages) were treated with fish oil, equivalent to 5.1 g eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) at a ratio of 1.5 EPA to DHA. Twenty-two patients had 23 pregnancies (one patient had two pregnancies) over a period of 3 years. There was only one intrauterine fetal death at the 27th week associated with pre-eclampsia. Twenty-one pregnancies, 19 of which ended after the 37th week, produced a baby. Two pregnancies ended with cesarean section for pre-eclampsia at 30th and 35th week of gestation and one is ongoing at 32nd week. All babies are well. The weight at birth of babies delivered at term was always > 2500 g. These encouraging results favour a therapeutic role, without any adverse reaction, of fish oil to prevent recurrent miscarriage in PAPS.

Mechanisms of chromium carcinogenicity and toxicity.

Chromium, like many transition metal elements, is essential to life at low concentrations yet toxic to many systems at higher concentrations. In addition to the overt symptoms of acute chromium toxicity, delayed manifestations of chromium exposure become apparent by subsequent increases in the incidence of various human cancers. Chromium is widely used in numerous industrial processes, and as a result is a contaminant of many environmental systems. Chromium, in its myriad chemical forms and oxidation states, has been well studied in terms of its general chemistry and its interactions with biological molecules. However, the precise mechanisms by which chromium is both an essential metal and a carcinogen are not yet fully clear. The following review does not seek to embellish upon the proposed mechanisms of the toxic and carcinogenic actions of chromium, but rather provides a comprehensive review of these theories. The chemical nature of chromium compounds and how these properties impact upon the interactions of chromium with cellular and genetic targets, including animal and human hosts, are discussed.
