The lower susceptibility to Plasmodium falciparum malaria of Fulani of Burkina Faso (west Africa) is associated with low frequencies of classic malaria-resistance genes.

The gene frequencies in 1993-94 for haemoglobin S, haemoglobin C, alpha-3.7 deletional thalassaemia, G6PDA-, HLAB*5301 were estimated in Fulani, Mossi and Rimaibé ethnic groups of Burkina Faso, West Africa. The aim of the study was to verify whether the previously reported Fulani lower susceptibility to Plasmodium falciparum malaria was associated with any of these malaria-resistance genes. Similar frequencies for haemoglobin S were recorded in the 3 ethnic groups (0.024 +/- 0.008, 0.030 +/- 0.011, 0.022 +/- 0.013; in Mossi, Rimaibé and Fulani, respectively). The Mossi and Rimaibé showed higher frequencies when compared to Fulani for haemoglobin C (0.117 +/- 0.018, 0.127 +/- 0.020, 0.059 +/- 0.020), alpha-3.7 deletional thalassaemia (0.227 +/- 0.040, 0.134 +/- 0.032, 0.103 +/- 0.028), G6PDA- (0.196 +/- 0.025, 0.187 +/- 0.044, 0.069 +/- 0.025) and HLA B*5301 (0.189 +/- 0.038, 0.202 +/- 0.041, 0.061 +/- 0.024). Among Fulani the proportion of individuals not having any of these protective alleles was more than 3-fold greater than in the Mossi-Rimaibé group (56.8% vs 16.7%; P < 0.001). These findings exclude the involvement of these genetic factors of resistance to P. falciparum in the lower susceptibility to malaria of Fulani. This evidence, in association with the previously reported higher immune reactivity to malaria of Fulani, further supports the existence in this ethnic group of unknown genetic factor(s) of resistance to malaria probably involved in the regulation of humoral immune responses.

Bionomical and cytogenetic heterogeneities of Anopheles funestus in Senegal.

Malaria transmission by Anopheles funestus was investigated from May 1994 to September 1997 in different locations from western to eastern Senegal along the northern border of The Gambia. 10515 A. funestus were captured on human volunteers or by indoor pyrethrum spraying. Circumsporozoite protein rates showed that A. funestus had a high infection rate, 2-7%, in the whole of the study area. Analysis of feeding behaviour showed great variation of anthropophilic rates from western Senegal, where populations were highly anthropophilic, to eastern Senegal, where they were much more zoophilic. In eastern Senegal many females captured in bedrooms had fed outside on horses. Polytene chromosome analysis showed that the general pattern of karyotype distribution is consistent with the hypothesis of 3 chromosomally differentiated populations of A. funestus. In samples from a central part of the study area, analysis showed lack of karyotype intergradation with a deficit of heterokaryotypes, suggesting the presence of 2 genetically differentiated populations in an area of sympatric.

[Effect of the photoperiod of the site of oviposition of Aedes mariae (Diptera,Culicidae)].

The overwintering biology of Aedes (Ochlerotatus) mariae (one of the sibling species of the mariae complex) was studied in populations from the Tyrrhenian coast of Central Italy (Sperlonga and Formia). Ae. mariae has in this zone several generations per year and its larvae are commonly found from March to October exclusively in rock pools along the coast. Field and laboratory observations show in these populations an embryonic winter diapause induced by short-day photoperiods (9-12 hours). The photoperiod acts primarily on the parental females during their preimaginal life. The expression of diapause is influenced by temperatures and photoperiods experienced by the embryos. Full embryonic diapause is observed when the eggs are incubated at relatively low temperatures (less than 16 degrees C) and at short photoperiods. Short-day photoperiods also induce a remarkable change in the oviposition behavior of Ae. mariae. Laboratory observations show that the adult females readily oviposit on water surfaces when originating from larvae reared at long-day photoperiod while they are very reclutant to oviposit in the same situation when reared at short day photoperiod. Choice experiments involving four alternative oviposition sites (see Plate 1) demonstrate a preference for outside free water in long-day mosquitoes and for inside moist surfaces in short-day mosquitoes. The behavioral difference persists in successive gonotrophic cycles and it is not apparently affected by the photoperiod acting on the adult females. The above evidence together with recovery of overwintering eggs in holes and crevices of rock pools suggest that the shift in oviposition site shown in the laboratory reflects a similar shift occurring in nature. Such photoperiodically induced change in oviposition behavior seems to have an important adaptive significance in providing more constant microclimates to the diapausing eggs and in protecting them from the mechanical action of winter storms.

[Propensity for feeding on blood under laboratory conditions and chromosomal polymorphism in Anopheles stephensi].

Propensity to blood feeding under laboratory conditions was studied in females of A. stephensi carriers of different arrangements of a polymorphic paracentric inversion of chromosome 2R (+/+, +/b, b/b). One hour experiments were performed at various hours of the day, in continuous light, with temperature between 26 and 28 degrees C and relative humidity between 65 and 75%, using unfed mosquitoes 4-5 days old and guinea pigs as hosts. The frequency of blood fed females was found to be constantly much lower in the +/+ homokaryotypes, than in the other two karyotypes. Smaller differences in biting activity were observed between the heterokaryotypes and the b/b homokaryotypes, the latter appearing significantly more active at least during the scotophase hours. The evidence of a lower activity of the +/+ homokaryotypes was confirmed when the mosquitoes were stimulated by a human arm. Such marked and constant behavioural difference is tentatively considered as the expression of a diversified response to the environmental conditions whose deterrent effect on biting activity would be more pronounced on the +/+ homokaryotypes than on the carriers of the 2Rb arrangement.

[Seasonal variations in microfilarema and effects of ambient temperature in dogs parasitized by Dirofilaria repens].

1 - Dogs infected with Dirofilaria repens from Italy were found to show seasonal variations in the number of microfilariae in the peripheral blood. 2 - The rise in the number of microfilariae occured in August and September and is coincidental with the higher probability of trasmission. 3 - Rise and fall of microfilaraemia was induced experimentally by exposure of the host to high and low temperatures respectively. 4 - The importance of such variations in microfilaraemia is stressed in relation to the evaluation of the infection from a clinical, pharmacological and epizoological point of view.

Immunity to Plasmodium sporozoites: recent advances and applications to field research.

The presence of antibodies against Plasmodium falciparum sporozoites in humans living in malaria endemic areas was measured using as antigen the synthetic peptide (NANP)3, which represents the immunodominant region of the circumsporozoite (CS) protein. The results indicate that: i) the production of anti-CS antibodies is unrelated to the presence in the circulation of blood-stage parasites; ii) anti-CS antibodies, raised by natural inoculation, could exert a protective role against natural malaria infection; iii) anti-CS antibodies can be used as indicators of the intensity of malaria transmission.

Ivermectin against larval stages of Dirofilaria repens in dogs.

The prophylactic efficacy of ivermectin against Dirofilaria repens infections in dogs was investigated. A first trial was carried out on 15 dogs exposed to four inoculations of L3 larvae at 15-day intervals and treated, in groups of five, with 0, 6 or 12 micrograms/kg body weight of ivermectin given per os 30 and 60 days after the first inoculation. Necropsy, performed about 9 months later, revealed that worm burdens were reduced by 86.6 and 92.8% for the 6 and 12 micrograms/kg dose levels, respectively. In a second trial with an otherwise identical protocol, a dose rate of 24 micrograms/kg of ivermectin was tested in 12 dogs. Only one of the six treated dogs was found worm free at necropsy. The worm burden was reduced by 87.9% in treated animals as opposed to controls. A lengthening of the prepatent periods, which might be considered dose related, was apparent in all treated groups. Ivermectin was not completely effective in preventing establishment of experimental infections with D. repens in dogs.

Genetic characterization of Dirofilaria repens and D. immitis by electrophoretic analysis of gene-enzyme systems.

A multilocus genetic analysis based on protein electrophoresis was carried out on 39 adult specimens of Dirofilaria repens and 31 of D. immitis. Seventeen enzymatic loci provided reliable electrophoretic patterns which were compared in the two species. All loci except one were monomorphic. Fixed alternative allozymes were found at 13 loci while only 3 loci shared apparently the same allozyme in the two species. The polymorphic locus, Pgm, also showed alternative allozymes. This remarkably high genetic divergence, which presumably reflects a very old speciation process, allows an easy characterization of D. repens and D. immitis and supports their classification in the subgenera Nochtiella and Dirofilaria, respectively.

CS antigen localization in malaria vectors: hypothetical refractoriness to transmission observed in the field.

Indoor resting Anopheles gambiae s.l. were collected in two villages near Ouagadougou, Burkina Faso, and processed to investigate the presence and distribution of Plasmodium sporozoites. Salivary glands were dissected, examined by phase contrast microscopy and further processed by IRMA in order to reveal the presence of the circumsporozoite (CS) antigen of P. falciparum. The corresponding thoraces were homogenized and processed by IRMA. In the village characterized by the higher inoculation rate more than 40% of the infected mosquitoes were not found infective since CS antigen was detected in the thorax in absence of sporozoites and CS antigen in the corresponding salivary glands.
