Variance analysis of error in selected ion monitoring assays using various internal standards. A practical study case.

This study contains a practical application of variance analysis for the evaluation of precision in selected ion monitoring using stable isotope variants or chemically related compounds as internal standards. Variance analysis is applied to determine the relative contributions of the errors due to long-term and short-term effects of instrument stability. The technique allows the resolution of variance due to sample manipulation from that introduced by the gas chromatography mass spectrometry instrumentation. This provides an objective measure of potential internal standards. Evaluation of the dependency of these errors upon concentration indicates appropriate weighing factors to be applied for regression analysis.

Simultaneous measurement of imipramine and desipramine by selected ion recording with deuterated internal standards.

A gas chromatographic mass spectrometric method for the quantitative determination of imipramine and its N-demethylated metabolite desipramine in plasma samples at the nanogram level is reported. The method involves derivatization of the extracted drugs with trifluoroacetylimidazole, a mild derivatizing reagent. Specificity is provided by selected ion recording of the [M + H]+ ions, formed upon chemical ionization with methane as reagent gas. Quantitation is achieved by stable isotope dilution techniques, using deuterium labeled analogs, prepared by acid-catalyzed exchange, as internal standards. Data on patient samples are presented.

Antiplatelet and antithrombogenic effects of suloctidil.

Platelet aggregation induced in mice or rats by i.v. ADP can be antagonized by oral administration of suloctidil. This effect on platelet behaviour appears to be sufficient to reduce the rate of thrombotic occlusion of the femoral artery in the dog and to protect the rat against occurrence of thrombophlebitis.

Mass spectrometric methods for the characterisation and differentiation of isomeric O-diglycosyl flavonoids.

Tandem mass spectrometric methods have been evaluated for the characterisation of the type and the differentiation of the interglycosidic linkage of isomeric flavonoid O-diglycosides. Based on the occurrence of internal monosaccharide residue loss and the relative abundances of Y-type ions formed by fragmentation at glycosidic bonds, four pairs of isomeric flavonoid O-diglycosides can be unambiguously differentiated. The different techniques used, i.e. linked scanning at constant B/E without collisional activation and low-energy collision-induced dissociation using methane or helium as collision gas, have been shown to be useful for distinguishing the two most common (1, 2- and 1, 6-) interglycosidic linkages, e.g. flavonoid O-neohesperidosides and O-rutinosides.

Structure and chemotherapeutical activity of a polyisoprenylated benzophenone from the stem bark of Garcinia huillensis.

The stem bark of Garcinia huillensis grown in Zaïre and used in central-African traditional medicine has been subjected to a bioassay-guided fractionation. The chemotherapeutically active petroleum ether extract afforded fatty acids, aliphatic alcohols, triterpenes and a polyisoprenylated benzophenone, which was identified as garcinol, also named camboginol. This compound has been shown to exhibit chemotherapeutical activity gram-positive and gram-negative cocci, mycobacteria and fungi. On the other hand garcinol has been found to be inactive against gram-negative enteric bacilli, yeasts and viruses.

The POP Program: the patient education advantage.

In 1992, a preoperative education program was developed for total joint replacement patients in a small community hospital. The goals of the program were to increase educational opportunities for the joint replacement patients, prepare patients for hospitalization, plan for discharge needs, and increase efficiency of the orthopaedic program. Since 1992, approximately 600 patients have attended the education program. Outcomes have included positive responses from patients regarding their preparedness for surgery, increased participation in their plan of care, coordinated discharge planning, decreased length of stay, and progression across the continuum of care. A multidisciplinary approach to preparing patients for surgery allows for a comprehensive and efficient education program. Marketing of successful programs can enhance an institution's competitive advantage and help ensure the hospital's viability in the current health care arena.
