Diminished calcium homeostasis and increased susceptibility to excitotoxicity of JS 3/16 progenitor cells after differentiation to oligodendroglia.

JS 3/16, derived from passaged oligodendroglial cultures prepared from rat cerebral white matter, differentiate from progenitors (OP) into complex process-bearing, galactocerebroside-positive but myelin basic protein-negative immature oligodendrocyte-like cells (ImO) after withdrawal of trophic factors. We found that JS 3/16 ImO are markedly more susceptible than OP to cell death after sustained alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionate glutamate receptor (AMPA-GluR) activation. This excitotoxicity is preceded by loss of intracellular Ca(2+) homeostasis, which is more marked in ImO than OP. We identified three factors likely to contribute to the diminished Ca(2+) homeostatic capacity of ImO. First, signal intensities of immunoreactive GluR2, GluR3, and GluR4 AMPA-GluR subunits are increased 1.3- to 2.2-fold in ImO over OP without comparable changes in RNA editing and alternative splicing. Second, transcriptional levels of genes encoding Na(+)-Ca(2+) exchanger proteins and a plasma membrane ATPase (PMCA1), which are necessary for Ca(2+) extrusion across the plasma membrane, are lower in ImO than in OP. Third, ImO have more depolarized basal mitochondrial membrane potential (Delta Psi) than OP, and Delta Psi collapses within 15 min after onset of AMPA-GluR activation in almost all ImO, but not in the majority of OP. This Delta Psi collapse limits the capacity of ImO mitochondria to buffer the rise in intracellular Ca(2+) caused by AMPA-GluR activation. The JS 3/16 line provides a valuable system for analysis of intracellular Ca(2+) homeostasis and AMPA-GluR-mediated excitotoxicity in the oligodendroglial lineage.

Age- and myopathy-dependent changes in connexins of normal and cardiomyopathic Syrian hamster ventricular myocardium.

The conduction of cardiac action potentials depends on the flow of excitation through gap junctions, which are hexameric protein associations of connexins (Cxs). The major Cx reported in the heart is Cx43, although some Cx40 and Cx45 are also present. There is some evidence for altered Cx content in heart failure. In heart failure, conduction is depressed and slowed conduction may contribute to arrhythmogenesis and (or) the maintenance of arrhythmia. Cx content and distribution were determined in ventricular tissues from normal and cardiomyopathic Syrian hamsters, an animal model of heart failure which has reproducible age-specific cardiomyopathy resulting in heart failure and age-matched controls in three groups: young (3-5 weeks), adult (13-18 weeks), and old (>45 weeks). Frozen, unfixed sections of ventricular tissues were immunofluorescently stained using antibodies against Cx43, Cx40, and Cx45. Cx43 was the predominant Cx detected in all samples. In normal hamsters, Cx43 was localized predominantly at the intercalated disc region, while in myopathic myocytes, it was scattered. In Western blots, Cx43 content of normal hamster hearts was highest in the adult hearts compared with young and old hamster hearts. In contrast, Cx43 content was significantly lower in adult cardiomyopathic hamster hearts compared with all other groups. The alterations of content and distribution of gap junction Cx43 may contribute to diminished conduction, pump function, and arrhythmogenesis in heart failure.

Humoral and cellular immune responses to the GB virus C/hepatitis G virus envelope 2 protein.

Immune responses to two recombinant envelope 2 (E2) proteins, representing genotypes 1 and 2 of the GB virus C, or the hepatitis G virus (GBV-C/HGV), were studied in mice and in 48 individuals with, or without, chronic, or past GBV-C/HGV infection. Immunised mice developed E2-specific antibodies (mean titres, 1:1,167 to 1:9,360), recognising linear antigenic regions and proliferative and IL-2, IL-6 and gammaIFN cytokine responses regardless of the viral genotype. Individuals with past GBV-C infection had E2 antibody titres from 1:1,500 to 1:7,500 that did not recognise the E. coli derived E2 protein or linear antigenic regions. Proliferative E2-specific responses were detected in peripheral blood mononuclear cells from 6/22 (27%) persons with, and in none without GBV-C markers (P<0.05). Thus, E2-specific immune responses are mainly crossreactive between different variants of GBV-C/HGV, although proliferative responses appear to be rare.

Tuboovarian abscess mimicking malignancy: report of two cases.

Tuboovarian abscess is a well-known sequela of acute or chronic salpingitis. In a small percentage of patients, these inflammatory masses compress or even rupture into the adjacent viscera, thus simulating the condition of pelvic malignancy, particularly when the clinical presentations are indolent. We describe two cases of tuboovarian abscess mimicking malignancy. Case 1: A 39-year-old woman with an intrauterine device had a clinical presentation mimicking an exophytic submucosal colorectal tumor with suspicious mucosal invasion. She complained of tenesmus but did not experience fever or adnexal tenderness. A right tuboovarian abscess with fistula formation into the rectosigmoid colon was noted during laparotomy. Case 2: A 46-year-old woman with an intrauterine device had a preoperative diagnosis of uterine myoma with degeneration. At laparotomy, an omentum cake with dense pelvic adhesions was noted. Malignancy appeared to be present, and debulking surgery was performed. The final pathologic examination revealed bilateral chronic tuboovarian abscesses and focal omental abscess.

[Effects of stress level on the biomechanical behavior of the temporomandibular joint disc in domestic pigs].

This study aimed to examine the responses of the temporomandibular joint (TMJ) disc against compression, to clarify the property of viscoelasticity and permeability of the disc, and to provide experimental data for further analyzing the mechanism of stress dissipation within the TMJ disc. With the use of confined compression mechanics and biphasic theory, creep experiments on eight TMJ discs of four domestic pigs were performed under three stress levels(0.07 MPa, 0.13 MPa and 0.30 MPa). The results showed that at 0.07 stress level, the compressive stiffness at 2 seconds of the anterior band(AB), intermediate zone(IZ) and posterior band(PB) of the disc was 4.48 MPa, 3.93 MPa and 6.31 MPa, respectively; the coefficient K of permeability was 0.272 x 10(-12) m4/NS, 0.30 x 10(-12) m4/NS, and 0.042 x 10(-12) m4/NS. At 0.30 Mpa level, the compressive stiffness increased to 14.07 MPa, 13.68 Mpa and 14.00 MPa; the coefficient K lowered to 0.017 x 10(-12) m4/NS, 0.012 x 10(-12) m4/NS and 0.005 x 10(-12) m4/NS. In conclusion, the TMJ disc has viscoelastic and biobiphasic properties against compression. The stiffness of the disc increases with the increment of stress level but the coefficient of permeability decreases. These findings demonstrate that the modulation of stress and of permeability may be two important factors influencing stress dissipation and shock absorption behavior of the disc during compressive and impact loads.

[Expression of beta 2-adrenoceptor on peripheral lymphocyte and the level of plasma cAMP in patients with pregnancy induced hypertension].

OBJECTIVE: To evaluate the alteration of beta 2-adrenoceptor-cAMP system in patients with pregnancy-induced hypertension (PIH). METHODS: The Bmax of beta 2-adrenoceptor on the lymphocyte was examined by radioligand binding technique. The level of plasma cAMP was observed by radio competitive protein binding assay. RESULTS: The Bmax of beta 2-adrenoceptor (259 fmol/10(6) cell +/- 22 fmol/10(6) cell) on the lymphocytes was significantly higher in PIH group than in control group (247 fmol/10(6) cell +/- 23 fmol/10(6) cell). There was no significant difference between the PIH group and the controls. The content of plasma cAMP (2.94 nmol/40 microliters +/- 0.91 nmol/40 microliters) was significantly higher in PIH group than in control group (0.19 nmol/40 microliters +/- 0.05 nmol/40 microliters). CONCLUSION: The mechanism of PIH may be related with the activity of beta 2-adrenoceptor-cAMP system.

[Measurements of proton response of two lithium fluoride detectors with different thicknesses].

OBJECTIVE: To study the response characteristics of LiF detectors to proton fluence rate and energy, and to observe the thickness effect of the detector. METHOD: Protons were generated by an accelerator. Proton energy was changed in two ways, i.e. changing the accelerator energy directly, or using detector stacks to absorb the proton energy. The incident proton energy on each chip of detector stacks was calculated according to proton range in LiF. RESULT: The response of the detector to proton fluence rate was almost constant; when proton energy was above 9 MeV, the response of the detector to proton energy was constant (less than 10% errors). When proton energy was below 9 MeV, the response reduced gradually with the decrease of proton energy. Thickness effect for LiF thicknesses of 0.4mm and 0.8mm was not obvious. CONCLUSION: The homemade LiF detector is suitable for measurement of space radiation dose. When proton component (below 9 MeV) was abundant in radiation field, the decrease of the relative thermoluminescence efficiency should be taken into consideration.

[The effects on melanoma and immunological mechanism of human interferon-alpha gene therapy by fibroblasts].

OBJECTIVE: To establish a model of fibroblast cell-mediated human IFN-alpha gene therapy for anti-melanoma. METHODS: BMGNeo-IFN-alpha DNA was transferred into NIH3T3 fibroblasts by the liposome method. The clone secreting the highest level of IFN-alpha was selected and implanted to mice intraperitoneally. RESULTS: From 12 hours after implantation, IFN-alpha activity could be detected in serum of the animal and was sustained for a long time. We used the model to treat the nude mice inoculated with melanoma and found that the growth of M21 cells was inhibited obviously. CONCLUSIONS: The results demonstrated that fibroblasts could successfully transfer and express hIFN-alpha gene in vivo and the fibroblast cell-mediated gene therapy could be used to treat human melanoma.

An experimental study of endovascular brachytherapy using liquid 32P filled balloon catheter.

OBJECTIVE: To observe the effect of endovascular brachytherapy on the prevention of restenosis after interveneional therapy and to investigate its possible mechanisms. METHODS: In the balloon-injuried rabbit model, pathological sections of iliac arteries were observed and the changes of vascular histomorphology were estimated by computer analysis of photomicrograms. Using immunohistochemical techniques, proliferating cell nuclear antigen (PCNA) was quantified to assess the proliferation of vascular smooth muscle cells. RESULTS: After rabbit iliac arteries were injured by balloon overstretch angioplasty, neointima were shown to be less proliferative in the irradiated group than in the control group, from PCNA scores. The formation of neointima was suppressed with the external elastic laminar area increasing and without the luminal area decreasing in the irradiation group. CONCLUSION: Endovascular brachytherapy using a liquid 32P filled balloon catheter system could prevent restenosis possibly by inhibiting the formation of neointima and improving positive vascular remodeling.

[Automatic modulation of refractoriness of His-Purkinje system during atrioventricular nodal reentrant tachycardia].

OBJECTIVE: To illustrate the automatic modulation of refractoriness of His-Purkinje system during atrioventricular nodal reentrant tachycardia (AVNRT) and to discuss the possible mechanisms. METHODS: Programmed electrical stimulations were performed in high right atrium (HRA) in 8 patients with AVNRT before ablation to induce tachycardia and electrocardiagraphic recordings were done synchronically when AVNRT appeared. RESULTS: All the patients had 2:1 atrioventricular (A-V) conduction when AVNRT began, 2 of whom were blocked below His bundle, 5 above His bundle and 1 unclear. After a duration of 14.03 +/- 10.03 s of 2:1 A-V conduction, 1:1 A-V conduction with bundle banch block appeared, 3 of which were right bundle branch block (RBBB), 3 left bundle branch block (LBBB), and 2 with both. Bundle branch block disappeared after a duration of 6.87 +/- 11.26 s. CONCLUSION: Effective refractory period (ERP) of His-Purkinje system at the beginning of AVNRT was modulated automatically within less than 30-60 s and thus facilitated nodal-ventricular conduction. The mechanism of this is electrical remodeling.

[Multicenter evaluation of the antimicrobial activity in vitro for six broad-spectrum beta-lactams in China using the E-test method].

OBJECTIVE: To investigate the in vitro activities of cefepime and other 5 beta-lactam antimicrobial agents against 937 strains isolated from 10 teaching hospitals in different geographical areas in China during the early 1998. METHODS: The MIC values of cefepime, ceftazidime, ceftriaxone, imipenem, cefoperazone/sulbactum, and piperacillin against 937 isolates were tested by a gradient concentration method (E-test((R)); AB Biodisk Co.). RESULTS: Quality control isolates were tested in all working processes. MIC values of 96.5% (829/859 times) of quality assurance tests were within expected ranges of NCCLS. Only imipenem and cefepime could inhibit 90% of enterobacter and citrobacter strains, Cefepime were significantly more active than 3rd Generation cephalosporins in enterobacter and citrobacter strains, and little better active than E. coli and Klebsiella spp. (98%, 95% susceptibility). All tested antimicrobial excluding ceftazidime had very high activity to oxacillin susceptible Staphylococci (95% approximately 100% susceptibility). cefepime, ceftazidime, imipenem, and cefoperazone/sulbactum had similar activity to Pseudomonas aeruginosa (83% approximately 79% susceptibility). Both imipenem and cefoperazone/ sulbactum were very active to Acinetobacter spp. (97%, 89% susceptibility). CONCLUSION: The rank order of activity of the six tested agents is imipenem (97% Susceptible) > cefepime (89%) > cefoperazone/sulbactum (86%) > ceftazidime (76%) > ceftraixone (67%) > piperacillin (57%). The data are helpful to guide empiric therapy with beta-lactams in China.

[Morphological changes of mitochondria in apoptosis of esophageal carcinoma cells induced by As(2)O(3)].

OBJECTIVE: In order to make clear how As(2)O(3) targets the cells of esophageal carcinoma, the morphology of mitochondria and expression of bcl-2/bax were selected to study the SHEEC1 cell line during early apoptosis. METHODS: The esophageal carcinoma cell line SHEEC1 was treated by As(2)O(3) with concentrations of 1, 2 and 3 micromol/L for the periods of 2, 4, 6, 12 and 24 h. Annexin-V-Fluorescein and DNA histogram were used to detect the early apoptotic cells by flow cytometry. The morphologic changes of the apoptotic cells were examined by means of light and electron microscopy. Changes in mitochondria of living cells were detected with Rhodamin 123 fluorescent probe by fluorescent microscopy. The expression of bcl-2 and bax in floating cells were determined by immunohistochemical procedures. RESULTS: Typical apoptotic morphological changes were found in SHEEC1 at 24 h after As(2)O(3) treatment. Cells in the early stage of apoptosis were detected by Annexin-V labeled at 4 h. The ultrastructural alterations of mitochondria were found as following: within 4 h after administrating As(2)O(3), mitochondrial hyperplasia with small electron-dense materials in its matrix was the earliest manifestation of cell injury. At 6 h progressive mitochondrial swelling lead to balloon-like appearance and its outer membrane disrupted at 12 h. In all groups, down regulated expression of bcl-2 and over expression of bax were always found in damaged cells. CONCLUSION: In the SHEEC1 cell line morphological changes of mitochondria are the early events in apoptosis induced by As(2)O(3) and apoptosis is closely related to over expression of bax and down-regulating expression of bcl-2. It is possible that As(2)O(3) is a mitochondriotoxic anticancer agent for esophageal carcinoma.

[Study on the expression of integrin alpha(5), beta(1) during wound healing].

OBJECTIVE: To explore the potential role of integrin family in mediating the interaction between cells and extracellular matrix (ECM) during wound healing. METHODS: In situ hybridization technique was employed in the determination of the expression of the mRNA of the subunits of integrin alpha(5), beta(1) on the surface of fibroblasts in the donor sites of partial thickness skin of 12 burned patients. RESULTS: Evidently more integrin alpha(5), beta(1) positive staining fibroblasts were found in the donor site than in normal skin (P < 0.05). CONCLUSION: The results suggested that increased expression of integrin alpha(5), beta(1) in the wound might be involved in the activation of collagen-- overproductive subpopulation of fibroblasts.

[Molecular diagnosis in a Korean family with thalassemia intermedia due to co-inheritance of triplicated alpha-globin genes (alphaalpha/alphaalphaalpha(anti 3.7)) and beta-thalassemia trait (IVS-II-1 G-->A)].

OBJECTIVE: To analyze the molecular abnormalities of beta-thalassemia intermedia in a Korean family with thalassemia intermedia. METHODS: Polymerase chain reaction (PCR), Southern blot hybridization and double strand DNA cycle sequencing were used to analyse alpha, beta and gamma globin gene organization. RESULTS: In the Korean family the interaction between a triplicated alpha-globin locus and a heterozygous beta-thalassemia gave rise to a clinical phenotype of thalassemia. The molecular defect was a heterozygosity for a single beta-thalassemia mutation (beta IVS-II-1 G-->A) and a triplicated alpha-globin gene (alphaalpha/alphaalphaalpha(anti 3.7)). CONCLUSION: Beta-thalassemia heterozygotes conjuncted with alpha-globin gene triplication was the major cause of the beta-thalassemia intermedia in this Korean family.

[The equipment of using Azolla for O2-supplementation (correction of supplimentation) and its test].

The equipment of using Azolla for O2-supplementation and food-production in future space station was developed and tested. Dog was used as the O2-consuming animal. The design of this device considered both the requirement of Azolla growth, such as illumination, temperature, humidity, nutrition and biomass harvesting, and also the food supplement, excretion draining and temperature controlling for the dog under the condition of an airtight chamber for a relatively long duration. This device was preliminarily tested for O2-release by Azolla, and data about O2-supplement by Azolla were obtained.

[An analysis on thickness effect of lithium fluoride detector in proton measurement].

OBJECTIVE: To theoretically analyze the thickness effect of a LiF detector for proton measurement. METHOD: Thickness effect of the detector was calculated by using the definition of the detector dose under ideal and reasonable hypothesis. RESULT: The thickness effect of the detector was not obvious when protons could penetrate the detector: when proton energy was above 12 MeV, the thickness effect was obvious only on condition that the detector thickness approaches proton range, when proton energy was less than 12 MeV, the thickness effect was not obvious. The thickness effect of the detector was obvious, when protons could not penetrate the detector. CONCLUSION: Thin detectors should be used in low energy proton measurements to avoid thickness effect of the detector.

[Tensile mechanics of mandibular condylar cartilage].

OBJECTIVE: To study the biomechanical properties of the mandibular condyle cartilage and determine its functional importance. METHODS: Eight condyle specimens of the four pig temporomandibular joints were obtained by autopsy and stored in sealed plastic bags at -20 degrees C. These pigs were healthy, aged 1 year +/- 2 months. In the coronal direction, 300 microns wide, rectangular cartilage-bone specimens were prepared on the right condyles using specified knife. In the sagittal direction, rectangular cartilage-bone specimens with the same width were obtained on the left condyles. Standard rectangular testing specimens were prepared from cartilage-bone specimens in a microtome kept at -20 degrees C (300 microns wide and 250 microns thick). The specimens were inserted in a soft-tissue uniaxial tensile test machine and immersed in a bath of Ringer's solution at room temperature. Preconditioning of the specimens was carried out prior to the uniaxial tensile experiment and the specimens were elongated to failure with the constant strain rate (0.05 mm/s). The resulting mean values were curved fitted with Fung's two-parameter (A, B) exponential stress strain equation using a nonlinear regression program. RESULTS: The data obtained showed that a typical nonlinear biomechanical behavior of the condyle cartilage. Toe-region and quasi-linear zone were obvious on the experimental stress strain relation curve. Theoretically fitted parameter A and B were 0.47 and 4.13 (coronally) and 0.60 and 4.70 (sagittally). The sagittal specimens showed greater tensile stress (2.92 MPa), tensile stiffness (9.04 MPa), energy absorption (6.02 N.mm) but less tensile strain (37.34%) than that of the coronal specimens (2.15 MPa, 6.55 MPa, 4.91 N.mm, and 41.12%). The differences were statistically significant (P < 0.05). CONCLUSION: The mandibular condyle cartilage is a kind of biophasic material which shows nonlinear viscoelastic behavior, anistrophic characteristics in biomechanical function and high ability to resist tensile stress in the longitudinal direction.
