Immunohistochemical evaluation of minichromosome maintenance protein 7 in astrocytoma grading.

The minichromosome maintenance (MCM) proteins, which play an important role in eukaryotic DNA replication, represent a group of proteins that are currently under investigation as novel diagnostic tumor markers. Several studies have proved a greater reliability of MCM proteins to stain proliferating cells compared to Ki67 protein, a routinely used proliferation marker in histopathology. In the present study, the expressions of MCM7 and Ki67 were estimated in 66 primary human astrocytomas in relation to tumor grade (Grade I-IV, WHO). MCM7 significantly stained more nuclei compared to Ki67 in all the histopathological grades investigated. In addition, a stronger increase of the MCM7 labelling index, in relation to the tumor aggressiveness, was observed.

[Study of memory in cerebral damaged patients related to the side of lesion (author's transl].

Twenty-four subjects with cerebral lesions, 21 of vascular and 3 of neoplastic origin, have been tested by the Authors with a battery of tests for memory evaluation. They comprehended short term, middle term and long term memory tests. For the last ones, the subjects was requested to recall remote events presented under visual, verbal and auditory modality. For each test, results comparison between left and right hemispheric cerebral damaged patients was performed. The diagnosis of the hemispherical localisation, on the basis of clinical data established, have been confirmed by instrumental exams (EEG, CAT, Angiography). A clear difference in the results between the two groups of patients does not appear except in specific tests related to memory selective types, in which the sensitive modality of tests administration seems to be more closely in relation with the damaged hemispherical functions. Ribot's law on the longer life of old memories has not been confirmed by the long term memory tests in cerebral damaged patients.

[Clinical and CT findings in ischaemic cerebrovascular diseases (author's transl)].

120 patients with an ischaemic stroke visualized by computerized tomography were investigated: the clinical examination of the patients (double-blind study) showed a wrong diagnosis in 15,8%, the topographic diagnosis was correct in a much higher percentage (81%). Possible relationships between clinical severity, CT localization and extension of the damage were investigated; a statistically significant relationship was ascertained only in the group of 38 cases who were examined within 21 days from the stroke. The site and extension of the lesion did not seem to influence the clinical evolution or to be a good prognostic indicator in this study.

[(Negative) CT findings in subjects with ischaemic strokes (author's transl)].

50 subjects who had one or several ischaemic attacks without detectable attenuation area in the CT scan were examined. CT findings grouped as follows: normal, cortical generalized atrophy and cortical localized atrophy with or without ventricular or cysternal enlargement, diffuse cortical and ventricular atrophy. The relationships between CT scan, clinical evolution, number of ischaemic attacks and predisposing risk factors were also considered. The mainly morphological value of CT and the limited dynamic and functional aspects of this technique are emphasized in agreement with other authors.

[CT findings in chronic cerebrovascular insufficiency (author's transl)].

CT examination in 100 patients affected by chronic cerebrovascular insufficiency showed a normal picture in 16 cases, while 84 subjects showed different degrees of atrophy, generalised and focal, controlateral or sometime omolateral to the infarct, with differing patterns. The mean age of subjects with normal CT was significantly lower in comparison with patients with definite atrophy. Besides the neurological picture and the time course, the patients' psychiatric status was also assessed: no definite relationship seems to exist between psychiatric disturbances and brain atrophy. Pseudobulbar syndrome was found in 46 cases and many of them showed cortical and mainly ventricular atrophy. This picture was very seldom found in patients with vertebro-basilar insufficiency. Localised cortical atrophy, on the contrary, was often associated with ischaemic stroke. The incidence of predisposing risk factors (hypertension, diabetes mellitus or high lipid levels) was greater in subjects with brain atrophy and older age than in patients with normal CT scan.

High incidence of gastrointestinal cancer in a family with ataxia-telangiectasia.

Literature suggests that not only homozygotes but also heterozygotes for ataxia-telangiectasia have a high incidence of cancer, probably due to an impairment in DNA repair. The most frequent associations are with breast, lung, bladder, prostate and stomach tumors, while no correlation with colorectal cancer has been demonstrated. The affected family reported in this paper seems to have a high incidence of gastrointestinal tract tumors, including the colorectal ones.

Activated lymphoid cells in human gliomas: morphofunctional and cytochemical evidence.

To study defensive infiltrating cells, smear preparation from 20 fresh gliomas and autologous normal peritumoural brain tissue, used as control, were analysed. May-Grünwald Giemsa staining and cytochemical reaction markers of cellular functions [acid phosphatase (AP), dihydrofolate reductase (DHFR), dipeptidilpeptidase (DAP IV) and serine esterase BLT-dependent (SE)], were employed to characterize the cells. The extent of the leukocytic infiltration and the percentage of activated lymphocytes ("hand mirror" shape lymphocytes: HMS; lymphocytes binding tumor: LBT) were also studied. In addition serum levels of T cell growth factor interleukin-2 (IL-2) and of the soluble IL-2 receptor (sIL-2R) were determined in the affected patients. In tumoural imprints, mostly in astrocytomas, we observed an increased percentage of DHFR and DAP IV positive lymphocytes, of HMS lymphocytes and of lymphocytes binding tumoral cells. Serum levels of IL-2 were significantly increased in all patients whilst sIL-2R levels, were high only in glioblastoma. These findings indicate that in malignant gliomas there is stimulation of the immune system as witnessed by the presence of activated cells inside tumor tissue and soluble activating factors in serum.

Cytochemical demonstration of nitric oxide synthase and 5' nucleotidase in human glioblastoma.

Both nitric oxide (NO), formed from L-arginine by the enzyme nitric oxide synthase (NOS), and adenosine, which is produced by 5' nucleotidase (5' N) acting on adenosine 5' monophosphate (5' AMP) are implicated in several neurophysiological processes. In addition, 5' N is a linker protein involved in cell motility. Alterations of both enzyme activities seem to be responsible for some pathological states of the central nervous system (CNS). In the present report, we have studied the cytochemical demonstration of NOS and 5' N activities in human glioblastoma cells. Enzyme activity of both was observed in tumor cells; moreover, the coincidence of enzyme histochemistry and immunohistochemistry for NOS was noted in most cases. The findings were interpreted on the basis of the cytotoxic effects due to NO production by tumor cells, and on the non-catalytic role of membrane 5' N which acting as an adhesive molecule can favour tumor invasiveness.
