To determine when spinal bone density reaches its peak, the trabecular vertebral density was assessed, via quantitative computed tomography, among females from two age groups: (1) adolescents (aged 14-19 years; N = 24); and (2) young adults (aged 25-35 years; n =24). The adolescent girls had a higher mean trabecular vertebral density (P less than 0.01), suggesting that spinal density reaches its peak around the time of cessation of longitudinal growth and epiphyseal closure.
We studied the cardiac uptake of 2-14C-D-deoxyglucose (14C-DG) in 6 dogs subjected to a 15 min left anterior descending coronary artery (LAD) occlusion followed by 30 min reflow. Coronary arterial flows during occlusion were determined with 46Sc microspheres. After 30 min reflow, 50 microCi 14C-DG were given intravenously to determine cardiac uptake. The animal was killed 45 min later and 46Sc counts/min were determined in gamma and beta counters. Quenching was determined with non radioactive blood for both isotopes in the beta counter. Contribution of 46Sc counts/min to the 14C channel was subtracted. 14C uptake was in DPM/g and was higher in the cardiac ischemic regions of four of the six dogs. In three of the six animals there was an inverse curvilinear relation between 14C-DG cardiac uptake and occlusion flow. This observation was further confirmed by high resolution autoradiography, indicating that enhanced cardiac uptake of radiolabeled DG may be a useful positron emission tomographic marker for reversible myocardial ischemia.
Past research has shown rather consistently that positive mood states lead to increased helpfulness. In an expanded analysis of the published literature, we examined six distinct views about this relation: the focus of attention, objective self-awareness, separate process, social outlook, mood maintenance, and concomitance hypotheses. For each of 61 positive affect conditions in which it was possible to generate an effect-size estimate corresponding to the relative degree of helpfulness exhibited by positive mood subjects (compared with neutral affect subjects), judges assessed the contextual levels of variables relevant to each of the six hypotheses by reading the Method section of each article. Higher-order partial correlation coefficients were then calculated to isolate the independent contribution of each of the theoretically relevant variables to the variation among the 61 effect sizes. The results support the focus of attention, separate process, social outlook, and mood maintenance hypotheses, and partially support the objective self-awareness and concomitance hypotheses.
The SNF3 gene is required for high-affinity glucose transport in the yeast Saccharomyces cerevisiae and has also been implicated in control of gene expression by glucose repression. We report here the nucleotide sequence of the cloned SNF3 gene. The predicted amino acid sequence shows that SNF3 encodes a 97-kilodalton protein that is homologous to mammalian glucose transporters and has 12 putative membrane-spanning regions. We also show that a functional SNF3-lacZ gene-fusion product cofractionates with membrane proteins and is localized to the cell surface, as judged by indirect immunofluorescence microscopy. Expression of the fusion protein is regulated by glucose repression.
A high-performance liquid chromatographic (HPLC) assay for isosorbide dinitrate in pharmaceutical formulations is described. The method employs a reversed-phase C18 column with a mobile phase containing methanol/water/acetate buffer and is specific for isosorbide dinitrate with respect to its 2- and 5-mononitrate degradation products and other organic polynitrate esters. The method is applicable to the analysis of the diluted bulk drug and dosage forms, including sublingual, oral, chewable, and timed-release preparations.
To determine changes in bone density during growth, trabecular vertebral density and an index of spinal cortical bone were measured with quantitative computed tomography in 101 children. The children were divided by age into three groups: prepubertal, indeterminate, and pubertal. Compared with prepubertal children, pubertal adolescents had significantly higher trabecular bone density and more compact bone in the spine (P less than .001). After controlling for puberty, vertebral bone density failed to correlate significantly with age, sex, weight, height, surface area, and body mass index. The results indicate that bone density increases markedly during puberty.
