DNA and histone H1 interact with different domains of HMG 1 and 2 proteins.

High mobility group (HMG) proteins 1 and 2 from calf thymus have been digested under structuring conditions (0.35 M NaCl, pH 7.1) with two proteases of different specificities, trypsin and V8. The two proteases give a different but restricted pattern of peptides in a time course digestion study. However, when the interactions of the peptides with DNA are studied by blotting, a closely related peptide from HMG-1 and -2 does not show any apparent binding. This peptide, from the V8 protease digestion, has been isolated by DNA-cellulose chromatography and has the amino acid composition predicted for a fragment containing the two C-terminal domains of the protein, i.e., approximately residues 74-243 for HMG-1. The same peptide shows the only interaction detectable with labelled histone H1. A separate function for the different domains of HMG proteins 1 and 2 is proposed.

Hepatosplenic large cell immunoblastic lymphoma in a bottlenose dolphin (Tursiops truncatus) with high levels of polychlorinated biphenyl congeners.

This report describes a large cell immunoblastic lymphoma in a bottlenose dolphin found stranded alive in Gran Canaria, Spain. Diffuse infiltration of round neoplastic cells was observed in the splenic cords and sinuses and in hepatic sinusoids, resulting in moderate organ enlargement. The tumour cells (immunophenotype IgG+ and CD3-) showed scant, lightly eosinophilic or basophilic cytoplasm, distinct cell boundaries and hyperchromatic nuclei, each with one or more nucleoli. Mitoses were common. On the basis of histopathological, immunohistochemical and ultrastructural features, the tumour was classified as an immunoblastic lymphoma. Eleven polychlorinated biphenyl (PCB) congeners, 23 organochlorine pesticides and 16 polycyclic aromatic hydrocarbons (PAHs) were detected in the blubber and liver. High concentrations of PCBs 153, 180, 138 and 187 found in the liver may have been associated with the hepatosplenic lymphoma.

Evaluation of an enzyme immunoassay and a modified coagglutination assay for typing gonococcal isolates with monoclonal antibodies.

In order to resolve methodologic and interpretation problems associated with the serotyping of Neisseria gonorrhoeae isolates with monoclonal antibodies specific for Protein I (PI) by coagglutination (the standard method; CoA1), two methods were compared with CoA1: an enzyme immunoassay (EIA) and a modified coagglutination assay (CoA2), in which methylene blue was added to the monoclonal antibody reagents. When the EIA was compared with the CoA1 method, 92.7% of the WI isolates were typed identically and 89% of the WII/III isolates were typed identically. When the CoA2 was compared with the CoA1 method, 92.7% of WI isolates were typed into similar serovars, in comparison to only 80.5% of the WII/III isolates. With the EIA method, only 21 of 328 isolates (6.4%) were retested because duplicate results were inconsistent, in comparison to 72 isolates (22%) with the CoA1 method and 120 isolates (36.6%) with the CoA2 method. In most cases result inconsistencies between duplicate tests were due to differences in reading weakly positive coagglutination reactions with single reagents (which on duplicate testing might be negative), the most notable being PIA reagent 4A12(b) and PIB reagents 1F5(b) and 2G2(g). The EIA method was the most reproducible method, and provides some degree of automation when serotyping large numbers of isolates.

Evaluation of the application of pig slurry to an experimental crop using agronomic and ecotoxicological approaches.

The agronomic and ecotoxicological effects of the application of pig (Sus scrofa) slurry during a maize (Zea mays L.) crop cycle under conditions of forced irrigation were evaluated. The 0.2-ha experimental area, of typical xerofluvent soil and of known vulnerability to nitrate (NO3-) contamination, was divided into 12 plots and provided with water measurement instruments (TDR-probes, vertical tensiometers, and ceramic candles). Samples of soil, water, soil organisms, and the crop were subjected to analytical, agronomic, and biological test procedures. The following fertilizer treatments were applied to triplicate plots: urea (U;170 kg N ha(-1)), and an optimized (P1; 162 kg N ha(-1)) and triple (P3; 486 kg N ha(-1)) dose of pig slurry. Unfertilized plots (P0) served as controls. Calculation was made of seasonal drainage and leached NO3- and sodium losses during the experimental period. Conductivity, heavy metal concentration, hardness, pH, and redox potential were determined in soil solutions. The ecotoxicological evaluation of the soil solution and matrix was based on ecotoxicity bioassays and the quantification of organic and inorganic compounds [phenols, indols, polychlorinated biphenyls (PCBs)]. The results suggest that the P3 treatment is highly contaminating due to the leaching of nitrates and increased soil salinity. Despite the fact that a Folsomia candida reproduction test indicated chronic ecotoxicological effects on the soil in plots treated with P1 and P3, the absence of organic compounds suggests that these effects may be attributable to contaminants not considered in this study.

[A clinical and epidemiological description of a series of patients diagnosed as suffering from progressive supranuclear palsy].

INTRODUCTION: Progressive supranuclear palsy is a disease that normally presents only sporadically in adults and courses in a progressive, chronic manner. It is characterised by the presence of supranuclear ophthalmoplegia, postural instability, a Parkinsonian syndrome, pseudobulbar affect, cervical dystonia and cognitive impairment. PATIENTS AND METHODS: We conducted a descriptive study of clinical and epidemiological features in a series of 18 patients who satisfied the mandatory NINDS-SPSP clinical eligibility criteria for the likely diagnosis of progressive supranuclear palsy, using the scale developed by Golbe et al. RESULTS AND CONCLUSIONS: The mean age of onset of the disease was 58.6 +/- 8.2 years, 55.5% of the patients were males, the average history of the disease at the time of diagnosis was 4.39 +/- 2.3 years, and there was a diagnostic subregister in the first 4 years of the disease. Gait disorders, falls and slowness were the most frequently observed presenting forms of the disease. During their first four years with the disease, 75% of the patients were totally independent when it came to carrying out activities of daily living, whereas after the fourth year there was a predominance of the need for aid and absolute dependence. Dysphagia was more frequent in the later stages of the disease. Ocular motility disorders and impaired cognitive functioning were obvious in the initial stages of the disease, and there was a strong correlation between the length of time the disease had been coursing and the severity of the ocular and cognitive disorders.
