Adenovirus type 2 mRNA in transformed cells: map positions and difference in transport time.

Adenovirus type 2 rat transformed cells produced two polyadenylic acid-terminated mRNA's with approximate coordinates 1.5-4.4 and 4.4-11.0 on the physical map of the adenovirus type 2 genome. These mRNA's were also formed early during lytic infection in addition to one or more smaller mRNA's from the 4.4-11.0 region. In transformed cells, the 1.5-4.4 mRNA appeared in the cell cytoplasm without detectable lag, whereas the 4.4-11.0 mRNA required at least 20 to 30 min for the maximal rate of accumulation.

Chronic intravenous dosing and blood collection in the unanesthetized monkey.

A procedure is described which allows chronic dosing and blood collection from awake monkeys. The procedure involves only minor surgical anesthesia and allows the same vein of the animal to be used several different times with home-cage recuperation periods intervening between sequential chronic catheterizations.

Esophageal cannulation for oral drug administration in the sub-human primate.

A technique of esophageal cannulation used in rats was adapted for use in the sub-human primate. Cannulation of the esophagus requires general surgical skills and a very short postoperative recovery period. No interference in eating habits or tendency toward vomiting was noted following cannulation. This technique allows the investigator an effective method for oral dosing of drugs with minimal physical and emotional stress to the animals and is quite appropriate for oral drug self-administration studies.

The effect of cocaine and D-amphetamine on punished responding.

Various dosages of d-amphetamine (0.1, 0.5, 2.5 mg/kg) and of cocaine (5.0, 20, 40 mg/kg) were administered i.p. to each of 7 rats trained in an experimentally induced conflict procedure. Sessions were 1 hr in duration and consisted of five 12 min periods; responding was reinforced with food on a F124 sec schedule of reinforcement during each period; however, in periods 2 and 4 each response was followed by the application of footshock. Significant increase in responding did not occur in any period following any of the pretreatments. Cocaine (5.0, 20 mg/kg) and d-amphetamine (0.5, 2.5 mg/kg) significantly decreased responding in both punished and unpunished periods. Following these treatments the rate of responding in punished and unpunished components was not significantly different. This suggest that psychomotor stimulants may not selectively increase anxiety, at least at dosages which are not at the same time anorexic.

Screening for breast cancer.

The feasibility of mass population screening for breast cancer by clinical examination and x-ray mammography was studied. The results indicate that such a programme could be conducted effectively by non-medical staff and be safe from the dangers of irradiation. The response rate of women invited for screening suggests that such a service is acceptable to the general public. The additional work load produced by screening would not overburden the existing surgical services.

Cryosurgical haemorrhoidectomy.

One hundred consecutive cases of haemorrhoids were treated by cryosurgery. The cryoprobe used is a convenient and simple instrument and gave a satisfactory result in 94 of the patients. The technique and complications are described. The patients were treated as outpatients, usually without anaesthesia.

Mazindol self-administration in the rhesus monkey.

The ability of the intravenous administration of mazindol (SaH 42-548) to act as a reinforcer in monkeys previously conditioned to self-administer cocaine was ascertained. Unit dosages i.e. dosage per injection, of 50 and 100 mug/kg resulted in self-administration rates significantly greater than that which occurred with saline. An inverse relationship existed between unit dosage and frequency of self-administration over the unit dosage range 50-200 mug/kg. The total mazindol dosage self-administration per session was however independent of unit dosage. Approximately 2-3 mg/kg was self-administered by each animal during a 4 hr session at each of the 3 unit dosages. This tends to indicate that the 200 mug/kg unit dosage was also reinforcing even though the self-administration rate was similar to that of saline. This study indicates that mazindol can serve as a reinforcer and that the relationship between total session intake, unit dosage, and self-administration frequency of mazindol are similar to these seen with other reinforcing psychomotor stimulant drugs.

Dissociation between LH release and pituitary cyclic nucleotide accumulation in response to synthetic LH-releasing hormone in vivo.

In order to clarify the role of cyclic nucleotides in the mediation of LH-releasing hormone (LH-RH) action on LH release, the effect of LH-RH on LH release and pituitary cyclic AMP and cyclic GMP accumulation was studied in vivo. Pituitary cyclic AMP levels were increased 10 min after the intraveneous injection of 2 mg dibutyryl cyclic AMP (db-cyclic AMP) or 25 mg aminophylline. However, serum LH levels were not altered at 10, 30 or 60 min after the administration of either agent. Synthetic LH-RH (100 ng/rat) increased serum LH levels as measured 10 min after injection, but no effect on either pituitary cyclic AMP or cyclic GMP was seen 1, 5 or 10 min after LH-RH administration. These in vivo results are contradictory to the second messenger hypothesis for either cyclic AMP or cyclic GMP in the mediation of LH-RH-induced LH released and suggest that further work must be done in order to confirm or reject this hypothesis.

Acute pharmacological activity of intravenous cocaine in the rhesus monkey.

The effects of the intravenous administration of cocaine (0.05-5.0 mg/kg) on body temperature, heart rate, respiration rate and several unconditioned behavioral categories were ascertained in unanesthetized male rhesus monkeys. Statistically significant increases in body temperature, respiration rate and heart rate occurred only after the largest dosage tested. Subjective increases in pupil size, activity, reactivity and vocalization as well as the occurrence of stereotyped behaviors, mydriasis and refusal to ingest fruit, were observed following injection with 0.2, 0.5 mg/kg and 5.0 mg/kg. A large degree of consistent intersubject variability in the magnitude of these responses was present. There was no consistent correlation across subjects between the magnitude of these responses and the plasma level of cocaine. However, within a given subject a direct correlation existed between these parameters.
