Studies of the protein synthetic activity of lysates from HeLa cells incubated in hypertonic medium.

When HeLa S-3 cells are incubated with medium made hypertonic by adding NaCl, protein synthesis is inhibited. When the cells are returned to normal conditions protein synthesis is restored. To study the molecular mechanism of this regulation of protein synthesis, lysates were prepared from HeLa S-3 cells grown in minimum essential medium (normal, N); from cells which were incubated with additional (100 mM) NaCl (hypertonic, H); and from cells which were treated similarly in hypertonic medium and then restored to isotonic conditions (hypertonic-isotonic, H-I). Lysates of H cells exhibited reduced endogenous protein synthesis. Studies with mixed lysates from H and N cells implicated that the H lysate did not contain a soluble, non-labile macromolecule (greater than 10 000 daltons) with an inhibitory activity upon the protein synthesis. Analysis by Edman reaction of H lysates showed a reduced incorporation of [35S]methionine into N-terminal suggesting that the initiation step of protein synthesis was affected. However, sucrose gradietn analysis indicated that lysates of H cells were still able to form 80-S initiation complexes with [35S] methionyl-tRNAIMet. The block in initiation was not complete. The lesion could not be reversed by adding post-ribosomal supernatant or a ribosomal salt wash from N cells to ribosomes from the H cells. The data show that the ribosomal fraction is primarily involved in the inhibition.

Characterization of rat chorionic mammotropin.

A placental lactogen or chorionic mammotropin (rCM) has been identified in the serum of pregnant rats by radioreceptor assay (RRA). Two peaks of activity were found, the first between days 11-13 and the second between days 17-21 of pregnancy. Gel filtration of day 12 serum or placental extracts revealed two peaks of lactogenic activity, the first eluting ahead of and the second appearing immediately after 125I-hPRL (human prolactin). On the other hand, when serum or placental extracts from day 17-21 pregnant rats were fractionated in the same manner, only a single peak of lactogenic activity was eluted after 125I-hPROL. The placental concentration of rCM increased from 11 mug/g wet weight at day 12 to 74 mug/g at days 14-15, after which the concentration declined to 29 mug/g on day 21. Electrophoresis of a fraction from G-100 column of a day 12 0r day 20 placental extract or serum with a V-e/V-o ratio of 2.08 revealed a single peak of lactogenic activity by RRA with an Rf equal to 0.43. Large MW rCM (V-e/V-o ratio of 1.35) from serum had an Rf equal to 0.52 whereas the large MW species from placental extracts had a mobility similar to that of small MW fraction. The half-time disappearance rate of serum rCM on day 12 is 19.5 min compared to 1.2 min on days 17-21 of pregnancy.

Disease, impairment, disability and social handicap: a community based study of people aged 70 years and over.

The aim of this research was to investigate the prevalence of disability in a total population-based sample aged 70 years and over, the social handicap resulting from the disability and the diseases and impairments contributing to disability in the most disabled subjects. From the initial sample of 856 subjects, 782 (91.4%) participated. Disability in the tasks examined varied from 1.3% of subjects unable to feed themselves to 24.4% unable to carry out housework. In the 74 most disabled subjects comorbidity was common. The major clinical disorders that contributed to impairment and disability were heart failure, osteoarthritis, stroke and dementia. Those who were disabled were considerably more likely to be handicapped than those not disabled (odds ratio 6.65, 95% confidence interval 4.73-9.36). When social support was considered, the estimated risk of handicap associated with disability ranged from 3.19 (95% CI 1.92-5.30) for the subset of subjects who had a spouse, to 52.00 (95% CI 4.03-670.6) for subjects without emotional support.

Elderly people who fall: identifying and managing the causes.

Falls are not a mundane and inevitable consequence of ageing, but are a serious threat to the health and the independence of many older people. There is now very good evidence that the risk of falls can be significantly lowered by careful clinical evaluation and institution of a physical retraining programme.
