Hormonal cross-talk between auxin and ethylene differentially regulates the expression of two members of the 1-aminocyclopropane-1-carboxylate oxidase gene family in rice (Oryza sativa L.).

Two cDNA clones, pOS-ACO2 and pOS-ACO3, encoding 1-aminocyclopropane-1-carboxylate (ACC) oxidase were isolated from rice seedling cDNA library. pOS-ACO3 is a 1,299 bp full-length clone encoding 321 amino acids (Mr=35.9 kDa), while pOS-ACO2 is 1,072 bp long and is a partial cDNA clone encoding 314 amino acids. These two deduced amino acid sequences share 70% identity, and display a high degree of sequence identity (72-92%) with previously isolated pOS-ACO1 of deepwater rice. The chromosomal location studies show that OS-ACO2 is positioned on the long arm of chromosome 9, while OS-ACO3 on the long arm of chromosome 2 of rice genome. A marked increase in the level of OS-ACO2 transcript was observed in IAA-treated etiolated rice seedlings, whereas the OS-ACO3 mRNA was greatly accumulated by ethylene treatment. Results of ethylene inhibitor studies indicated that auxin promotion of the OS-ACO2 transcription was not mediated through the action of auxin-induced ethylene. Thus, it appears that there are two groups of ACC oxidase transcripts in rice plants, either auxin-induced or ethylene-induced. The auxin-induced OS-ACO2 expression was partially inhibited by ethylene, while ethylene induction of OS-ACO3 transcription was completely blocked by auxin. These results indicate that the expression of ACC oxidase genes is regulated by complex hormonal networks in a gene specific manner in rice seedlings. Okadaic acid, a potent inhibitor of protein phosphatase, effectively suppressed the IAA induction of OS-ACO2 expression, suggesting that protein dephosphorylation plays a role in the induction of ACC oxidase by auxin. A scheme of the multiple regulatory pathways for the expression of ACC oxidase gene family by auxin, ethylene and protein phosphatase is presented.

Primary synovial sarcoma of the kidney.

The authors present two cases of primary synovial sarcoma of the kidney. Both patients had a mass in the upper part of the right kidney without any primary extrarenal neoplastic lesions. Grossly, the tumors were soft to rubbery masses measuring 5.5 cm and 5 cm in diameter, respectively. Histologically, both tumors were poorly differentiated synovial sarcoma. The lesions exhibited a hypercellular solid or lobular growth of round, oval, or short spindle cells in variably solid sheets, in intersecting fascicles, or in a haphazard fashion. Areas of solid aggregation or fascicles of the tumor cells alternating with hypocellular myxoid tissues, together with areas displaying a prominent hemangiopericytoma-like pattern, were found. Immunohistochemically, vimentin was diffusely positive and a few tumor cells were positive for cytokeratin, epithelial membrane antigen, and neurofilament. The tumor cells were negative for S- 100 protein, CD34, smooth muscle actin, and desmin, whereas CD56 and CD99 were positive. In both cases, reverse transcription-polymerase chain reaction using ribonucleic acid extracted from formalin-fixed, paraffin-embedded tissues detected SYT-SSX2 fusion gene transcripts, which are characteristic molecular findings of synovial sarcoma. One patient died 10 months after diagnosis. These tumors are unique cases of primary synovial sarcoma of the kidney confirmed by molecular study.

Knowledge, barriers, and motivators related to cervical cancer screening among Korean-American women. A focus group approach.

Cervical cancer is a significant health problem for Korean-American women. It currently is the number one female cancer diagnosed among women in South Korea. Despite this fact, Korean-American women have very low rates of cervical cancer screening. The purpose of this research were to gain an understanding of Korean women's knowledge about cervical cancer, and to identify major barriers to early screening for cervical cancer and the motivators for prevention and early detection. It is hoped that the findings will guide the development of community-based cervical cancer education and screening programs for adult Korean-American women. The health belief model (HBM) provided the theoretical basis for the study. A qualitative study with eight focus groups (n = 102) was conducted using 11 questions derived from the HBM. Focus group discussions revealed that there was misinformation and a lack of knowledge about cervical cancer. The women therefore were confused about the causative factors and preventive strategies related to cervical cancer. The findings showed that major structural barriers were economic and time factors along with language problems. Many participants were recent immigrants with no medical insurance and long work hours. The main psychosocial barriers were fear/fatalism, denial, and Confucian thinking. Participants stated that medical advice and education would influence them most to undergo a Pap test. Recommendations were made to reduce certain barriers and to increase knowledge and motivations.

Adhesive force of chondrocytes to cartilage. Effects of chondroitinase ABC.

Chondrocyte transplantation is a clinical procedure for cartilage repair. Transplanted cells may have difficulty attaching to the surface of chondral lesions because of the anti-adhesive properties of the proteoglycan rich matrix. This study used micromanipulation methods to determine if pretreatment of cartilage with chondroitinase ABC affects chondrocyte adhesion to cartilage and if chondrocytes adhere preferentially to the superficial, middle, or deep layers of cartilage. Bovine chondrocytes were transplanted in vitro on articular cartilage sections cut perpendicular to the articular surface. At various times between 15 and 75 minutes after seeding, a micropipette micromanipulation system was used to measure the adhesion force of individual chondrocytes to cartilage. The chondrocyte adhesion force increased with chondroitinase ABC treatment and seeding time but generally was similar for the different regions of articular cartilage (superficial, middle, deep layer) to which the cells were attached. For normal cartilage, the adhesion force increased from 1.29 +/- 0.24 mdyne after 15 to 30 minutes seeding to 5.29 +/- 0.25 mdyne after 60 to 75 minutes. Treatment with chondroitinase ABC at certain concentrations and durations (1.0 U/mL for 5 minutes or 0.5 or 1 U/mL for 15 minutes) led to an increase in adhesion force, whereas relatively low concentration or treatment time (0.25 U/mL for 15 minutes or 0.5 U/mL for 5 minutes) had little or no detectable effect. The increase in adhesion attributable to chondroitinase ABC treatment appeared most marked (+144% to +292%) for short (15 to 30 minutes) seeding durations but was still significant (+46%) for the longest seeding period (60 to 75 minutes) studied after the 1 U/mL for 15 minute treatment condition. These results provide direct biomechanical evidence that enzymatic treatment of a cartilage surface can enhance chondrocyte adhesion.

Usefulness of 99Tcm-MDP knee SPET for pre-arthroscopic evaluation of patients with internal derangements of the knee.

The aim of this study was to ascertain whether knee SPET can localize lesion sites in patients with internal derangements of the knee. We performed knee SPET as a pre-arthroscopic examination in 63 consecutive patients. SPET imaging was performed with a triple-headed SPET camera 4 h after the injection of 99Tcm-methylene diphosphonate. Arthroscopic diagnoses were as follows: 28 medial meniscus injuries, 24 lateral meniscus injuries, 31 anterior cruciate ligament (ACL) injuries, three posterior cruciate ligament injuries and one medial collateral ligament injury. Of 30 patients with crescent-shaped increased activity at the medial tibial plateau, 22 had medial meniscus injuries (positive predictive value: PPV 73%); of 17 patients with crescent-shaped activity at the lateral tibial plateau, 13 had lateral meniscus injuries (PPV 76%). Of 18 patients with increased activity at ACL attachment sites (primary sign), 17 had ACL injuries (PPV 94%). Of 27 patients with increased activity at bone impaction sites of ACL injury (secondary sign), 22 had ACL injuries (PPV 81%). Of 32 patients who had either a primary or secondary sign, 26 had ACL injuries (PPV 81%). We conclude that knee SPET is very useful in the management of internal derangements of the knee, particularly in determining the need for arthroscopy by localizing lesion sites.

Reproducibility of assessment of myocardial function using gated 99Tc(m)-MIBI SPECT and quantitative software.

PURPOSE: We investigated the reproducibility of the quantification of left ventricular volumes and ejection fraction, and grading of myocardial wall motion and systolic thickening with gated myocardial single-photon emission computed tomography (SPECT) and Cedars quantification software. PATIENTS, MATERIALS AND METHODS: We performed post-stress gated myocardial SPECT in 33 consecutive patients twice sequentially in situ. Sixteen frames per cycle were used for gating. Using Cedars quantitative gated SPECT software ventricular volumes and ejection fraction (EF) were calculated. Wall motion and thickening was graded using 5- and 4-point scores, respectively. Coefficients of variation for re-examination of volumes and EF were calculated. Kappa values for assessing reproducibility of wall motion or wall thickening were calculated. RESULTS: Root mean square of errors was 5.0 ml for end-diastolic volume, 3.9 ml for end-systolic volume and 1.9% for EF. No bias or difference was found between the first and second acquisition by Bland-Altman analysis. Kappa values for wall motion and thickening of repeated acquisition was 0.76 and 0.87, respectively. The value of 2 SD from Bland-Altman analysis was 14.4 ml for EDV, 11.2 ml for ESV and 5.3% for EF. CONCLUSION: We conclude that quantification of functional indices and assessment of wall motion or thickening using gated 99Tc(m)-sestamibi (MIBI) SPECT was reproducible and we found that their ranges of physiological fluctuation were narrow enough.

Localization of epileptogenic zones in F-18 FDG brain PET of patients with temporal lobe epilepsy using artificial neural network.

For an objective interpretation of cerebral metabolic pattern to find epileptogenic zones in patients with temporal lobe epilepsy (TLE), we developed a computer-aided classifier using an artificial neural network (ANN). We studied 261 epilepsy patients diagnosed as no abnormal findings (NA, n = 64), left TLE (n = 116), or right TLE (n = 81) on interictal brain F-18-fluorodeoxyglucose positron emission tomography (FDG PET) by the consensus of two expert physicians. Seventeen asymmetry indexes between the mean counts of the 34 mirrored regions were extracted from the spatially normalized images and used as input parameters. The three diagnoses of NA, left TLE, and right TLE were used as outputs of the ANN. The structure of the ANN was optimized with variable error goals and the number of hidden units. On the criteria of agreement of diagnoses with those of expert viewers, the best classifier was chosen, which yielded a maximum average agreement of 85% for the test set when we used an error goal of 20 (sum of squared error) and ten hidden units. We could devise an ANN that performed as well in diagnosing left or right TLE on FDG PET as human experts and could be used as a clinical decision support tool.

Monte Carlo and experimental investigations of multileaf collimated electron beams for modulated electron radiation therapy.

Modulated electron radiation therapy (MERT) has been proposed as a means of delivering conformal dose to shallow tumors while sparing distal structures and surrounding tissues. Conventional systems for electron beam collimation are labor and time intensive in their construction and are therefore inadequate for use in the sequential delivery of multiple complex fields required by MERT. This study investigates two proposed methods of electron beam collimation: the use of existing photon multileaf collimators (MLC) in a helium atmosphere to reduce in-air electron scatter, and a MLC specifically designed for electron beam collimation. Monte Carlo simulations of a Varian Clinac 2100C were performed using the EGS4/BEAM system and dose calculations performed with the MCDOSE code. Dose penumbras from fields collimated by photon MLCs both with air and with helium at 6, 12, and 20 MeV at a range of SSDs from 70 to 90 cm were examined. Significant improvements were observed for the helium based system. Simulations were also performed on an electron specific MLC located at the level of the last scraper of a 25x25 cm2 applicator. A number of leaf materials, thicknesses, end shapes, and widths were simulated to determine optimal construction parameters. The results demonstrated that tungsten leaves 15 mm thick and 5 mm wide with unfocused ends would provide sufficient collimation for MERT fields. A prototype electron MLC was constructed and comparisons between film measurements and simulation demonstrate the validity of the Monte Carlo model. Further simulations of dose penumbras demonstrate that such an electron MLC would provide improvements over the helium filled photon MLC at all energies, and improvements in the 90-10 penumbra of 12% to 45% at 20 MeV and 6 MeV, respectively. These improvements were also seen in isodose curves when a complex field shape was simulated. It is thus concluded that an MLC specific for electron beam collimation is required for MERT.

Predictors of survival among elders suffering strokes in Taiwan: observation from a nationally representative sample.

BACKGROUND AND PURPOSE: Cerebrovascular disease was the leading cause of death in Taiwan from 1963 to 1982. Deaths due to stroke now rank second only to cancer, with more deaths resulting from strokes than from any other single pathology. It is important to understand stroke prognosis among elderly stroke survivors, with respect to survival and attendant predictive factors, because aged population in Taiwan is growing rapidly. The aim of the present study was to discern factors affecting survival in stroke patients from a nationally representative elderly sample. METHODS: A total of 99 stroke survivors, from a representative national sample of elders aged >/=65 years on December 31, 1988, whose strokes occurred in the period 1989-1993, were followed for mortality until July 1, 1995. Personal data were gathered through home interviews conducted by well-trained community nurses, and mortality data were obtained from the national census office by using identification card numbers. Cox proportional hazards regression analysis and the stepwise technique were used to search for important prognostic factors of survival. RESULTS: Women experienced a higher mortality rate (139.8 per 1000 person-years) than men (126.4 per 1000 person-years), as age-adjusted for World Health Organization world-population figures. Stroke patients who received continuous treatment for diabetes experienced mortality risks similar to those of patients without diabetes and much lower risks than those with discontinuous diabetes treatment. Cognitive impairment was also an independent predictor of survival (relative risk 2.69, P<0.05). In addition, patients with both cognitive and mobility impairments had a 2- to 3-fold greater risk of mortality than those with only a single abnormality. CONCLUSIONS: This first report on the various prognostic factors related to survival of elderly stroke patients in Taiwan's Chinese population emphasized the benefit of continuous diabetes treatment in improving survival chances. These stroke patients should also be monitored for cognitive and mobility impairments and undergo rehabilitation.

Detecting p53 gene mutation of breast cancer and defining differences between silver staining PCR-SSCP and immunohistochemical staining.

This study detects and defines the patterns of p53 gene mutations in breast cancers. We analyse p53 gene mutations through comparing the results of single-strand-conformation-polymorphism (SSCP) and immunohistochemistry (IHC), and we try to define the differences between the results of SSCP and IHC. Twenty-seven fresh primary breast cancer tissues and eight normal breast tissues were studied. The IHC was done with the usual streptavidin-biotin peroxidase complement method by using monoclonal antibody DO-7. The results of staining was scored. The SSCP method was done by using Cold SSCP Electrophoresis System. Overexpressions of p53 protein were seven (25.9%) among 27 cancer cases on IHC. Four (57.1%) of seven cases were positive in SSCP. In SSCP, the mutations were detected in 10 (37%) among 27 cancer cases. The mutations were two in exon 5, one in exon 8, and seven cases in exon 7. All of 10 mutations were proved by sequencing analysis. Of them, only four (40%) were positive in IHC. We consider the IHC as a screening method for p53 gene mutations.

Congenital cytomegalovirus infection: three autopsy case reports.

We report three autopsy cases of congenital cytomegalovirus (CMV) infection in fetuses with a review of literature. The clinical manifestations in these cases of congenital CMV infection include intrauterine fetal death, hydrops fetalis, and CMV pneumonia associated with cardiovascular defect. The pathological characteristics were as follows: 1) the kidney was the most frequently involved organ, followed by lung and liver, 2) CMV inclusions were found predominantly in epithelial cells and to a lesser degree in endothelial cells, 3) intrahepatic bile duct epithelial cells were frequently involved, and 4) inflammatory reaction around CMV inclusions was not prominent in the early stage of pregnancy. Diagnostic confirmation was obtained by in situ hybridization (ISH) using a biotinylated CMV-DNA probe, which demonstrated intranuclear inclusions and sometimes recognized cells that did not show intranuclear inclusion.

Human colon cancer cells express multiple glycoprotein ligands for E-selectin.

The interaction between the colon tumor cell surface and the endothelial cell layer is an important component of tumor intravasation, extravasation, and metastasis. Multiple studies suggest that tumor cells may bind to E-selectin expressed on endothelial cells during these processes. To identify possible E-selectin ligands on tumor cells that may participate in this mechanism, we used E-selectin-Ig chimera affinity chromatography to isolate glycoproteins from the human colon cancer cell line Colo-205. Binding of these cells to E-selectin was specific, required the presence of calcium, and could be blocked by antibodies against E-selectin. We identified LAMP-1 (lysosomal membrane glycoprotein-1), LAMP-2, and two high molecular weight glycoproteins (>400 kDa and 300 kDa) as the main E-selectin ligands on Colo-205 cells. Treatment of the cells with N-glycanase and O-sialoglycoprotease abolished their binding to E-selectin. The high MW glycoproteins contained sialyl Lewis X and/or sialyl Lewis A glycoconjugates, and appeared to be either alternatively spliced or alternatively glycosylated forms of MUC-1 (mucin-1).

Transient prolonged stunning induced by dipyridamole and shown on 1- and 24-hour poststress 99mTc-MIBI gated SPECT.

UNLABELLED: We investigated whether poststress gated SPECT, which was believed to show resting wall motion, revealed stunning induced by dipyridamole stress. METHODS: In 62 patients with coronary artery disease (n = 57) or chest pain (n = 5), dipyridamole stress gated 99mTc-hexakis-2-methoxyisobutyl isonitrile (MIBI) SPECT and rest 201Tl SPECT were performed on the first day; 24-h delayed 201Tl SPECT and rest gated 99mTc-MIBI SPECT were performed on the second day. Stress and rest gated 99mTc-MIBI SPECT was performed 1 h after injection. The myocardium was divided into 17 segments, and perfusion was scored on a 4-point scoring system (scores, 0-3 for normal to defect); wall motion during first-day poststress gated and second-day rest gated SPECT was also scored on another 4-point scale (scores, 0-3 for normal to dyskinesia). RESULTS: Thirty-one of 62 patients showed wall motion abnormality that was worse after stress than during resting. Three hundred eight (29%) of the total 1054 segments showed wall motion abnormality on poststress gated SPECT. In 198 of these segments, wall motion abnormality was the same on poststress and rest gated SPECT, and 106 segments showed wall motion that was worse on 1-h poststress than on rest gated SPECT. Perfusion was normal either during rest (n = 113) or after a 24-h delay (n = 18) in 131 segments with the poststress wall motion abnormality. Of these 131 segments, 69 showed the same wall motion abnormality between poststress and resting periods (persistent stunning). However, in 40 segments, abnormal wall motion on 1-h poststress gated SPECT normalized on rest gated SPECT (transient prolonged stunning). The other 20 segments showed improvement of wall motion during rest compared with the poststress period but still showed abnormal wall motion during the resting period (between transient prolonged stunning and persistent stunning). Stress perfusion decrease was more severe in transient prolonged stunning than in persistent stunning. Poststress wall motion abnormality was more severe in persistent stunning. CONCLUSION: Using gated 99mTc-MIBI SPECT, stunned myocardium was found on 1-h poststress SPECT compared with normal resting wall motion found on rest gated SPECT on the next day. We conclude that some myocardial walls did not show true resting wall motion on 1-h poststress gated SPECT; hence, caution is necessary when using wall motion on 1-h poststress gated SPECT to assess resting wall motion.

Value of morphometric nuclear image analysis using the Feulgen reaction in renal cell carcinoma.

OBJECTIVE: To evaluate retrospectively the ability of morphometric nuclear image analysis to predict survival in patients with renal cell carcinoma. STUDY DESIGN: The subjects were 40 patients with previously untreated renal cell carcinoma. Pathologic stage was determined using Robson's stage system. Nuclear grade was assigned according to the criteria of Fuhrman et al. We used the Feulgen staining technique, which has been widely used for the histochemical assessment of nuclear DNA content. A minimum of 300 nuclei were analyzed from each subject. Five variables in morphometric nuclear image analysis were measured: nuclear area, nuclear perimeter, nuclear ellipticity, nuclear regularity and DNA content. Cox's proportional hazard model was applied to identify prognostic usefulness with respect to survival time. RESULTS: All nuclear morphometric variables but nuclear regularity correlated with tumor grade. According to univariate survival analyses, Robson stage and nuclear ellipticity revealed a prognosis on survival with statistical significance. After adjustments for age and sex, nuclear ellipticity remained the only significant prognostic factor related to survival (P < .01). The survival rates were relatively high for patients with nuclear ellipticity > 773 as compared to those with nuclear ellipticity < 773 (P < .05). CONCLUSION: These findings indicate that morphometric nuclear image analysis using the Feulgen reaction is a reliable and efficient technique and that nuclear ellipticity is the most discriminating morphometric variable for predicting the prognosis of renal cell carcinoma patients.

Cerebellar vasoreactivity in stroke patients with crossed cerebellar diaschisis assessed by acetazolamide and 99mTc-HMPAO SPECT.

UNLABELLED: Crossed cerebellar diaschisis (CCD) tends to persist or even worsen after supratentorial infarction. Several studies have shown impairment of cerebral vasomotor responsiveness in the hemispheric area of diaschisis in patients with hemispheric infarction. This finding has led to the concern that the lack of CCD reversibility might be associated with chronic circulatory abnormalities. We therefore assessed the vasoreactivity in the cerebellar hemisphere in which diaschisis is manifested using acetazolamide (ACZ) and SPECT. METHODS: Eight stroke patients with CCD (5 with unilateral hemispheric infarcts and 3 with unilateral intracerebral hemorrhage) had 99mTc-HMPAO SPECT scanning at rest and 20 min after intravenous injection of 1.0 g ACZ. The time interval after stroke ranged from 25 to 904 d. From the total counts obtained from each cerebellar hemisphere, the asymmetry index (AI) was calculated as (unaffected - affected cerebellar hemisphere)/unaffected cerebellar hemisphere x100. RESULTS: After ACZ, the mean AI (8.7+/-6.6) was significantly decreased (P<0.05) compared with that at rest (17.7+/-5.8). Seven of the 8 patients showed decrease in the AI after ACZ. In 1 patient, the direction of the asymmetry was reversed after ACZ so that the AI was negative. The ACZ-induced change in the AI did not show a significant correlation with the time interval after stroke, whether calculated in absolute terms or as a percentage change. CONCLUSION: This study shows that normal vascular supply is maintained in the CCD-affected cerebellar hemisphere over long periods of time after a stroke. Thus, the lack of CCD reversibility may not be attributed to a chronic circulatory insufficiency. The results lend support to the concept of functional deactivation and subsequent transneuronal degeneration as a likely explanation for CCD. It is unclear whether decreased AI after ACZ indicates a higher vascular response of the affected cerebellar hemisphere than that of the normal side.

Limited incremental diagnostic values of attenuation-noncorrected gating and ungated attenuation correction to rest/stress myocardial perfusion SPECT in patients with an intermediate likelihood of coronary artery disease.

UNLABELLED: Either gated myocardial perfusion SPECT or attenuation-corrected SPECT can be used to improve specificity in the diagnosis of coronary artery disease (CAD). We investigated whether attenuation-noncorrected gating and ungated attenuation correction could improve the diagnostic performance of rest/stress perfusion SPECT in patients having an intermediate pretest likelihood of CAD. METHODS: Sixty-eight patients (29 men, 39 women; mean age, 59 +/- 12 y) with coronary artery stenosis > or =70% (1 vessel, n = 13; 2 vessels, n = 18; 3 vessels, n = 8; normal, n = 29) underwent rest attenuation-corrected 201TI SPECT and dipyridamole stress gated attenuation-corrected 99mTc-methoxyisobutyl isonitrile SPECT with an ADAC vertex camera. Three physicians graded the post-test likelihood of CAD for each arterial territory using a 5-point scale (1, normal; 2, possibly normal; 3, equivocal; 4, possibly abnormal; 5, abnormal). The sensitivity, specificity, and areas under receiver-operating-characteristic curves were compared for each operator by 3 methods: attenuation-noncorrected rest/stress SPECT, gated poststress SPECT plus attenuation-noncorrected rest/stress SPECT, and attenuation-corrected rest/stress SPECT plus gated poststress SPECT plus attenuation-noncorrected rest/stress SPECT. RESULTS: When higher than grade 3 was used as the criterion for CAD, no differences in sensitivity and specificity were found among the 3 methods for each operator. Areas under receiver-operating-characteristic curves for the diagnosis of CAD and stenosis revealed no differences for each modality (P > 0.05 for each comparison). CONCLUSION: In patients with an intermediate risk of CAD, viewing attenuation-noncorrected gated poststress SPECT and ungated attenuation-corrected rest/stress SPECT images did not improve the diagnostic performance for CAD and stenosis.

An epidemiologic study of dog bites among postmen in central Taiwan.

BACKGROUND: The purpose of this study was to investigate the occupational hazard of dog bites among postmen. METHODS: Data were collected from postmen working for 9 post offices in central Taiwan using the postmen's 1994 annual health examinations. Of 237 eligible postmen, 192 postmen (81.0%), including 183 men and 9 women, participated in this study. Detailed personal and occupational information and experiences of dog bites during a 3-year period (1991 to 1994) were obtained through a structured questionnaire. Baseline variables and occupational data were examined as risk factors for the dog bites among the postmen using multiple logistic regression analysis. RESULTS: We found that, during the 3-year period of study, 71.4% (137/192) of the postmen had had the experience of dog bites. Only 48.9% (67/137) had sought medical attention after a dog bite injury. There was also a regional difference among postmen experiencing dog bites. After controlling multiple variables, there was a 2.70-fold increase in risk of having dog bite accidents among postmen who worked in rural areas compared with those who worked in urban areas (95% confidence interval, 1.39 to 5.25). CONCLUSION: Dog bites are a common occupational hazard for postmen. The incidence of dog bites among postmen is higher than the reported rates among general populations. There is a need to develop an effective prevention strategy to reduce the occurrence of dog bites among postmen. We also emphasize the importance of seeking adequate medical treatment after a dog bite injury.

Transforming growth factor beta one (TGF-beta 1) enhancement of the chondrocytic phenotype in aged perichondrial cells: an in vitro study.

BACKGROUND: Perichondrium is recognized as a tissue with chondrogenic potential yielding cells which can be used for osteochondral repair. Factors which influence the proliferative ability and chondrocytic phenotype of such cells include age and presence of specific growth factors, i.e. TGF-beta 1. The present in vitro study assessed proliferation and markers of chondrocytic phenotype in cells extracted from the rib perichondrium of four- to five-year-old aged rabbits, and assessed the effects of exogenously added TGF-beta 1 on those cells. METHODS: Assays included 3H-thymidine incorporation (cell proliferation), 35S-sulfate incorporation (proteoglycan synthesis) and quantitative RT-PCR for determination of type II collagen gene expression. RESULTS: The results demonstrated that addition of TGF-beta 1 to the culture media stimulated thymidine incorporation and proteoglycan synthesis up to four- and five-fold, respectively, in aged perichondrium-derived cells. Moreover, the exogenous addition of TGF-beta 1 to the culture media resulted in an upregulation of transcriptional expression of the type II collagen gene. CONCLUSIONS: In summary, the present study has demonstrated that exogenously added TGF-beta 1 can stimulate proliferation and chondrocytic phenotype in aged perichondrium-derived cells in vitro.
