Cloning of a human complement component C4 gene.

Six overlapping cosmid clones having an average insert size of 40 kilobase pairs were identified and isolated from a human genomic library by using a cDNA probe, Alu-7, specific for the amino acid sequence of C4d, a known region of the fourth component of human complement. Analysis of these genomic clones by restriction digestion and Southern blotting shows that all six probably contain the same complete C4 gene. Nucleotide sequence comparison of the genomic clone Cos-A and the cDNA clone Alu-7 shows an identical sequence except for the presence of a 1,500-base-pair intron in the genomic sequence. The amino acid sequence predicted from the nucleotide sequence agrees with the known C4d region amino acid sequence with one exception.

Identification of H-2-controlled structural variants of the murine Slp protein and demonstration of cis-regulation of its expression.

H-2 haplotype-related structural variation in Slp beta-chains was detected by the limited proteolysis peptide mapping technique. Two distinct peptide patterns were distinguished: Slp.1 was found in strains carrying the S regions of the H-2d and H-2s haplotypes, and Slp.2 was found in strains bearing the S region of H-2w7. These different patterns were expressed codominantly in male (Sd X Sw7)F1 hybrids, whereas only the Slp.2 pattern was expressed in female heterozygotes. The 2 beta-chains are most likely the products of alleles of the Slp structural gene, which must then be located in the murine major histocompatibility complex, very probably in the S region.

Structural studies on the murine fourth component of complement (C4). IV. Demonstration that C4 and Slp are encoded by separate loci.

The S region of the murine major histocompatibility complex (MHC) encodes the structural gene for the fourth complement component (C4) and controls C4 serum levels and expression of the sex-limited protein (Slp). Although Slp was first throught to be an allele of C4, more recent studies have suggested that Slp is either a modified product of the C4 locus or is encoded at a separate locus. By using a sensitive peptide analysis technique, each of the three constitutive polypeptide chains of C4 and Slp was compared, and multiple distinct peptides as well as several shared peptides were detected. These results demonstrate that although C4 and Slp undoubtedly share a common genetic origin, they are encoded by separate structural genes.

Studies on murine Ss protein: demonstration that S region encodes structural gene for fourth component of complement.

Although genes controlling the expression of certain complement components have been shown to be linked to the major histocompatibility complex of several species, the structural genes that encode these molecules have been more difficult to map. In this study, the three constitutive polypeptide chains of the fourth component of murine complement (C4) (alpha, beta, and gamma) were isolated from 14 different inbred strains and compared by peptide mapping on analytical sodium dodecyl sulfate gels. The peptide patterns of the alpha and gamma subunits appeared to be nearly identical, but two distinctly different patterns were observed for the C4 beta chain. This structural variant was mapped to the S or G region and, as such, provides direct evidence that a structural gene for a complement component is encoded within a major histocompatibility complex.

Studies on the murine Ss protein: demonstration that the Ss protein is functionally the fourth component of complement.

This report describes a simple and reproducible assay for the fourth component of murine complement (C4). Utilizing this assay, experiments are described which conclusively demonstrate that the murine Ss protein (serum substance) is functional C4 and that ascitic fluid may be used as a source for isolating Ss. The evidence that Ss is functional C4 is: (i) pretreatment of mouse plasma with F(ab')2 anti-Ss completely abrogated the C4 hemolytic titer; and (ii) in the isolation of the Ss protein from ascitic fluid, Ss immunochemical activity and C4 functional activity coeluted in each step of a four-step purification procedure.

Using self-care theory to guide nursing management of the older adult after hospitalization.

This article describes Orem's Self-Care Theory (Orem, 1985) and applies it to the home setting. The purpose of the study was to identify how older adults manage self-care activities at home following discharge from a hospital. Results of the survey indicated that 66% of the clients reported self-care deficits after discharge. In addition, 15% reported inability to care for themselves totally after discharge. Analysis of the information indicated the lack of a supportive-educative system to promote self-care at home. The solution to the management of self-care is the rehabilitation nurse clinical specialist, who can provide nursing strategies that promote partnership with the older adult in planning self-care activities after hospitalization.

National Council Licensure Examination success: accurate prediction of student performance on the post-1988 examination.

This study was designed to identify variables that might be used as predictors for success on the post-1988 National Council Licensure Examination (NCLEX) and to identify those students at risk of failing the examination. The convenience sample consisted of 257 graduates of a baccalaureate nursing program from 1988 to 1990. The investigators examined 15 variables to determine their predictive value for NCLEX success. Grades in the first senior level nursing course and graduation grade point index were the two best predictors. Results of the discriminant analyses identified seven significant predictor variables leading to successful classification of 86 per cent of students at the end of the junior year, 88 per cent during the senior year, and 91 per cent by the time of graduation. Findings from this study indicate that reasonably accurate predictive data on individual student performance is available by the end of the junior year, allowing faculty to begin interventions for at-risk students. The greater precision of prediction during the senior year and at graduation provides direction for later remediation to increase the probability of passing the NCLEX.

Correlation between a DNA restriction fragment length polymorphism and C4A6 protein.

The fourth component of complement (C4) in man, is coded for by two separate but closely linked loci (C4A and C4B) within the major histocompatibility region (MHC), on the short arm of chromosome 6. Like class I and II loci of this region, the C4 genes are highly polymorphic with more than 30 alleles, including null alleles, assigned to the two loci. This extensive polymorphism, based mainly on electrophoretic mobility, provides a useful marker for studies of disease susceptibility. Several disorders, including systemic lupus erythematosus and type I diabetes, show associations with C4 phenotypes. We have used the technique of Southern with a C4 specific probe to examine the genomic DNA of individuals typed for C4 by protein electrophoresis. We have identified 10.7 and 3.8 kilobase (kb) BglII restriction fragments in each of 9 unrelated individuals with a C4A6 allele, and in none of 22 unrelated individuals in whom this allele was not expressed. This clear correlation of restriction fragment length polymorphism with C4 phenotype provides a precise basis for analysis of C4 polymorphism. It is likely to be of value in clinical investigations of autoimmune disease.

A molecular map of the human major histocompatibility complex class III region linking complement genes C4, C2 and factor B.

Four human complement genes, which have previously been mapped between HLA-D and HLA-B on chromosome 6, have now been aligned on a 98-kilobase (kb) section of the chromosome on the basis of four overlapping cosmid clones of genomic DNA. The C2 and factor B genes, less than 2 kb apart, are about 30 kb from two C4 genes separated from each other by about 10 kb.

Assessing students' perceived threats to safety in the community: instrument refinement.

The shift to community-based practice presents challenges to traditional nursing education. One such challenge is that of student safety. The authors discuss the continuing efforts to develop an instrument to assess student perceptions of threats in the community as well the findings of a pilot study. Analysis of the data indicates that the Environmental Comfort Scale III is valid, reliable, and easy to administer and interpret.

The role of complement and natural antibody in intestinal ischemia-reperfusion injury.

Activation of the complement cascade is central to many types of injury. Ischemia-reperfusion is an important example of such an event. Using intestinal ischemia-reperfusion as a model, we have further elucidated the importance and mechanism of this activation. Of novel importance is the evidence that natural antibody is a trigger for these events via recognition of self-antigen. In this article, we review the role of natural antibody and complement in intestinal ischemia-reperfusion injury. It is hoped that this study will ultimately lead to better understanding of these important modulators and their role in this type of injury.

Microbiological validation of a new manufacturing complex for an injectable biological product.

The Raritan Biological Production Facility (RBPF) at Ortho Pharmaceutical Corporation, Raritan, NJ, is a unique facility designed and built exclusively for the production of a sterile, injectable biological product of murine monoclonal origin. This product is the first injectable monoclonal antibody product to be licensed by FDA's Center for Biologics-Evaluation and Research (CBER). Thus, Ortho's Biotechnology Division had a unique opportunity to work very closely with CBER throughout all aspects of facility design, construction and validation, including microbiological validation of the facility and its equipment. This paper will address how existing guidelines for pharmaceutical and sterile products were used to develop initial validation protocols for the different areas and applications within the facility, and how the data gathered were used, with the assistance of CBER, to develop operating specifications and monitoring programs, for the operations within the complex.
