[Specific cell-mediated cytotoxicity in patients with cerebral infarct].

The purpose of the work was to assess changes in the activity of cytotoxic lymphocates in patients with cerebral infarction. The material comprised 27 patients with the diagnosis of cerebral infarction and 18 controls. The authors analyzed the activity of cytotoxic lymphocytes, against the cerebral antigen-encephalitogenic peptide (EP). As target cells they used syngenic lymphocytes coated with EP and labelled with 51Cr, the activity of which released during reaction with supernatan is proportional to the amount of cells disrupted by cytotoxic lymphocytes. The reaction is specific for the encephalitogenic peptide. The activity of specific cytotoxic lymphocytes was in patients with cerebral infarction almost four times higher than in controls. The percentage activity was higher in "seriously affected patients" and two weeks after the onset of the disease. Then the activity declined. Investigation of the activity of cytotoxic lymphocytes could prove important in patients with cerebral infarction with symptoms of sensitization against cerebral antigen.

HLA-D/DR identity results in a positive CML reaction.

Investigating HLA-A, HLA-B, HLA-C, and HLA-DR antigens, MLR, CML, and PLT reactivity in two unrelated persons it was found that despite their HLA-D/DR identity cytotoxic T lymphocytes (CTL) could be induced in the CML assay. The HLA-DP antigens proved to provide the necessary proliferative impetus for the generation of CTL.

A review: HLA and diseases--current status.

Since the discovery of polymorphism of the HLA complex the relationship between HLA antigens and various diseases has been followed. A number of HLA disease associations have been found. A review of the most important ones is given in this article. Explanation of the relations: "HLA and diseases" remains obscure till now what however does not hinder to take profit from them in practical medicine at diagnosis, differential diagnosis, therapy, prevention and also in genetic advisory service.

[Myocarditis in childhood].

In a group of 82 children followed up for prolonged periods the authors draw attention to some problems of virus myocarditis. They focused attention on the diagnosis of Coxsackie myocarditis for which they elaborated a scoring system. Based on this system they separated the prognostically most adverse group where cardiac decompensation was the dominant sign from the very beginning. They confirmed the theoretical assumption that in some children virus myocarditis develops into cardiomyopathy. This was proved in six children by histological examination and in 19 children by means of echocardiography.

[The development of cytotoxic T lymphocytes in HLA-D/DR identity].

The development of cytotoxic T-lymphocytes is controlled by genes of the HLA complex and occurs, in the majority of cases, when the responding and stimulating subjects differ both in HLA-A, -B and HLA-D/DR antigens. We examined a married couple for kidney transplantation and found that they differed in HLA-A, -B antigens and were identical in HLA-DR antigens. The negative results of the MLC test confirmed the serological findings. Nevertheless, cytotoxic T-lymphocytes developed despite HLA-D/DR identity. The presented case is one of the few published exceptions to the rule governing the development of cytotoxic T-lymphocytes.

[Expression of HLA antigens using the radioimmunoanalysis method].

Expression of HLA antigens class II on stimulated T lymphocytes was studied by radioimmunoassay. The products of all three HLA loci --DR, --DP, --DQ were present on stimulated cells in contrast to resting control T lymphocytes which do not express these antigens and are Ia negative.

Genetic determination of phagocytic activity of polymorphonuclear leucocytes.

Sixty-three persons with the known HLA-A, -B, -C, -DR and -Dw antigens were investigated for phagocytic ability, candidacidal activity, respiratory burst values, and the levels of circulating immune complexes, C2, C4, IgG, IgM, and IgA. The investigated persons were divided into the HLA-DR2/Dw2, the HLA-DR3/Dw3 and a control group, members of which possessed neither antigen in question. The phagocytic activity of polymorphonuclear leucocytes ingesting dead cells of C. albicans and the candidacidal activity were found to be significantly lower (P less than 0.02) in the HLA-DR3/Dw3 compared to the control group. The respiratory burst values did not considerably differ in all three groups under study. The levels of circulating immune complexes were higher in both the HLA-DR2/Dw2 and HLA-DR3/Dw3 in comparison with the control group.

Supplementary characteristics of anti-MHC class II monoclonal antibodies elicited by an ALL cell line: immunofluorescence cytofluorometry, C-dependent cytotoxicity, two-dimensional analysis of antigen.

Monoclonal antibodies directed to MHC class II antigen(s), elicited by a non-T, non-B ALL cell line, were characterized by immunofluorescence flow cytofluorometry and ELISA immunofiltration measurements of their immunoreactivity with selected neoplastic hemopoietic cell lines, determination of their complement-dependent cytotoxic activity against isolated peripheral blood B and T lymphocytes and by two-dimensional electrophoretic analysis (isoelectric focusing, SDS-PAGE) of radiolabeled, immunoprecipitated by these antibodies cell surface antigens. Patterns of these immunological reactivities, as well as two-dimensional radioimmunoprecipitation patterns (acidic heavy chain p35 and basic light chain p30) of antigens recognized by these antibodies confirm their anti-MHC class II specificity. One of these antibodies (braFB6; IgG2b) displayed identical pattern of expression on cell lines and cell types as the typical anti-MHC class II antibodies, but immunoprecipitated only a single chain p30 radioiodinated cell surface protein (with two-dimensional pattern close to the beta-chain of MHC class II DR antigen). These properties indicate the ability of braFB6 monoclonal antibody to recognize a nonpolymorphic determinant of DP-MHC class II antigen.

The characteristics of the HLA antigens determined by cellular techniques.

The complexity of the D region of the HLA complex is rather great. Three loci--HLA-DP, DQ, DR are very well established in this time. However, the question about the independence of the first known locus of this region--HLA-D--remains elusive--it is still not known if HLA-D antigens are really independent or they rather represent some epitopes of HLA-DR/DQ antigens. The HLA-DP antigens represent the additional series of specificities of the HLA complex recognized by PLT technique. On the other hand HLA-PL antigens, detected by PLT too, correlate as to HLA-D as to HLA-DR antigens and with a great probability they do not form an independent group of HLA antigens.
