Mechanism of immune transfer by RNA extracts. Immune RNA induces the synthesis of idiotype-bearing antigen receptors in noncommitted cells.

Immune RNA (I-RNA) was extracted form lymphoid organs of BALB/c mice immunized with AKR lymphoid cells. Incubation of normal BALB/c spleen cells with this I-RNA (but not with normal RNA) resulted in leukocyte migration inhibition reactions (LMIR) against AKR extracts but not against purified protein derivative or BALB/c sarcoma extracts. This transfer was abolished by pretreating I-RNA with RNAse but not with pronase. The active fraction of I-RNA was retained by and could be eluted from Poly-U Sepharose columns. Normal cells pretreated with I-RNA also reacted in the presence of an anti-idiotypic anti-serum of anti-(BALB/c anti-AKR) specificity. Pretreatment of cells with anti-idiotypic serum plus complement did not inhibit the subsequent transfer of LMIR with I-RNA. Idiotypic receptors were expressed on I-RNA treated cells less than one hour after I-RNA treatment. Using an I-RNA of double specificity, the results suggested that I-RNA entered into and acted on the cells through a nonspecific mechanism. Finally, I-RNA could induce BALB/c anti-AKR idiotypic markers in C57Bl/6 cells, genetically committed for different idiotypes, while RNA extracted from C57Bl/6 immune cells could not induce in BALB/c cells their own genetically acquired idiotypes. This series of data would prove that I-RNA acting as a mRNA is able to induce in normal noncommitted cells the de novo synthesis of antigen receptors similar or identical to those present in the surface of in vivo immunized lymphoid cells of the same strain.

Single intravenous dose kinetics accumulation of atenolol in patients with impaired renal function and on hemodialysis.

The concentration of atenolol in plasma and urine was determined following an intravenous (i.v.) dose given to 17 hypertensive patients with a glomerular filtration rate (GFR) between 5 and 105 ml/min and in 4 patients on hemodialysis. In patients with normal renal function the mean half life of elimination was calculated to be 6.8 h. This value increased to a mean of 50.1 h in patients with a GFR below 10 ml/min. In patients on hemodialysis the half life of elimination was about 4 h. The elimination rate constants as well as the body and renal clearances of atenolol have a significant correlation with the GFR. Although accumulation of atenolol was observed, especially after multiple oral doses and in patients with a GFR below 30 ml/min, no toxic side effects occurred.

Normal murine endogenous lymphoid factor(s) inhibiting lymphocyte functions. I. In vivo effect on allogeneic tumor growth.

Murine lymphoid chalones and their in vivo effect was studied. Cytoplasmic extracts were prepared from normal murine spleen lymphocytes. When assayed in vitro, cytoplasmic extracts (CE) were able to inhibit a blastomitogenic phytohemagglutinin (PHA) response of syngeneic and allogeneic normal murine lymphocytes, as well as cell replication of murine lymphoma cells. CE also had an inhibitory effect on human lymphoblastoid cell line division but it had no effect on nonlymphoid cell division. In vivo, CE had an enhancing effect on a murine allogeneic tumor, probably by suppressing the host's immune rejection response.

[The therapeutical profit of encymatic analysis of serum digoxin on geriatric patients (author's transl)].

The develpment of an encymimmunological method of analysis for serum digoxin level makes it possible to analyse serum digoxin in hospital as well as in practice. The therapeutical profit of this encymimmunological method was tested with 32 patients being digitalised because of having a cardiac insufficiency besides various diseases and different degree of disablement. The therapeutical profit should be an extended individual radius of action. There are compared two periods of therapy. In the first way the level of serum digoxin was unknown and in the second way the level of serum digoxin was known and taken into consideration by the medical treatment. The dose of digitalis was fixed under consideration of all clinical symptoms and not only under consideration of the level of serum digoxin. That the method of analysis of serum digoxin is useful was demonstrated by a sixth part of the patients, where a wrong assessment of clinical symptoms turned out and could be corrected. The clinical symptomatic of overdigitalisation was easier to recognise and less necessary to be changed than underdigitalisation.

Enumeration of bacteria forming acetate from H2 and CO2 in anaerobic habitats.

A method has been worked out that allows the detection and isolation of bacteria fermenting molecular hydrogen and carbon dioxide to acetic acid. The ratio of methanogenic to acetogenic bacteria in sludge and lake sediment samples has been found to be approximately 100 to 1. Acetogenic bacteria could not be detected in rumen samples.

Idiotypic determinants in alloantigen receptors. I. Role in a leucocyte migration inhibition reaction.

An anti-idiotypic serum was prepared by injecting BALB anti-AKR serum into (BALB X AKR)F1 mice. Pretreatment of BALB anti-AKR immune cells with this anti-idiotypic serum plus complement abrogated a leucocyte migration inhibition reaction (LMIR) against AKR extracts but not against purified protein derivative By itself, the serum induced LMIR in immune cells but not in normal cells. The reaction was strain-specific and anti-Thy 1,2 plus complement sensitive. These results indicate that alloantigen receptors, able to trigger in LMIR in immune cells, have idiotypic determinants similar to those of circulating antibodies of the same specificity. Thus the LMIR is a good model for the study of receptors cell-mediated immune reactions of primed cells.

Rosette forming T-cells in human lymphomas: relationship with age and clinical parameters.

Peripheral blood spontaneous rosette forming T cells (RFC) were studied in 40 Hodgkin's Disease (HD) and 20 lymphosarcoma (LS) patients, classified according to the histological type and spread of disease and to their age, treatment, clinical condition and evolution. LS patients had near normal RFC values; in HD, the lowest values of RFC corresponded to the most severe histological picture. In all patients, differences could be seen according to the spread of the disease and the clinical condition, but none in relation with treatment nor with the clinical evolution in a one year period after the test. While in controls and LS patients, RFC were higher in younger than older subjects, in HD, values were higher after 50 years of age. The usefulness of RFC test in the evaluation of lymphoma patients is discussed.

14-3-2 protein in rat primary and transplanted gliomas and neurinomas and in clonal cell lines.

Experimental neurogenic tumors were induced transplacentally in rats by single injections of ethylnitrosourea (ENU). The resulting primary tumors as well as isogenic transplantation tumor lines and clonal cell lines derived therefrom were examined for their content of the brain specific protein 14-3-2 by a quantitative microcomplement fixation assay. The content of S-100 protein in the samples studied is given as well. Some of the tumors of glial or Schwann cell origin did contain 14-3-2 protein ranging from 0.6 to 1.5 mug 14-3-2 per mg total soluble protein. Our experiments also showed that the ability of a tumor to produce this specific protein is transplantable over a series of subcutaneous isogenic transplantations while in the transplantation tumors the content of this protein seemed to be reduced. We were not able so far to find a correlation between the morphology of a tumor and its capability to produce a specific protein. The clonal cell line RN2 of Schwann cell origin which has been previously described in detail contained both the brain specific proteins 14-3-2 and S-100 in comparable amounts ranging from 0.3 to 0.6 and from 0.4 to 1.0 mug specific protein per total soluble protein respectively.
