Extracellular glycosaminoglycans (GAG) released by chick embryonic fibroblasts. A possible involvement of surface receptors.

Administration of Concanavalin A (Con A) to cultured skin fibroblasts derived from chick embryos at two developmental stages produce variations in the relative concentration of individual glycosaminoglycan (GAG) secreted by the cells. This effect is different: at 7 days (increase of hyaluronic acid and dermatan sulphate and decrease of chondroitin sulphate) and at 14 days (dermatan sulphate is not detectable). All the cells bind the Con A specifically, but a different pattern of agglutination is present in fibroblasts of the two embryonic ages. Since Con A is well known to bind carbohydrate-containing surface proteins, the result suggests that the release of GAG by chick embryonic fibroblasts can be modulated by cell surface receptors.

Effects of ascorbic and dehydroascorbic acid on the multiplication of tumor ascites cells in vitro.

The effects of AA and DHA on ATP C+ cell multiplication in vitro were studied by measuring incorporation of 3H thymidine into DNA. The results obtained demonstrate that both AA and DHA have the same effects: they favor cell multiplication at low doses and inhibit it at high doses. Experiments carried out with serial doses of both these substances revealed that AA is more efficient in determining both stimulating and inhibiting effects. The lesser efficiency of DHA may be attributed to its limited stability in culture medium. Studies on the effect of high doses of AA and DHA added to the culture medium in single or fractionated doses revealed that fractionated administration is more efficient in inhibiting cell multiplication than single administration.

[Effect of serum from patients with kerion celsi on organ culture of chick embryo skin].

In the present work the Authors have studied the differentiation and the cellular multiplication of chick embryo skin incubated in vitro with normal human sera or with pathological human sera from patients affected by Kerion Celsi. In the following research the Authors have studied the skin structure by an optical and electron microscope and the thymidine 3H incorporation. The result demonstrates that the pathological human serum does not interfere with the multiplication and the differentiation of the skin in vitro, but displays a cytoaggressive action that induces a noticeable cytoplasmic vacuolation.

Stimulating effect of ascorbic acid on ascites tumor cell multiplication in vitro.

In a preceding report evidence has been presented that ascorbic acid (AA) accelerated solid tumor growth obtained by inoculating tumor ascites cells into the subcutaneous dorsal region of inbred Balb/cf/Had/Se substrain mice. The present work investigates whether this effect is the result of a direct action of AA on neoplastic cell multiplication or of an indirect action on the tumor cells mediated by changes induced by AA in the organism bearing the tumor. The results obtained demonstrate that AA at low doses (microgram 10-30/ml) stimulates ascites tumor cell multiplication in vitro, while at high doses (microgram 60-120/ml) the opposite occurs. These results suggest that the major growth of solid tumors observed in mice treated with AA may be attribute to the capacity of this substance to favor the multiplication of ATP cells which were used to induce the tumors.

[Use of aspiration biopsy for the cytodiagnosis of non-Hodgkin's lymphoma].

The data on application of fine-needle aspiration biopsy followed by cytologic investigation in the diagnosis of non-Hodgkin's lymphomas are reported. Changes in cell population and presence of proliferating cells, characteristic of non-Hodgkin's lymphomas, can be detected by this method. Since the procedure is simple, painless, complication-free and assures a more than 90% accuracy, its application is indicated for diagnosis of non-Hodgkin's lymphomas.

Inhibition of in vitro keratinization of chick embryo skin cultured with hepatectomized rat serum.

In the present study organ cultures of 6 day-old chick embryo matatarsal skin were used to determine the effect of hepatectomized rat serum in modulating epithelial differentiation. Six day-old chick embryo skin, maintained in organ culture for 6-7 days with a medium containing adult rat serum, was seem to undergo differentiation and keratinization faster then in vivo. Epidermis cultured for 6-7 days with a medium containing partially hepatectomized rat serum does not differentiate and the squamous layer is absent. Studies with 3H-cystine show a sharp decrease in the labelled amino-acids incorporation in the cultures added with serum obtained after partial hepatectomy.

Further researches on the action of hepatectomized rat serum on epithelial differentiation in chick embryo skin.

Skin explants obtained from 12-day old chick embryos, maintained in organ culture containing adult rat serum, show a lower epidermal growth in comparison with skin explants cultured with other adult animal serum; nevertheless the keratogeneous differentiation of the epidermal cell is evident. When the cultures are incubated with medium containing partially hepatectomized rat serum epidermal-cell differentiation and proliferation are absent.

Fine-needle biopsy of thyroid gland.

Critical analysis is provided by the authors on cytologic examination combined with histology of 131 patients with thyroid alterations. Cytologic examination yielded seven false-negative and four false-positive findings. Sensitivity of the procedure was 0.78 and the specificity 0.97. Aspiration cytology of the thyroid gland combined with other methods of examination can be considered an advance in the diagnostics of thyroid diseases.

Diagnostic accuracy of fine needle aspiration cytology of the thyroid: impact of ultrasonography and ultrasonographically guided aspiration.

OBJECTIVE: To determine the impact of ultrasonography on the diagnostic accuracy of fine needle aspiration cytology (FNAC) of the thyroid. STUDY DESIGN: We compared two FNAC series. In period 1 we used neither ultrasonograph nor ultrasonographically-guided FNAC in the evaluation of thyroid nodules, while both tools were routinely applied in period 2. Moreover, in the former period all cellular follicular lesions were encountered among suspicious findings, while in period 2 we advised regular follow-up examinations instead of immediate surgery for patients with no significant atypia. The basis of follow-up examinations was ultrasonographic volumetry of the nodule and repeat FNAC on growing nodules. RESULTS: The number of FNAC, positive predictive value of FNAC, number of malignancies, sensitivity, the specificity and diagnostic accuracy were higher in period 2 as compared with period 1: 3,446 vs. 1,448; 45% vs. 16%, 39 vs. 29, 92% vs. 76%, 65% vs. 87% and 66% vs. 87%, respectively. CONCLUSION: The introduction of ultrasonography permits higher diagnostic accuracy of the evaluation process. This is achieved not only by lessening false negative reports but, more important, by decreasing false positive results.

Long-term expression of a fluorescent reporter gene via direct injection of plasmid vector into mouse skeletal muscle: comparison of human creatine kinase and CMV promoter expression levels in vivo.

Expression of a fluorescent reporter gene has been studied using two alternate promoters to transcribe the green fluorescent protein (gfp) from Aequorea victoria. The human cytomegalovirus (CMV) enhancer/ promoter or the human muscle-specific creatine kinase promoter (CKM) were inserted along with the gfp cDNA into a plasmid expression vector based on a modified adeno-associated virus genome. Naked plasmid DNA was injected into the hamstring muscle of mdx mice and gfp gene expression determined from frozen muscle sections taken at 4, 14, and 42 days postinjection. Fluorescence patterns obtained by photomicroscopy and quantitative fluorescence measurements indicated a near-linear increase in the accumulation of the gfp in skeletal muscle during the length of the study, with gfp expression at 42 days being roughly four times the values obtained at 4 days. The levels of expression of gfp from the CKM construct were consistantly higher than for the CMV construct. The CKM promoter/expression vector combination demonstrates significant potential for simple, direct delivery and long-term, high-level expression of genes in skeletal muscle.

Interleukin-1 alpha: regulation of cellular proliferation and collagen synthesis in cultured human osteoblast-like cells.

In this work the authors studied the effects of interleukin-1 alpha on metabolic activities of human osteoblast-like cells in vitro. The bone nature of the cells was established by assaying for specific bone protein, the osteonectin, and the parathormone receptor, an osteoblast marker. Administration of interleukin-1 alpha to cultured osteoblasts produce an increase in cellular proliferation as suggested by 3H-thymidine incorporation and cell growth studies. Interleukin-1 alpha also affected collagen synthesis confirming its potential role on bone-formation and resorption processes.

The increasing incidence of prostate cancer in Hungary.

Prostate cancer is becoming an increasing health concern world wide, and its significance is also increasing in Hungary, where mortality rates are very similar to those in the United States of America. Mortality from prostate cancer is increasing disproportionately to the aging of Hungarian men or overall population growth, and the explanation for this alarming trend is as yet unexplained. Nevertheless, the data suggest that prostate cancer will continue to assume increasing importance as a medical problem and the need for fundamental epidemiologic investigations is clear.
